Abstract
Description
[1] がん幹細胞を製造する方法であって、下記の工程:
(a) Oct3/4、Sox2およびKlf4、またはそれらをコードする核酸を含む因子をがん細胞に導入する工程、および
(b) 工程(a)により得られた細胞を胚性幹(ES)細胞の維持培養条件以外の条件で培養する工程
を含み、前記工程(b)により得られた細胞集団は、Oct3/4、Sox2およびKlf4、またはそれらをコードする核酸を含む因子を導入していないがん細胞の薬剤排除能を抑制するのに有効な濃度のATP結合カセット(ABC)トランスポーター阻害剤を添加した条件において、薬剤排除能を有する細胞を含有することを特徴とする、方法。
[2] 以下の工程(c)をさらに含む、[1]に記載の方法:
(c)工程 (b)により得られた細胞から、Oct3/4、Sox2およびKlf4、またはそれらをコードする核酸を含む因子を導入していないがん細胞の薬剤排除能を抑制するのに有効な濃度のABCトランスポーター阻害剤を添加した条件において、薬剤排除能を有する細胞を抽出する工程。
[3] 前記薬剤排除能がHoechst33342を排除する能力である、[1]または[2]に記載の方法。
[4] 前記ABCトランスポーター阻害剤が、ベラパミルである、[1]から[3]のいずれか1項に記載の方法。
[5] 薬剤排除能を抑制するのに有効な濃度が50μM以上、250μM未満である、[4]に記載の方法。
[6] がん幹細胞を製造する方法であって、下記の工程:
(a) Oct3/4、Sox2およびKlf4、またはそれらをコードする核酸を含む因子をがん細胞に導入する工程、
(b) 工程(a)により得られた細胞を胚性幹(ES)細胞の維持培養条件以外の条件で培養する工程、
(c) 工程(b)により得られた細胞をトリプシンで処理する工程、および
(d) 工程(c)により解離しなかった細胞をがん幹細胞として取得する工程
を含む方法。
[7] 前記工程(c)が、0.25%トリプシンで5分間以上処理するものである、[6]に記載の方法。
[8] 前記がん細胞が、ヒト由来のがん細胞である、[1]から[7]のいずれか1項に記載の方法。
[8a] 前記がん細胞が、大腸がん細胞である、[1]から[8]のいずれか1項に記載の方法。
[9] [1]から[8]および[8a]のいずれか1項に記載の方法により作製されたがん幹細胞。
[10] 下記の特徴を有する、がん幹細胞:
(a) 外来性のOct3/4、Sox2およびKlf4をコードする核酸を含み、
(b) CD133、CD44、CD26、ABCG2およびLGR5が陽性であり、および
(c) Oct3/4、Sox2およびKlf4、またはそれらをコードする核酸を含む因子を導入していないがん細胞の薬剤排除能を抑制するのに有効な濃度のABCトランスポーター阻害剤を添加した条件において、薬剤排除能を有する。
[11] 前記薬剤排除能がHoechst33342を排除する能力である、[10]に記載の細胞。
[12] 前記ABCトランスポーター阻害剤が、ベラパミルである、[10]または[11]に記載の細胞。
[13] 前記薬剤排除能を抑制するのに有効な濃度が50μM以上、250μM未満である、[12]に記載の細胞。
[14] 前記がん幹細胞が、大腸がん幹細胞である、[10]から[13]のいずれか1項に記載の細胞。
[15] 以下の工程を含む、抗がん剤をスクリーニングする方法。
(1)[9]から[14]のいずれか1項に記載の細胞と候補薬剤とを接触させる工程、
(2)Oct3/4、Sox2およびKlf4、またはそれらをコードする核酸を含む因子を導入していないがん細胞の薬剤排除能を抑制するのに有効な濃度のABCトランスポーター阻害剤を添加した条件において、薬剤排除能を有する細胞数を測定する工程、および
(3)工程(2)で測定された細胞数が、候補薬剤と接触させなかったときの細胞数より減少した場合、当該候補薬剤を抗がん剤として選択する工程。
[16] 前記薬剤排除能がHoechst33342を排除する能力である、[15]に記載の方法。
[17] 前記ABCトランスポーター阻害剤が、ベラパミルである、[15]または[16]に記載の方法。
[18] 前記薬剤排除能を抑制するのに有効な濃度が50μM以上、250μM未満である、[15]から[17]のいずれか1項に記載の方法。
[19] 以下の工程を含む、抗がん剤をスクリーニングする方法。
(1)[6]または[7]に記載の方法で製造された細胞と候補薬剤とを接触させる工程、
(2)細胞数を測定する工程、および
(3)工程(2)で測定された細胞数が、候補薬剤と接触させなかったときの細胞数より減少した場合、当該候補薬剤を抗がん剤として選択する工程。
本発明において「誘導因子」とは、体細胞に導入することにより、その体細胞をがん幹細胞様の細胞に変換させる物質(群)であれば、タンパク性因子またはそれをコードする核酸(ベクターに組み込まれた形態を含む)、あるいは低分子化合物等のいかなる物質から構成されてもよい。誘導因子がタンパク性因子またはそれをコードする核酸の場合、好ましくは、誘導因子は、Oct3/4、Sox2およびKlf4、またはそれらをコードする核酸である。

上記の誘導因子は、DNAの形態で、あるいはタンパク質の形態で導入することができる。DNAの形態で導入する場合、例えば、ウイルス、プラスミド、人工染色体などのベクター、リポフェクション、リポソーム、マイクロインジェクションなどの手法によって体細胞内に導入することができる。ウイルスベクターとしては、レトロウイルスベクター、レンチウイルスベクター(以上、Cell, 126, pp.663-676, 2006; Cell, 131, pp.861-872, 2007; Science, 318, pp.1917-1920, 2007)、アデノウイルスベクター(Science, 322, 945-949, 2008)、アデノ随伴ウイルスベクター、センダイウイルスベクター(Proc Jpn Acad Ser B Phys Biol Sci. 85, 348-62, 2009)などが例示される。また、人工染色体ベクターとしては、例えばヒト人工染色体(HAC)、酵母人工染色体(YAC)、細菌人工染色体(BAC、PAC)などが含まれる。プラスミドとしては、哺乳動物細胞用プラスミドを使用しうる(Science, 322:949-953, 2008)。ベクターには、誘導因子が発現可能なように、プロモーター、エンハンサー、リボゾーム結合配列、ターミネーター、ポリアデニル化サイトなどの制御配列を含むことができる。使用されるプロモーターとしては、例えばEF1αプロモーター、CAGプロモーター、SRαプロモーター、SV40プロモーター、LTRプロモーター、CMV(サイトメガロウイルス)プロモーター、RSV(ラウス肉腫ウイルス)プロモーター、MoMuLV(モロニーマウス白血病ウイルス)LTR、HSV-TK(単純ヘルペスウイルスチミジンキナーゼ)プロモーターなどが用いられる。なかでも、EF1αプロモーター、CAGプロモーター、MoMuLV LTR、CMVプロモーター、SRαプロモーターなどが挙げられる。さらに、必要に応じて、薬剤耐性遺伝子(例えばカナマイシン耐性遺伝子、アンピシリン耐性遺伝子、ピューロマイシン耐性遺伝子など)、チミジンキナーゼ遺伝子、ジフテリアトキシン遺伝子などの選択マーカー配列、緑色蛍光タンパク質(GFP)、βグルクロニダーゼ(GUS)、FLAGなどのレポーター遺伝子配列などを含むことができる。また、上記ベクターには、体細胞への導入後、誘導因子をコードする遺伝子もしくはプロモーターとそれに結合する誘導因子をコードする遺伝子を共に切除するために、それらの前後にLoxP配列を有してもよい。別の好ましい一実施態様においては、トランスポゾンを用いて染色体に導入遺伝子を組み込んだ後に、プラスミドベクターもしくはアデノウイルスベクターを用いて細胞に転移酵素を作用させ、導入遺伝子を完全に染色体から除去する方法が用いられ得る。好ましいトランスポゾンとしては、例えば、鱗翅目昆虫由来のトランスポゾンであるpiggyBac等が挙げられる(Kaji, K. et al., (2009), Nature, 458: 771-775、Woltjen et al., (2009), Nature, 458: 766-770 、WO 2010/012077)。さらに、ベクターには、染色体への組み込みがなくとも複製されて、エピソーマルに存在するように、リンパ指向性ヘルペスウイルス(lymphotrophic herpes virus)、BKウイルスおよび牛乳頭腫(Bovine papillomavirus)の起点とその複製に係る配列を含んでいてもよい。例えば、EBNA-1およびoriPもしくはLarge TおよびSV40ori配列を含むことが挙げられる(WO 2009/115295、WO 2009/157201およびWO 2009/149233)。また、複数の誘導因子を同時に導入するために、ポリシストロニックに発現させる発現ベクターを用いてもよい。ポリシストロニックに発現させるためには、遺伝子をコードする配列の間は、IRESまたは口蹄病ウイルス(FMDV)2Aコード領域により結合されていてもよい(Science, 322:949-953, 2008およびWO 2009/092042、WO 2009/152529)。
本発明において、がん幹細胞を誘導するための元となる体細胞は、好ましくは、がん細胞であり得る。「がん細胞」は、自律的に制御できない増殖能を獲得した細胞であれば特に限定されず、任意の細胞が包含される。本発明におけるがん細胞は、例えば、上皮細胞由来の癌細胞であり得るが、非上皮性の肉腫細胞や血液がん細胞であってもよい。例えば、頭頸部のがん(例えば、上顎がん、咽頭がん、喉頭がん、舌がん、甲状腺がん)、胸部のがん(例えば、乳がん、肺がん(非小細胞肺がん、小細胞肺がん))、消化器のがん(例えば、食道がん、胃がん、十二指腸がん、大腸がん(結腸がん、直腸がん)、肝がん(肝細胞がん、胆管細胞がん)、胆嚢がん、胆管がん、膵がん、肛門がん)、泌尿器のがん(例えば、腎がん、尿管がん、膀胱がん、前立腺がん、陰茎がん、精巣(睾丸)がん)、生殖器のがん(例えば、子宮がん(子宮頸がん、子宮体がん)、卵巣がん、外陰がん、膣がん)、皮膚のがん(例えば、基底細胞がん、有棘細胞がん)を含むが、これらに限定されない。本発明におけるがん細胞は、好ましくは、大腸がん細胞である。
本発明におけるがん細胞はヒト由来のがん細胞であることが好ましい。
本発明においてがん幹細胞を誘導するための培地は、動物細胞の培養に用いられる培地を基礎培地として調製することができる。基礎培地としては、例えば、IMDM培地、Medium 199培地、Eagle's Minimum Essential Medium (EMEM)培地、αMEM培地、Dulbecco's modified Eagle's Medium (DMEM)培地、Ham's F12培地、RPMI 1640培地、Fischer's培地、StemPro34(invitrogen)、およびこれらの混合培地などが包含される。培地には、血清が含有されていてもよいし、あるいは無血清でもよい。必要に応じて、培地は、例えば、アルブミン、トランスフェリン、Knockout Serum Replacement(KSR)(FBSの血清代替物)、N2サプリメント(Invitrogen)、B27サプリメント(Invitrogen)、脂肪酸、インスリン、コラーゲン前駆体、微量元素、2-メルカプトエタノール(2ME)、チオールグリセロールなどの1つ以上の血清代替物を含んでもよいし、脂質、アミノ酸、L-グルタミン、Glutamax(Invitrogen)、非必須アミノ酸、ビタミン、増殖因子、低分子化合物、抗生物質、抗酸化剤、ピルビン酸、緩衝剤、無機塩類などの1つ以上の物質も含有し得る。
本発明の方法によって得られるがん幹細胞は、特定の濃度のATP結合カセット(ABC)トランスポーター阻害剤を添加してもなお薬剤排除能を有する細胞である。薬剤排除能を有する細胞とは、例えば、SP細胞であり、SP細胞とは、フローサイトメトリーでの解析でHoechst33342という蛍光色素を細胞に取り込ませてUVで励起すると405nmおよび600nmに蛍光を発する通常の細胞(未分化細胞以外の細胞)からサイトグラム上は異なった位置(蛍光の暗い部分、すなわち、“Hoechst Blue弱陽性かつHoechst Red弱陽性”)に出現する細胞集団のことである。従って、本発明における好ましい排出される薬剤は、Hoechst33342である。
本発明において、トリプシン処理とは、細胞とトリプシンを接触させることで、細胞間接着および細胞-培養器材間接着を解離させることを意味する。本発明においてトリプシン処理は、トリプシンを希釈して細胞と接触させることが好ましく、希釈率として、特に限定されないが、例えば、0.1〜1%(例:0.1%、0.15%、0.2%、0.25%、0.3%、0.35%、0.4%、0.45%、0.5%、0.55%、0.6%、0.65%、0.7%、0.75%、0.8%、0.85%、0.9%、0.95%および1%)が挙げられる。より好ましくは、0.25%である。
別の態様において、本発明はまた、前述の方法により得られたがん幹細胞を提供する。本発明の方法により作製され得る限りそのすべての細胞は、本発明のがん幹細胞の概念に包含される。
(a) 外来性のOct3/4、Sox2およびKlf4をコードする核酸を含み、
(b) がん幹細胞マーカーが陽性であり、および
(c) Oct3/4、Sox2およびKlf4、またはそれらをコードする核酸を含む因子を導入していないがん細胞の薬剤排除能を抑制するのに有効な濃度のABCトランスポーター阻害剤を添加した条件において、薬剤排除能を有する。
本発明によるがん幹細胞は、出願時において公知であった任意の方法を用いてがん細胞へ分化させることができる。分化誘導されるがん細胞は、頭頸部のがん(例えば、上顎がん、咽頭がん、喉頭がん、舌がん、甲状腺がん)、胸部のがん(例えば、乳がん、肺がん(非小細胞肺がん、小細胞肺がん))、消化器のがん(例えば、食道がん、胃がん、十二指腸がん、大腸がん(結腸がん、直腸がん)、肝がん(肝細胞がん、胆管細胞がん)、胆嚢がん、胆管がん、膵がん、肛門がん)、泌尿器のがん(例えば、腎がん、尿管がん、膀胱がん、前立腺がん、陰茎がん、精巣(睾丸)がん)、生殖器のがん(例えば、子宮がん(子宮頸がん、子宮体がん)、卵巣がん、外陰がん、膣がん)、皮膚のがん(例えば、基底細胞がん、有棘細胞がん)を含むが、これらに限定されない。本発明によるがん幹細胞は、目的に応じて、均一ながん細胞のみの集団、不均一な(多様性のある)がん細胞の集団、その中間的な集団など、種々の細胞集団に分化誘導され得る。
本発明は、前述のように得られたがん幹細胞を用いて、抗がん剤をスクリーニングする方法を提供する。本発明のスクリーニング方法は、例えば、前述のように得られたがん幹細胞を試験物質の存在下または非存在下で培養した後、Oct3/4、Sox2およびKlf4、またはそれらをコードする核酸を含む因子を導入していないがん細胞の薬剤排除能を抑制するのに有効な濃度のABCトランスポーター阻害剤を添加した条件において、薬剤排除能を有する細胞の生存度を測定し、試験物質の非存在下で培養した場合と比較して、試験物質の存在下で培養した場合において、当該細胞の生存度が低下した場合に、当該試験物質は抗がん作用を有すると判定することにより行われ得る。
東北大学の医用細胞資源センター(Cell Resource Center for Biomedical Research)から、2種類のヒト大腸がん細胞株(SW480およびDLD-1)を入手した。10% FBS(Life Technologies)、100 Units/mlペニシリン(Life Technologies)および100μg/mlストレプトマイシン(Life Technologies)を含有するDMEM培地(Nacalai Tesque)(以下、10%FBS-DMEMという)を用いて、入手した大腸がん細胞を培養した。
3因子を導入したSW480細胞が、がん幹細胞特性を獲得しているか否かを判定するために、以下の観点から検討した。
実施例1に記載のとおり3因子をSW480細胞に導入後10日目に、細胞の形態を観察した。その結果、導入された因子の種類ごとに、異なる形態的な特徴を示した(図1C)。具体的には、親細胞(Wt-SW480)やM-SW480細胞は、主に紡錘形の細胞や小さな丸型の細胞から構成され、O-SW480細胞は、不明瞭なエッジを有する球形のクラスターから構成され、S-SW480細胞は、丸型の細胞の数が増加しており、K-SW480細胞は、わずかに細胞境界不明瞭でやや扁平なコロニーであり、OSK-SW480細胞は、細胞境界不明瞭な扁平に盛り上がったコロニーを構成していた。
実施例1に記載のとおり3因子をSW480細胞に導入後10日目に、トリプシン-EDTAを用いて培養プレートから細胞を解離し、細胞を回収した。製造者の指示にしたがって、1% 2-メルカプトエタノールを含有するRLTバッファーにより細胞株からの全RNAを抽出し、RNeasy Plus Mini Kit(QIAGEN)で処理した。次いで、Transcriptor High Fidelity cDNA Synthesis Kit(Roche)により1μgの全RNAアリコートを逆転写した。Fast Start Universal SYBR Green Master Mix(Roche)を用いて、大腸がん幹細胞関連マーカーであるCD133、CD44、CD26、ALDH1、ABCG2およびLGR5について、StepOne real-time PCR system(Life Technologies)で解析した。用いたプライマー配列は、下記表1に示す。
各細胞の細胞増殖速度を測定した。詳細には、実施例1に記載のとおり3因子または陰性対照を導入後7日目に、トリプシン-EDTAを用いて培養プレートから細胞を解離し、細胞を回収した。3×105個の細胞を6ウェルプレートに播種し、11日目(96時間後)に細胞数を計測した。その結果、OSK-SW480細胞の数は、M-SW480細胞など他の細胞株と比較して有意に少ないことが見出された(図2D)。
3因子を導入されたSW480細胞について、抗がん剤である5-FUに対する感受性を検討するために、WST-8アッセイにより細胞生存率を測定した。詳細には、実施例1に記載のとおり3因子をSW480細胞に導入後10日目に、トリプシン-EDTAを用いて培養プレートから細胞を解離し、細胞を回収した。回収した5×103個の細胞を96ウェルプレートに播種し、播種の24時間後に、培地を、1μg/mlまたは50μg/mlの5-FUを含むDMEM培地に置換した。これらの5-FUの濃度は、実地臨床において使用される、ボーラス投与と持続投与における5-FU血中動態を参考として設定した。5-FU含有培地での48時間のインキュベーション後、マイクロタイタープレートリーダーを用いて450nmの吸光度を測定した。細胞生存率は、M-SW480細胞での吸光度に対する割合として算出した。その結果、5-FUを1μg/mlおよび50μg/mlのいずれの濃度で用いた場合でも、OSK-SW480細胞の生存度は、M-SW480細胞の生存度と比較して有意に高くなることが見出された(図2F)。
3因子を導入されたSW480細胞について、無血清浮遊培養条件下での増殖能を検討するために、スフィア形成アッセイを行った。詳細には、実施例1に記載のとおり3因子をSW480細胞に導入後10日目に、トリプシン-EDTAを用いて培養プレートから細胞を解離し、遠心分離を行った後、無血清DMEMで細胞を再懸濁した。次いで、1×104個の細胞を、10ng/ml bFGF(WAKO)、10μg/ml ヒトインスリン(CSTI)、100μg/ml ヒトトランスフェリンおよび100μg/ml BSAを含有する無血清DMEM を入れたUltra Low Attachment plates(Corning)に播種し、5% CO2インキュベータ内において、37℃で10日間インキュベートした後、形成されたスフィア数を計測した(図3A)。その結果、O-SW480、K-SW480およびOSK-SW480細胞を培養したときのスフィア数は、M-SW480細胞を培養したときのスフィア数と比較して有意に増加することが見出された(図3B)。
3因子を導入されたSW480のin vivoでの腫瘍形成能を検討するために移植実験を行った。実験は、京都大学の動物実験委員会の承認を得て行われた。詳細には、実施例1に記載のとおり3因子を導入後10日目に、トリプシン-EDTAを用いて培養プレートから細胞を解離し、細胞を回収した。次いで、100μl PBS中に懸濁し、1×106個の細胞または3×105個の細胞を免疫不全ヌードマウス(KSN/Slc mouse, SLC)の背部両側面に皮下注射した。各々の細胞数について、移植の4週間後および8週間後に、それぞれ腫瘍体積を測定した。腫瘍体積は、0.5×L×W2(L:長さ、W:幅)により計算した。その結果、いずれの細胞数の場合においても、K-SW480細胞およびOSK-SW480細胞を移植した場合、100%の腫瘍形成率であった。一方、対照群であるWt-SW480およびM-SW480ではともに、1×106個の場合:75%, 3×105個の場合:25%であった。
3因子を導入されたSW480の薬剤の排出能について、ABCトランスポーター阻害剤により排出能を抑制することで検討を行った。詳細には、実施例1に記載のとおり3因子をSW480細胞に導入後10日目に、トリプシン-EDTAを用いて培養プレートから細胞を解離し、細胞を回収した。1×106個の細胞をPBSで洗浄し、2% PBSおよび1mM HEPESを含有するDMEMで再懸濁した。細胞は、0μM、50μMまたは250μMのベラパミル(Sigma-Aldrich)、および5μg/mlのHoechst33342(Invitrogen)の存在下で、37℃で90分間インキュベートした。このとき、30分ごとに反転させることで穏やかに撹拌した。インキュベーション後、細胞を2% PBSおよび1mM HEPESを含有するPBSに再懸濁し、2μg/ml PIで対比染色して死細胞を標識し、40nmのメッシュフィルターを通過させ、ソーティングのために測定まで氷上で保管した。薬剤排出能は、Hoechst33342に対してUVレーザー(355nm)で励起させ、670/50フィルター(Hoechst Red)および450/50フィルター(Hoechst Blue)を透過する波長の蛍光をFACS Aria IIを用いて検出することによって評価した。その結果、OSK-SW480細胞は、50μMのベラパミル(VM)の存在下においても、Hoechst33342で標識されない細胞集団を含んでいることが見出された(図4A)。このような、0μMおよび50μMのベラパミルの存在下でHoechst33342により標識されない細胞をそれぞれV0細胞およびV50細胞と名付けた。V50細胞は、OSK-SW480細胞の培養物中においてのみ有意にその含有量が多いことが見出された(図4B)。ベラパミルの濃度を250μMまで引き上げると、OSK-SW480細胞培養物中に存在していたV50細胞は、消失した(図4C)。
他のヒト大腸がん細胞株(DLD-1)を用いて、3因子導入による形質転換を受けた細胞の増殖能(図5A)、薬剤排出能(図5B)、腫瘍形成能(図5C)を確認したところ、SW480と同様に、3因子導入によりin vitroでの細胞増殖能が低下し、V50細胞を含有し、形成腫瘍の体積および個数において腫瘍形成能が高いことが確認された。
OSK-SW480細胞培養物に見受けられたがん幹細胞特性が、実施例6で単離されたV50細胞に起因するものであるか否かを次の通り検討した。詳細には、実施例6と同様の方法により、OSK-SW480細胞中に存在するV50細胞(OSK-V50)、M-SW480細胞中のnon-V0細胞、V0細胞およびnon-V50細胞(それぞれ、M-nonV0、M-V0およびM-nonV50)、ならびにOSK-SW480細胞中のnon-V50細胞(OSK-nonV50)をソーティングし、各細胞集団を10日間培養した。その後、各細胞集団の細胞の形態を観察した。その結果、OSK-V50の形態は、OSK-SW480細胞で特徴的に観察されていたもの(図1Cの実線で囲まれた細胞)と類似していることが見出された(図6A)。対照的に、OSK-nonV50細胞の形態は、M-V0、M-nonV0およびM-nonV50細胞の形態と類似していた(図6A)。
OSK-SW480のV50細胞ががん幹細胞特性の一つであるがん組織における多様な細胞の形成能(多様性形成能)を有しているか否かを調べる実験を行った。詳細には、実施例6と同様の方法により、M-V0細胞、M-nonV50細胞、OSK-V50細胞およびOSK-nonV50細胞をソーティングし、各細胞集団を23日間培養した後、FACS解析を行った。その結果、M-V0細胞およびM-nonV50細胞を培養しても、V50細胞が生じることはなかったが、OSK-V50細胞は、培養後もV50細胞(7.8%)を含み、さらにnon-V50細胞も含むことが確認された(図7A)。これらの結果から、V50細胞は、薬剤排出能について多様な種類の細胞を産生する能力を有していることが明らかとなった。
OSK-V50細胞に由来する腫瘍について評価するために、免疫組織化学による実験を行った。本実験による評価は、OSK-V50由来の腫瘍とM-SW480由来の腫瘍を比較することにより行った。
OSK-V50細胞が、in vivoにおける継代を繰り返しても、腫瘍形成能を有しかつ実施例10で示された組織学的にヒト大腸がん組織特異性を再現する能力を有するかを検討した。実施例6と同様の方法によりソーティングにより得られたM-nonV50またはOSK-V50をin vitroで培養した後(1st Culture)、3×106個の細胞を免疫不全ヌードマウスに皮下注射した。続いて、得られた腫瘍(1st Tumor)を摘出し、腫瘍片をgentleMACS Dissociator とHuman Tumor Dissociation Kit (ともにMiltenyi Biotec)を用いて単一細胞に解離させ、培養した(2nd Culture)。培養後、3×106個の細胞を免疫不全ヌードマウスに皮下注射した。同様に、得られた腫瘍(2nd Tumor)を摘出し、解離させ、培養した(3rd Culture)。さらに、得られた3×106個の細胞を免疫不全ヌードマウスに皮下注射し、腫瘍(3rd Tumor)を形成させた。同様に、腫瘍を摘出し、解離させ、培養した(4th Culture)(図9A)。このようにして得られた1st Culture、2nd Culture、3rd Culture、4th Cultureを顕微鏡で観察したところ、OSK-V50では、細胞境界不明瞭な扁平隆起したコロニーの出現と多様な細胞から構成されることが観察され(図9B)、また培養後の細胞数も少ない(細胞増殖速度が低い)ことが確認された(図9C)。これらの挙動は、実施例9と同様の結果であることから、連続移植により形成された腫瘍から得られた細胞においてもV50細胞の形態的特徴は保持されていることが示唆された。さらに、2nd Culture、3rd Culture、4th Cultureの細胞中のV50細胞の含有率を測定したところ、OSK-V50では、そのいずれの腫瘍中にも一定量(1-1.6%)のV50細胞を含有していることが明らかとなり、in vitroと異なってin vivoではV50細胞の含有率が維持できることが確認された(図9D)。
実施例11と同様に用意した3rd Cultureを実施例6と同様の方法でソーティングし、再度V50細胞およびnon-V50細胞を得た。これらの細胞を4日間培養後それぞれ免疫不全ヌードマウスに皮下注射し、腫瘍(3rd Tumor)を形成させた。実施例11と同様に、それぞれの腫瘍を切除し、解離させ、培養した(4th Culture)(図10A)。3rd CultureにおけるV50細胞およびnon-V50細胞の培養後の細胞数を測定したところ、これまでと同様にV50細胞でその数が少ないことが確認された(図10B)。また、このV50細胞およびnon-V50細胞を移植せず培養を継続し、形態を観察したところ、実施例9と同様に、V50細胞には、細胞境界不明瞭で扁平隆起したコロニーの出現と形態的に多様な細胞集団構成が、経時的に観察された(図10C)。
ヒト肺がん細胞株(A549)は、理化学研究所バイオリソースセンターから入手可能である。10% FBS(Life Technologies)、100 Units/mlペニシリン(Life Technologies)および100μg/mlストレプトマイシン(Life Technologies)を含有するDMEM培地(Nacalai Tesque)(以下、10%FBS-DMEMという)を用いて、入手した肺がん細胞を培養した。
In-Fusion HD Cloning Kit(タカラ)を用いて、OCT3/4をコードする核酸、SOX2をコードする核酸およびKLF4をコードする核酸を同時に有するベクター(以下、OKSベクターという)を作製した(図17)。なお、遺伝子間は2A領域で繋いで作成した。
Claims (19)
- がん幹細胞を製造する方法であって、下記の工程:
(a) Oct3/4、Sox2およびKlf4、またはそれらをコードする核酸を含む因子をがん細胞に導入する工程、および
(b) 工程(a)により得られた細胞を胚性幹(ES)細胞の維持培養条件以外の条件で培養する工程
を含み、前記工程(b)により得られた細胞集団は、Oct3/4、Sox2およびKlf4、またはそれらをコードする核酸を含む因子を導入していないがん細胞の薬剤排除能を抑制するのに有効な濃度のATP結合カセット(ABC)トランスポーター阻害剤を添加した条件において、薬剤排除能を有する細胞を含有することを特徴とする、方法。 - 以下の工程(c)をさらに含む、請求項1に記載の方法:
(c)工程 (b)により得られた細胞から、Oct3/4、Sox2およびKlf4、またはそれらをコードする核酸を含む因子を導入していないがん細胞の薬剤排除能を抑制するのに有効な濃度のABCトランスポーター阻害剤を添加した条件において、薬剤排除能を有する細胞を抽出する工程。 - 前記薬剤排除能がHoechst33342を排除する能力である、請求項1または2に記載の方法。
- 前記ABCトランスポーター阻害剤が、ベラパミルである、請求項1から3のいずれか1項に記載の方法。
- 薬剤排除能を抑制するのに有効な濃度が50μM以上、250μM未満である、請求項4に記載の方法。
- がん幹細胞を製造する方法であって、下記の工程:
(a) Oct3/4、Sox2およびKlf4、またはそれらをコードする核酸を含む因子をがん細胞に導入する工程、
(b) 工程(a)により得られた細胞を胚性幹(ES)細胞の維持培養条件以外の条件で培養する工程、
(c) 工程(b)により得られた細胞をトリプシンで処理する工程、および
(d) 工程(c)により解離しなかった細胞をがん幹細胞として取得する工程
を含む方法。 - 前記トリプシンの処理が、0.25%トリプシンで5分間以上の処理である、請求項6に記載の方法。
- 前記がん細胞が、ヒト由来のがん細胞である、請求項1から7のいずれか1項に記載の方法。
- 請求項1から8のいずれか1項に記載の方法により作製されたがん幹細胞。
- 下記の特徴を有する、がん幹細胞:
(a) 外来性のOct3/4、Sox2およびKlf4をコードする核酸を含み、
(b) CD133、CD44、CD26、ABCG2およびLGR5が陽性であり、および
(c) Oct3/4、Sox2およびKlf4、またはそれらをコードする核酸を含む因子を導入していないがん細胞の薬剤排除能を抑制するのに有効な濃度のABCトランスポーター阻害剤を添加した条件において、薬剤排除能を有する。 - 前記薬剤排除能がHoechst33342を排除する能力である、請求項10に記載の細胞。
- 前記ABCトランスポーター阻害剤が、ベラパミルである、請求項10または11に記載の細胞。
- 前記薬剤排除能を抑制するのに有効な濃度が50μM以上、250μM未満である、請求項12に記載の細胞。
- 前記がん幹細胞が、大腸がん幹細胞である、請求項10から13のいずれか1項に記載の細胞。
- 以下の工程を含む、抗がん剤をスクリーニングする方法。
(1)請求項9から14のいずれか1項に記載の細胞と候補薬剤とを接触させる工程、
(2)Oct3/4、Sox2およびKlf4、またはそれらをコードする核酸を含む因子を導入していないがん細胞の薬剤排除能を抑制するのに有効な濃度のABCトランスポーター阻害剤を添加した条件において、薬剤排除能を有する細胞数を測定する工程、および
(3)工程(2)で測定された細胞数が、候補薬剤と接触させなかったときの細胞数より減少した場合、当該候補薬剤を抗がん剤として選択する工程。 - 前記薬剤排除能がHoechst33342を排除する能力である、請求項15に記載の方法。
- 前記ABCトランスポーター阻害剤が、ベラパミルである、請求項15または16に記載の方法。
- 前記薬剤排除能を抑制するのに有効な濃度が50μM以上、250μM未満である、請求項15から17のいずれか1項に記載の方法。
- 以下の工程を含む、抗がん剤をスクリーニングする方法。
(1)請求項6または7に記載の方法で製造された細胞と候補薬剤とを接触させる工程、
(2)細胞数を測定する工程、および
(3)工程(2)で測定された細胞数が、候補薬剤と接触させなかったときの細胞数より減少した場合、当該候補薬剤を抗がん剤として選択する工程。
