(III) comparing said activity of said human albumin-targeted reagent of step (I) with said activity of said human albumin-targeted reagent of step (II), and selecting methods that result in higher activity. ,Method.

(ii) said altered variable in step (II) is a route of administration for introducing said human albumin-targeted reagent to said non-human animal;
(iii) said altered variable in step (II) is the concentration or amount of said human albumin targeting reagent introduced into said non-human animal;
(iv) said altered variable in step (II) is in the form of said human albumin targeting reagent introduced into said non-human animal;
(v) said altered variable in step (II) is said human albumin targeting reagent introduced into said non-human animal;
(vi) said human albumin targeting reagent comprises a Cas protein or a nucleic acid encoding said Cas protein and a guide RNA or a DNA encoding said guide RNA, wherein said guide RNA is a guide RNA target sequence of the human albumin gene; is designed to target and:
(A) the altered variable element in step (II) is the guide RNA sequence or the guide RNA target sequence;
(B) said human albumin targeting reagent comprises a messenger RNA (mRNA) encoding a Cas protein and a guide RNA, and said altered variable in step (II) is the ratio of Cas mRNA to guide RNA; ;
(C) the altered variable in step (II) is a guide RNA modification; or
(vii) said human albumin targeting reagent comprises an exogenous donor nucleic acid, and:
(A) said modified variable element in step (II) is in the form of said exogenous donor nucleic acid;
(B) said exogenous donor nucleic acid targets a 5' homology arm targeting a 5' target sequence of said humanized endogenous albumin locus and a 3' target sequence of said humanized endogenous albumin locus; comprising an insert nucleic acid flanking a 3' homology arm, wherein said altered variable element in step (II) comprises a sequence or length of said 5' homology arm and/or a sequence or length of said 3' homology arm 48. The method of claim 47 , wherein the method is

(I)(a) modifying the genome of a pluripotent non-human animal cell to contain said humanized endogenous albumin locus;
(b) identifying or selecting said genetically modified pluripotent non-human animal cell comprising said humanized endogenous albumin locus;
(c) introducing the genetically modified pluripotent non-human animal cell into a non-human animal host embryo;
(d) gestating said non-human animal host embryo in a non-human animal surrogate mother; or modifying the genome of a cell stage embryo;
(b) selecting a one-cell stage embryo of said genetically modified non-human animal comprising said humanized endogenous albumin locus;
(c) gestating a non-human animal surrogate mother with a one-cell stage embryo of said genetically modified non-human animal.
