Abstract
Description
非特許文献1には、ADVやTDF内服中のB型肝炎患者のみで血清IFN−λ3値が高値を示し、in vitroの系を用いてADVやTDFが腸管細胞においてIFN−λ3を誘導することが開示されている。また、誘導されたIFN−λ3は、肝細胞内でIFN−stimulated genes (ISG)を誘導し、かつHBs抗原産生抑制効果を示すことが開示されている。
非特許文献2には、慢性B型肝炎の治療を受けた患者における肝細胞がんのリスクに関する韓国で行われたコホート研究の結果が示されている。
(1)
アデホビル若しくはテノホビル又はそれらのプロドラッグ、又はそれらの薬学的に許容可能な塩を含有する、mTOR阻害剤。
(2)
アデホビル若しくはテノホビル又はそれらのプロドラッグ、又はそれらの薬学的に許容可能な塩を含有する、Aktリン酸化阻害剤。
(3)
アデホビル若しくはテノホビル又はそれらのプロドラッグ、又はそれらの薬学的に許容可能な塩を含有する、抗腫瘍剤。
(4)
腫瘍が肝炎に起因しない腫瘍である、(3)に記載の抗腫瘍剤。
(5)
肝炎が、B型肝炎である、(4)に記載の抗腫瘍剤。
(6)
肝炎ウイルスに感染していない患者に投与するための、(3)〜(5)のいずれかに記載の抗腫瘍剤。
(7)
他の抗腫瘍剤と併用するための、(3)〜(6)のいずれかに記載の抗腫瘍剤。

腫瘍及び/又はがんを処置するためのmTOR阻害剤及び/又はAktリン酸化阻害剤であって、好ましくは患者、より好ましくはヒト患者に投与するために用いられ、治療上有効量/治療的有効量のアデホビル若しくはテノホビル又はそれらのプロドラッグ、又はそれらの薬学的に許容可能な塩を含有する、mTOR阻害剤及び/又はAktリン酸化阻害剤。
腫瘍及び/又はがんが、肝炎に起因しない腫瘍及び/又はがんであることが好ましく、B型肝炎に起因しない腫瘍及び/又はがんであってよい。患者又はヒト患者は、肝炎ウイルスに感染していない患者又はヒト患者に投与するためであってよい。他の抗腫瘍剤と併用してもよい。

腫瘍及び/又はがんが、肝炎に起因しない腫瘍及び/又はがんであることが好ましく、B型肝炎に起因しない腫瘍及び/又はがんであってよい。患者又はヒト患者は、肝炎ウイルスに感染していない患者又はヒト患者に投与するためであってよい。他の抗腫瘍剤と併用してもよい。

アデホビル若しくはテノホビル又はそれらのプロドラッグ、又はそれらの薬学的に許容可能な塩を含有する、mTOR阻害剤及び/又はAktリン酸化阻害剤は、治療上有効量/治療的有効量で、それを必要とする対象、好ましくは患者、より好ましくはヒト患者に投与することにより、腫瘍及び/又はがんを処置する方法に用いられてもよい。また、アデホビル若しくはテノホビル又はそれらのプロドラッグ、又はそれらの薬学的に許容可能な塩は、治療上有効量/治療的有効量で、それを必要とする対象、好ましくは患者、より好ましくはヒト患者に投与することにより、腫瘍及び/又はがんを処置する方法に用いられてもよく、mTOR阻害及び/又はAktリン酸化阻害する方法に用いられてもよく、mTOR阻害及び/又はAktリン酸化阻害することにより腫瘍及び/又はがんを処置する方法に用いられてもよい。
腫瘍及び/又はがんが、肝炎に起因しない腫瘍及び/又はがんであることが好ましく、B型肝炎に起因しない腫瘍及び/又はがんであってよい。患者又はヒト患者は、肝炎ウイルスに感染していない患者又はヒト患者に投与するためであってよい。他の抗腫瘍剤と併用してもよい。

腫瘍及び/又はがんを処置するために用いられる、mTOR阻害及び/又はAktリン酸化阻害するために用いられる、あるいは、mTOR阻害及び/又はAktリン酸化阻害することにより腫瘍及び/又はがんを処置するために用いられる、アデホビル若しくはテノホビル又はそれらのプロドラッグ、又はそれらの薬学的に許容可能な塩を含有する医薬製剤及び/又は医薬組成物の製造のための使用;及び
mTOR阻害及び/又はAktリン酸化阻害するための、腫瘍及び/又はがんを処置するための、あるいは、mTOR阻害及び/又はAktリン酸化阻害することによる腫瘍及び/又はがんを処置するためのアデホビル若しくはテノホビル又はそれらのプロドラッグ、又はそれらの薬学的に許容可能な塩の利用/使用。
アデホビル若しくはテノホビル又はそれらのプロドラッグ、又はそれらの薬学的に許容可能な塩は、治療上有効量/治療的有効量で、それを必要とする対象、好ましくは患者、より好ましくはヒト患者に投与してよい。
腫瘍及び/又はがんが、肝炎に起因しない腫瘍及び/又はがんであることが好ましく、B型肝炎に起因しない腫瘍及び/又はがんであってよい。患者又はヒト患者は、肝炎ウイルスに感染していない患者又はヒト患者に投与するためであってよい。他の抗腫瘍剤と併用してもよい。
アデホビルは、下記式(I)で表される化合物であり、以下、アデホビル若しくはアデホビルのプロドラッグ又はそれらの薬学的に許容可能な塩を総称して、本明細書において、「アデホビル等」と呼ぶ場合がある。

プロドラッグとしては、特に限定されないが、例えば、リン酸基が、ピボキシル基、ジソプロキシル基、アラフェナミド基等で置換されたプロドラッグが挙げられる。
アデホビル等及び/又はテノホビル等としては、特に限定されないが、例えば、アデホビル ピボキシル(アデホビル ジピボキシルともいう)及びテノホビル アラフェナミドフマル酸塩やテノホビル ジソプロキシルフマル酸塩を好適に用いることができる。
また、アデホビル等及びテノホビル等は、同位元素標識されていてもよい。同位元素としては、特に限定されないが、例えば、リン原子、炭素原子及び水素原子の安定同位体である33P、32P、13C及び2H(D)等が挙げられる。
Aktは、PI3K依存的に活性化されるキナーゼであり、BadやGSK−3等の下流シグナルを制御して、mTOR(mammalian target of rapamycin)を誘導する。したがって、本発明のAktリン酸化阻害剤により、mTORの誘導が抑制されるため、本発明のAktリン酸化阻害剤は抗腫瘍効果を奏する。
本発明における抗腫瘍剤が用いられる腫瘍としては、特に限定されないが、例えば、例えば、造血器腫瘍、固形がん等が挙げられる。
造血器腫瘍としては、例えば、急性白血病、慢性リンパ性白血病、慢性骨髄性白血病、真性多血症、悪性リンパ腫及び骨髄腫等が挙げられる。
固形がんとしては、例えば、脳腫瘍、頭頸部がん、食道がん、甲状腺がん、小細胞肺がん、非小細胞肺がん、乳がん、胃がん、胆のう・胆管がん、肝がん、膵がん、結腸がん、直腸がん、卵巣がん、絨毛上皮がん、子宮体がん、子宮頸がん、尿路上皮がん、腎細胞がん、前立腺がん、睾丸腫瘍、ウイルムス腫瘍、悪性黒色腫、神経芽細胞腫、骨肉腫、ユーイング肉腫及び軟部肉腫等が挙げられる。
中でも、腫瘍を縮小若しくは消滅させるか、あるいは腫瘍の増大を一定以下に抑制する効果を奏する。
肝炎としては、特に限定されないが、アデホビル及び/又はテノホビルが用いられる、B型肝炎であることが好ましい。
本発明の抗腫瘍剤は、肝炎に起因しない、好ましくはB型肝炎に起因しない、腫瘍に対する抗腫瘍剤として用いることができる。肝炎に起因しない、好ましくはB型肝炎に起因しない、腫瘍としては、上記造血器腫瘍や、固形がんであってよいが、肝炎に起因しない、好ましくはB型肝炎に起因しない、肝がんの抗腫瘍剤として用いてもよい。
経口投与には、錠剤、カプセル、コーティング錠、トローチ、溶液又は懸濁液などの液剤といった既知の投与用剤形を用いることができる。
非経口投与は、注射による静脈内、筋肉内若しくは皮下への投与、粉末、滴剤、スプレー若しくはエアロゾル等を用いた経鼻腔や口腔などの経粘膜投与、クリーム若しくは坐薬等を用いた直腸投与、パッチ、リニメント若しくはゲル等を用いた経皮投与などが挙げられる。
投与経路は、好ましくは、経口投与又は注射による静脈内投与である。
他の抗腫瘍剤としては、ターゲットとする腫瘍に応じて、適宜選択することができる。
本発明における抗腫瘍剤等は、好ましくは、末梢血リンパ球(PBMC)におけるリポ多糖(LPS)媒介サイトカインプロファイルをさらに調節する。具体的には、本発明における抗腫瘍剤等は、LPS誘導性インターロイキン(IL)−10産生を阻害し、LPS媒介性IL−12p70及び腫瘍壊死因子(TNF)−α発現を相互に誘導する。また、IFN−αと腫瘍剤等との組み合わせは、PBMCにおけるLPS媒介IL−12p70産生を相乗的に増強する。
ここで、過剰な免疫反応を抑制するIL−10のダウンレギュレーションはT細胞とNK細胞の機能を回復させることが期待される。したがって、本発明における抗腫瘍剤等は、mTOR阻害活性及び/又はAktリン酸化阻害活性のみならず、IL−10産生阻害剤としても機能することにより、他の抗腫瘍剤の抗腫瘍活性を高めることにもつながるため、他の抗腫瘍剤と併用することが好ましい。
本発明の抗腫瘍剤等は、mTOR阻害剤であり、かつ、IL−10産生阻害剤であってもよく、Aktリン酸化阻害剤であり、かつ、IL−10産生阻害剤であってもよく、mTOR阻害剤であり、Aktリン酸化阻害剤であり、なおかつ、IL−10産生阻害剤であってもよい。
また、本発明の抗腫瘍剤等は、IL−12p70産生増強剤であってもよく、TNF−α産生増強剤であってもよく、IL−12p70発現誘導剤であってもよく、TNF−α発現誘導剤であってもよい。
本発明の抗腫瘍剤等は、mTOR阻害剤であり、かつ、IL−12p70産生増強剤及び/又はTNF−α産生増強剤であってもよく、Aktリン酸化阻害剤であり、かつ、IL−12p70産生増強剤及び/又はTNF−α産生増強剤であってもよく、mTOR阻害剤であり、Aktリン酸化阻害剤であり、なおかつ、IL−12p70産生増強剤及び/又はTNF−α産生増強剤であってもよい。本発明の抗腫瘍剤等は、mTOR阻害剤であり、かつ、IL−12p70発現誘導剤及び/又はTNF−α発現誘導剤であってもよく、Aktリン酸化阻害剤であり、かつ、IL−12p70発現誘導剤及び/又はTNF−α発現誘導剤であってもよく、mTOR阻害剤であり、Aktリン酸化阻害剤であり、なおかつ、IL−12p70発現誘導剤及び/又はTNF−α発現誘導剤であってもよい。また、本発明の抗腫瘍剤等は、さらに、IL−10産生阻害剤であってもよい。
薬学的に許容される担体としては、特に限定されないが、例えば、抗酸化剤、安定剤、防腐剤、矯味剤、着色料、溶解剤、可溶化剤、界面活性剤、乳化剤、消泡剤、粘度調整剤、ゲル化剤、吸収促進剤、分散剤、賦形剤及びpH調整剤等が挙げられる。
投与量は、具体的には年齢、体重、一般的健康状態、性別、食事、投与時間、投与方法、排泄速度、薬物の組合せ及び患者の病状等に応じて決めることができ、さらに、その他の要因を考慮して決定してもよい。
本発明の抗腫瘍剤等として、アデホビル等及び/又はテノホビル等は、抗腫瘍効果を発揮し得る上で有効な量で含むことが好ましい。
アデホビル等及び/又はテノホビル等の1日の投与量は、患者の状態や体重、化合物の種類、投与経路等によって異なるが、非経口投与の場合は、通常、約0.01〜1000mg/人/日、好ましくは0.1〜500mg/人/日であり、また、経口投与の場合は、通常、約0.01〜500mg/人/日、好ましくは0.1〜100mg/人/日である。
それを必要とする患者は、特に限定されず、哺乳動物であり、好ましくは、ヒトである。
投与対象となる患者として、肝炎ウイルスに感染していない患者に、本発明における抗腫瘍剤等は投与するために用いられることが好ましい。
抗腫瘍剤等として、アデホビル等及び/又はテノホビル等は、治療上有効量で投与されることが好ましい。
本発明においては、アデホビル等及び/又はテノホビル等は、抗腫瘍作用を有するため、がんを発症する前に投与することにより、がんの予防に用いてもよく、がんの治療に用いてもよい。
健康成人(HV)及びB型慢性肝炎患者より採取した血液から濃度勾配法により末梢血リンパ球 (PBMC)を分離した。CD3+T細胞、CD19+B細胞、CD56+NK細胞、CD14+単球は、それぞれ磁気抗体を用いて分離した。
PBMCあるいはCD14+単球は、様々な濃度の格差アナログ製剤 (NUC)、ラパマイシン、ワートマニン、ペリフォシンとともに10%ウシ血清含有RPMI液にて2時間培養後に100ng/mLのリポ多糖(LPS)にて刺激を行い、指示された時間に上清又は細胞を採取した。使用した核酸アナログの濃度は、特別な表示がない限りは、ラミブジン(LAM),アデホビル(ADV)、エンテカビル(ETV)及びテノホビル(TDF)を、それぞれ50mM、500nM、 500nM、5mMで使用した。これらの濃度はそれぞれの核酸アナログにおける末梢血中の濃度の10倍である。その理由として、経口投与された薬剤の門脈内血中濃度は、末梢血中濃度の10倍と報告されているからである。
培養上清中の多種類のサイトカインは(一度に測定可能な)Bio-Plexヒトサイトカインパネルを用いて測定した。さらに、IL-10、IL-12及びTNF-aについては、ELISAにても解析した。
細胞質と細胞膜の分離には、MEM-PERTMPlus細胞膜蛋白抽出キットを用い、同製品の指示するプロトコールに従った。簡単に述べると、採取した細胞を洗浄後にバッファー(細胞膜に穴をあける)とともに10分間 4℃で反応させた。16,000gで15分間、遠心した後、その上清を新しいチューブに採取した(細胞質成分)。前の段階でチューブに沈殿したもの(細胞膜)を上記バッファーにて一度洗浄後に、細胞膜融解バッファーで30分 4℃で、時々振動を与えながら反応させた。その後、16,000gで15分間、遠心した後、上清を新しいチューブに採取した(細胞膜成分)。細胞質及び核成分は、細胞質/核抽出キットを用いて分離した。すなわち、採取した細胞を洗浄後にバッファー(細胞膜に穴をあける)とともに10分間 4℃で反応させた後、16,000gで15分間、遠心し、その上清を新しいチューブに採取した(細胞質成分)。前の段階でチューブに沈殿したもの(細胞膜+核)を上記バッファーにて一度洗浄後に、核融解バッファーで30分 4℃で、時々振動を与えながら反応させた。その後、16,000gで15分間、遠心した後、上清を新しいチューブに採取した(核成分)。
対照siRNAを含む化学合成ヌクレオチドsiRNAは、Santa Cruz(Dallas、TX、USA)から入手した。 Nucleofectorキット(Lonza、Cologne、Germany)を用いて細胞にsiRNAをトランスフェクトした。
細胞は、blue-loading bufferにて30分 4℃反応させ、16,000gで15分間、遠心し、その上清を採取した。同量の蛋白量の検体をSDS-PAGEゲルに流した後、膜に転写した。5%仔牛血清アルブミンで蛋白を転写した膜をブロッキングバッファーで反応させた後、同液にて一次抗体を希釈し、4℃で一晩反応させた。膜を洗浄後に二次抗体で1時間反応させ、さらに洗浄後にWestern Lighting ECL Pro NEL 12001EAにて反応させ、バンドをImageQuant LAS 4000にて検出した。
HAタグのついたAKT2-PH-EGFPをpCSII-EF1α-MCS-IRES2-Blasticidin lenti virus vectorに挿入した。同レンチウイルスをHT1080細胞に感染させ、同細胞を5mg/mlブラスチジン含有DMEM液にて培養した。PH-EGFPとリン酸化Akt、Akt-PH-EGFPの細胞内移動の観察には、それぞれを表出するHT1080細胞をカバーグラス上でDMSO、LAM、ADV、ETV、TDFと2時間培養した後に、3.7%ホルマリンにて細胞を固定し、その後リン酸化AKt (S473)及びDAPI(核染色)を行い、顕微鏡にて観察した。
化合物に対するペプチドの結合親和性は、表面プラズモン共鳴バイオセンサー(Biacore X 100、GE Healthcare)を用いて分析した。アミンカップリングキット(GE Healthcare)を用いてAkt1又はBSAをCM5センサーチップの表面に固定化した。5%DMSOを含有するリン酸緩衝生理食塩水(10mMリン酸緩衝液、2.7mM KCl、137mM NaCl、0.05%界面活性剤P20、pH7.4)で希釈した異なる濃度の化合物を20μL/minの流速で25℃で120秒間センサーチップに注入し、続いて、化合物を含まない緩衝液を180秒間適用した。参照フローセルを用いてDMSOのバルク効果を差し引き、そしてBiacore X100評価ソフトウェア(GE Healthcare)を用いてデータを分析することにより速度論的パラメータを決定した。
ノンパラメトリックなカテゴリカルデータを比較するために、ピアソンのカイ2乗検定とフィッシャーの直接検定を用いた。一元配置分散分析を分散分析に用いた。すべてのテストは、社会科学のためのIBM統計パッケージ、日本のための統計デスクトップ、V.19.0(IBM JAPAN)を用いて行われ、p値<0.05は統計的に有意と見なされる。
核酸アナログがLPS誘導性サイトカイン産生に影響を及ぼすかどうかを試験するために、3人のHVからのPBMCを核酸アナログで2時間エクスビボで前処理し、続いてLPSで刺激し、BioPlexサイトカインアッセイを用いて、サイトカイン産生を分析した。試験した15個のサイトカイン(granulocyte-colony stimulating factor (G-CSF)、IL-12、IL-10、interferon (IFN)-γ、granulocyte-macrophage (GM)-CSF、IL-13、IL-17、IL-1β、IL-2、IL-4、IL-6、IL-7、IL-8、monocyte chemotactic and activating factor (MCAF)、及びtumor necrosis factor (TNF)-α.)のうち、LPS誘導性の3個のサイトカイン(IL-10、IL-12p70及びTNF-α)の産生を変化させた。ADV又はTDFはIL-12p70及びTNF-αを有意に誘導し、IL-10を阻害した一方、LAM及びETVはDMSOと同様の効果を示したに過ぎなかった(図1A)。
他のHV由来のPBMCも同様に処理し、IL-12p70、IL-10及びTNF-αをELISAを用いて測定したところ、BioPlexアッセイの結果と同様の結果が示された(図1B)。
これらのサイトカイン調節はADV又はTDF用量依存的であった(図1C)。慢性肝炎(CH;図1D)又は肝硬変(LC;図1E)を有するHBV患者からのPBMCを用いて同様のパターンが観察された。
HBV感染のさまざまな段階から得られたPBMCにおけるサイトカイン産生を比較したところ、HV、無症候性キャリア及びCH患者の間で有意差は観察されなかった。 LC患者におけるすべてのサイトカインの産生は、HV、無症候性キャリア又はCH患者におけるのより低かった(図1F)。
PBMCをエクスビボで核酸アナログ、IFN -α及びLPSで処理し、IL-10及びIL-12p70上清レベルを測定した。ADV又はTDFを組み合わせた場合、IFN-αは相乗的にLPS媒介IL-12p70産生を増強した(図2)。
CD14 +単球をLPSで刺激し、MAPK関連タンパク質(pp38、pERK及びpMSK-1)のリン酸化を分析した。これらのタンパク質は、LPS刺激の30分後に最大にリン酸化された(図3)。
この条件下では、核酸アナログはこれらのタンパク質のリン酸化を阻害しなかったが、U0126及びSB202198はそれぞれERK及びMSK-1のリン酸化を阻害した(図4B)。
mTOR関連タンパク質は、LPS刺激の2時間後に最大にリン酸化された(図5)。
この条件下で、ADV及びTDFはAkt及びp70S6Kのリン酸化を阻害した(図4C)。
ADV及びTDFは用量依存的にAktのリン酸化を阻害した(図4D)。ADV及びTDFもまたGSK-3βのリン酸化を阻害した(図4E)。
mTOR阻害剤であるラパマイシン(図6A)及びワートマニン(図6B)は、用量依存的に、LPS媒介IL-10産生を阻害し、そしてLPS媒介IL-12p70産生を誘導した。これらの結果は、ADV及びTDFによるLPS媒介サイトカイン調節がmTOR経路の阻害によるものであることを示している。
ホスファチジルイノシトール-3キナーゼ及びホスホイノシチド依存性キナーゼ1などのAktの上流にあるいくつかのタンパク質のリン酸化を分析した。ADV及びTDFはこれらのタンパク質のリン酸化を阻害しなかった(図7A)。
安定なAkt 2 -PH-EGFP発現HT1080細胞を核酸アナログで2時間処理し、共焦点顕微鏡を用いて調べたところ、ADV及びTDFが、Akt-PHの細胞膜への転位を有意に阻害した。この結果は、HT1080細胞におけるAktリン酸化レベルの低下と一致した(図7B)。
ETV、TDF又はテノホビル一リン酸(TFV)に対するAktの結合親和性を表面プラズモン共鳴バイオセンサーを使用して分析したところ、TFVが用量依存的にAktタンパク質に結合した(図7C)。
Aktの細胞膜への移行を阻害することが周知なペリフォシンを用いて、PBMCを2時間前処理した後、LPSで刺激すると、IL-10及びIL-12p70の産生レベルはそれぞれ用量依存的に減少及び増加した(図7D)。また、ペリフォシンは用量依存的にAktのリン酸化も阻害した(図7E)。
ホスファターゼテンシンホモログ(PTEN)、mTOR複合体2、及び5'- AMP活性化プロテインキナーゼ(AMPK)は、mTOR経路を阻害することが知られている。ADV及びTDFは、LPS処理CD14+単球におけるPTEN又はpPTEN発現レベルに影響を及ぼさなかった(図9)。
ADV及びTDF誘導性IL-10レベルの減少はサイレンシングリクターによって回復されなかった(図10)。
AMPK誘導剤である5-アミノイミダゾール-4-カルボキサミド1-β-D-リボフラノシド(AICAR)は、LPS刺激PBMCと共にインキュベートした場合、IL-10産生を阻害し、IL-12p70を誘導した(図11A)。
しかしながら、AICARは用量依存的にAktのリン酸化を増強した(図11B)。AMPKサイレンシングを用いたPBMCにおけるIL-10産生は回復しなかった(図11C、D)。
これらの結果から、PTEN、mTOR複合体2及びAMPKが、ADV及びTDFによるIL-10産生の阻害に関与していなかったことが示された。
各種肝がん細胞株(HepG2, PLC/PRF/5, Huh7, Hep3B, SKHep-1, HLE)をETV又はTDFで72時間培養後にCellTiter-Glo(プロメガ)を用いて細胞内ATP量を測定することによって、ETV又はTDFが抗腫瘍効果を示すかを検討した。すべての細胞株において、TDFは用量依存性に細胞生存数を有意に低下させたが、ETVでは、その効果は認められなかった (図12)。
これらの結果から、単球で認められたADV及びTDFのmTOR経路抑制効果に基づく、肝がん細胞株における抗腫瘍効果がin vitro及びin vivoの両方で認められた。また、ADV及びTDFの発がん抑制効果は、ADV及びTDFのAktリン酸化抑制効果に基づくものと考えられた。
Claims (7)
- アデホビル若しくはテノホビル又はそれらのプロドラッグ、又はそれらの薬学的に許容可能な塩を含有する、mTOR阻害剤。
- アデホビル若しくはテノホビル又はそれらのプロドラッグ、又はそれらの薬学的に許容可能な塩を含有する、Aktリン酸化阻害剤。
- アデホビル若しくはテノホビル又はそれらのプロドラッグ、又はそれらの薬学的に許容可能な塩を含有する、抗腫瘍剤。
- 腫瘍が肝炎に起因しない腫瘍である、請求項3に記載の抗腫瘍剤。
- 肝炎が、B型肝炎である、請求項4に記載の抗腫瘍剤。
- 肝炎ウイルスに感染していない患者に投与するための、請求項3〜5のいずれか1項に記載の抗腫瘍剤。
- 他の抗腫瘍剤と併用するための、請求項3〜6のいずれか1項に記載の抗腫瘍剤。
