なお、本出願は2013年10月30日に出願された日本国特許出願2013−225922号に基づく優先権を主張しており、当該日本国出願の全内容は本明細書中に参照として援用されている。
例えば、ゲノム不安定なiPS細胞等の多能性幹細胞は生体内への移植に不適なため、再生医療に利用可能なiPS細胞等の多能性幹細胞を得るためには対象の多能性幹細胞集団の中からゲノム不安定な多能性幹細胞を除去する必要がある。上記ゲノム不安定な多能性幹細胞の除去に際しては、当然、多能性幹細胞のゲノム安定性を評価する必要がある。
そして、種々の多能性幹細胞におけるカルレティキュリンの発現量を調べたところ、驚くべきことに、ゲノム不安定な多能性幹細胞(例えばiPS細胞)はゲノム安定な同種の幹細胞と比較してカルレティキュリンの発現量が顕著に増大していることを見出し、本発明を完成するに至った。
抗カルレティキュリン抗体を用いた免疫学的手法によると、対象の多能性幹細胞培養物中(典型的にはiPS細胞培養物中若しくはES細胞培養物中)の多能性幹細胞におけるカルレティキュリンの発現を、特異的かつ高感度に調べることができる。したがって、iPS細胞等の多能性幹細胞のゲノム安定性若しくはゲノム不安定性を高精度且つ高い信頼性をもって判別することが可能となる。また、免疫学的手法を用いた多能性幹細のゲノム安定性の評価は、従来のゲノム安定性を評価する方法(例えばFISH法)と比較して短時間かつ簡便な操作でiPS細胞等の多能性幹細胞のゲノム安定性を評価し得るため好ましい。
本発明者は上記知見に基づき、本発明の他の側面として、ゲノム不安定な多能性幹細胞(典型的にはiPS細胞若しくはES細胞)に対してカルレティキュリン発現誘導活性を奏する人為的に合成されたペプチド(以下、「カルレティキュリン発現誘導ペプチド」という)を完成するに至った。
具体的には、本発明者は、以下のアミノ酸配列:
CRAKAGDPC(配列番号1);および
CEQKQEIRC(配列番号2);
のうちのいずれか若しくは該アミノ酸配列において1個又は数個(典型的には2個又は3個)のアミノ酸残基が置換、欠失及び/又は付加されて形成された改変アミノ酸配列のいずれかからなるカルレティキュリン発現誘導ペプチド配列を含む合成ペプチドを提供する。
上記改変アミノ酸配列からなるカルレティキュリン発現誘導ペプチド配列もまた、配列番号1又は2に示すカルレティキュリン発現誘導ペプチド配列と同様に良好なカルレティキュリン発現誘導活性を発揮することができる。
このような膜透過性ペプチドを有する上記カルレティキュリン発現誘導ペプチドは、カルレティキュリン発現誘導ペプチド配列を高効率にiPS細胞等の多能性幹細胞内(細胞膜及び/又は核膜の内側)に移入することができるため、本発明の実施に好適に用いることができる。

配列番号5としてここで開示されるアミノ酸配列は、膜透過性ペプチドを構成するアミノ酸配列の典型例であり、特にゲノム不安定な多能性幹細胞(典型的にはiPS細胞)におけるカルレティキュリン発現を高効率に誘導することができる。
このような短いペプチド鎖のペプチドは化学合成が容易であり、且つ、安価で取扱い性に優れるため、本発明の実施に特に好適に用いることができる。

かかる合成ペプチドは、特に、ヒト由来のゲノム不安定なiPS細胞に対してカルレティキュリン発現誘導活性を奏する。ヒト由来のiPS細胞を好適な適用対象とする上記合成ペプチドは、医療産業上の利用価値が極めて高い。
対象の多能性幹細胞培養物中(典型的にはiPS細胞培養物中若しくはES細胞培養物中)からゲノム不安定と判別された多能性幹細胞を除去することで、ゲノム安定と判別された多能性幹細胞を主体とする(好ましくはゲノム安定と判別された多能性幹細胞からなる)多能性幹細胞培養物(例えばiPS細胞培養物)を作製することができる。そのため、上記ゲノム不安定な多能性幹細胞の除去方法は、再生医療に利用可能なiPS細胞等の多能性幹細胞培養物の作製に好適に採用できる。また、上記多能性幹細胞のゲノム安定性評価方法によって対象の多能性幹細胞のゲノム安定性若しくは不安定性を判別することにより、高効率且つ高精度にゲノム不安定な多能性幹細胞(好ましくはiPS細胞)の除去を実現することができる。
細胞選別機(セルソーター)を使用することで、多能性幹細胞(典型的にはiPS細胞若しくはES細胞)を含む培養物中からゲノム不安定性であると判別された多能性幹細胞を効率よく除去することができる。そのため、細胞選別機を使用したゲノム不安定な多能性幹細胞の除去方法は、特に、多量の多能性幹細胞(典型的にはiPS細胞)を含む培養物からの細胞除去に好適である。
かかる製造方法により製造された多能性幹細胞培養物(典型的にはiPS細胞培養物若しくはES細胞培養物)は、上記除去方法によりゲノム不安定な多能性幹細胞が除去された多能性幹細胞培養物であるため、当該培養物中に含まれる実質的に全ての多能性幹細胞がゲノム安定な多能性幹細胞である多能性幹細胞培養物として把握され得る。該多能性幹細胞培養物(典型的には該多能性幹細胞培養物中の多能性幹細胞)は、腫瘍化のリスクが低い再生医療用多能性幹細胞として好適に利用することができる。特にヒト由来のiPS細胞を含む培養物を製造する上記方法は、医療産業上の利用価値が極めて高いため好ましい一実施形態である。
また、典型的には、上記カルレティキュリン発現誘導剤は、薬学上許容され得る少なくとも1種の担体(例えば上記ペプチドの安定性向上に資する少なくとも1種の基材、或いは生理食塩水や各種の緩衝液等の液状媒体)を含む。
かかるカルレティキュリン発現誘導剤は、単純な構造(直鎖状のペプチド鎖)の合成ペプチドを有効成分として含むため、例えば、iPS細胞等の多能性幹細胞(典型的には当該幹細胞培養物の培地中)に該カルレティキュリン発現誘導剤を供給するという簡易な処理を行うことによって、ゲノム不安定な多能性幹細胞のカルレティキュリン発現量を増大させることができる。これにより、上記幹細胞におけるカルレティキュリンの発現の程度に基づいて、当該幹細胞のゲノム安定性若しくはゲノム不安定性を高精度且つ高信頼性に判別可能となる。また、化学合成(若しくは生合成)によって容易に人為的に製造され得る合成ペプチドを有効成分として含むため、所望量のカルレティキュリン発現誘導剤を容易に調製することができる。なお、上記カルレティキュリン発現誘導剤は、特にゲノム不安定なヒト由来のiPS細胞のカルレティキュリンの発現量を増大させるために好適に用いることができる。

また、本明細書において「ペプチド」とは、複数のペプチド結合を有するアミノ酸ポリマーを指す用語であり、ペプチド鎖に含まれるアミノ酸残基の数によって限定されないが、典型的には全アミノ酸残基数が概ね100以下(好ましくは60以下、例えば50以下)のような比較的分子量の小さいものをいう。
また、本明細書において「アミノ酸残基」とは、特に言及する場合を除いて、ペプチド鎖のN末端アミノ酸及びC末端アミノ酸を包含する用語である。
なお、本明細書中に記載されるアミノ酸配列は、常に左側がN末端側であり右側がC末端側である。
なお、本明細書においてiPS細胞の由来は特に限定されないが、特にヒト由来のiPS細胞は医療産業上の利用価値が高いため本発明の実施対象として特に好ましい。
上記カルレティキュリン発現誘導ペプチドを対象の多能性幹細胞(例えばiPS細胞)に供給し、所定時間培養することで、ゲノム不安定な多能性幹細胞におけるカルレティキュリンの発現量を特に増大させることができる。これにより、ゲノム不安定な多能性幹細胞とゲノム安定な多能性幹細胞とのカルレティキュリン発現量の差を、カルレティキュリン発現誘導ペプチドを添加しない場合と比較してより明確にすることができる。そのため、該カルレティキュリンの発現量の程度に基づいた多能性幹細胞のゲノム安定性若しくはゲノム不安定性の判別を高精度且つ高い信頼性で実現可能となる。
なお、上記カルレティキュリン発現誘導ペプチドは、ゲノム不安定なヒト由来のiPS細胞のカルレティキュリン発現量を顕著に増大し得る。そのため、ここで開示されるiPS細胞のゲノム安定性評価方法は、特にヒト由来のiPS細胞について好適に実施することができる。
CRAKAGDPC(配列番号1);および
CEQKQEIRC(配列番号2);
のうちのいずれか若しくは該アミノ酸配列において1個、又は2個、又は3個のアミノ酸残基が置換、欠失及び/又は付加されて形成された改変アミノ酸配列のいずれかからなるカルレティキュリン発現誘導ペプチド配列を含む合成ペプチドである。配列番号1又は配列番号2に示される具体的なアミノ酸配列は、ヒト由来のセントリン2のsiRNAを構成するRNA配列を翻訳して得られるアミノ酸配列(以下「セントリン2siRNA関連配列」ともいう)であることに加え、ゲノム不安定なiPS細胞等の多能性幹細胞におけるカルレティキュリンの発現を誘導する或いはカルレティキュリンの発現量を増大させることが本発明者によって新たに見出された配列である。
ここで、セントリンとは、真核生物の中心体に存在し、中心子の構成タンパク質の一つとして中心子の複製や微小管の切断に関与する中心体関連タンパク質であり、セントリン2とは、セントリンファミリー(典型的には、セントリン1、セントリン2、セントリン3等)に属するタンパク質のうちの一つである(非特許文献3参照)。
RAKAGDP(配列番号3);および
EQKQEIR(配列番号4);
が挙げられる。配列番号3又は配列番号4に示すアミノ酸配列は、配列番号1又は配列番号2に示すアミノ酸配列のN末端及びC末端のシステイン残基(C)を欠失した配列である。ここに挙げるセントリン2siRNA関連配列の改変アミノ酸配列もまた、カルレティキュリン発現誘導ペプチド配列として好適に機能する。なお、配列番号3および配列番号4に示した上述の改変アミノ酸配列はあくまでも例示であり、使用可能なセントリン2siRNA関連配列の改変アミノ酸配列をかかる配列に限定することを意図するものではない。
上記膜透過性ペプチド配列は、細胞膜及び/又は核膜を通過し得る膜透過性ペプチドを構成するアミノ酸配列であれば特に限定なく使用することができる。多くの好適な膜透過性ペプチド配列が知られているが、例えばNoLS(核小体局在シグナル、Nucleolar localization signal)に関連するアミノ酸配列(改変アミノ酸配列を含む)がカルレティキュリン発現誘導ペプチドの膜透過性ペプチド配列のアミノ酸配列として好ましい。例えば、配列番号5に示すLIMキナーゼ2(LIM Kinase 2)に含まれるNoLS並びに配列番号6に示すIBV(トリ伝染性気管支炎ウイルス:avian infectious bronchitis virus)のNタンパク質(nucleocapsid protein)に含まれるNoLSのアミノ酸配列が挙げられる。また、他の膜透過性ペプチド配列の例として、配列番号7〜9に示すアミノ酸配列およびそれらの改変アミノ酸配列(膜透過性を保持しているものに限られる)が挙げられる。配列番号7は、HIV(ヒト免疫不全ウイルス:Human Immunodeficiency Virus)のTATに含まれる膜透過性ペプチド配列のアミノ酸配列を示している。配列番号8は、上記TATを改変した膜透過性ペプチド配列(PTD4)のアミノ酸配列を示している。配列番号9は、ショウジョウバエ(Drosophila)の変異体であるAntennapediaのANT関連アミノ酸配列を示している。
なお、配列表に示した上述の膜透過性ペプチド配列はあくまでも例示であり、使用可能なペプチド配列はこれに限定されない。本発明の実施に使用可能な様々な膜透過性ペプチド配列が本願出願当時に出版されている数々の文献に記載されている。それら膜透過性ペプチド配列のアミノ酸配列は一般的な検索手段によって容易に知ることができる。

のうちから選択されるいずれかのアミノ酸配列または該選択されたアミノ酸配列の改変アミノ酸配列を特に好ましく含有する。配列番号10又は配列番号11に示すアミノ酸配列は、上記配列番号1又は配列番号2に示すヒト由来のセントリン2siRNA関連配列と、上記配列番号5に示すLIMキナーゼ2のNoLS由来のアミノ酸配列とを、2個のグリシン(G)残基から成るリンカーを介して組み合わせることにより構築された合計24アミノ酸残基からなるアミノ酸配列である。また、配列番号12又は配列番号13に示すアミノ酸配列は、上記配列番号3又は配列番号4に示すヒト由来のセントリン2siRNA関連配列の典型的な改変アミノ酸配列と、上記配列番号5に示すLIMキナーゼ2のNoLS由来のアミノ酸配列とを、2個のグリシン(G)残基から成るリンカーを介して組み合わせることにより構築された合計22アミノ酸残基からなるアミノ酸配列である。
ここで開示されるカルレティキュリン発現誘導ペプチドは、少なくとも一つのアミノ酸残基がアミド化されているものが好ましい。アミノ酸残基(典型的にはペプチド鎖のC末端アミノ酸残基)のカルボキシル基のアミド化により、合成ペプチドの構造安定性(例えばプロテアーゼ耐性)を向上させることができる。
このような鎖長の短いペプチドは、化学合成が容易であり、容易にカルレティキュリン発現誘導ペプチドを提供することができる。なお、ペプチドのコンホメーション(立体構造)については、使用する環境下(生体外若しくは生体内)でカルレティキュリン発現誘導活性を発揮する限りにおいて、特に限定されるものではないが、免疫原(抗原)になり難いという観点から直鎖状又はヘリックス状のものが好ましい。このような形状のペプチドはエピトープを構成し難い。かかる観点から、本発明に適用するカルレティキュリン発現誘導ペプチドとしては、直鎖状であり比較的低分子量(典型的には60以下(特に30以下)のアミノ酸残基数)のものが好適である。
なお、本発明のカルレティキュリン発現誘導ペプチドとしては、全てのアミノ酸残基がL型アミノ酸であるものが好ましいが、カルレティキュリン発現誘導活性を失わない限りにおいて、アミノ酸残基の一部又は全部がD型アミノ酸に置換されているものであってもよい。
ここで開示されるカルレティキュリン発現誘導ペプチドは、市販のペプチド合成機(例えば、Intavis AG社、Protein Technologies社等から入手可能である。)を用いた固相合成法により、所望するアミノ酸配列、修飾(C末端アミド化等)部分を有するペプチド鎖を合成することができる。
一般的な技法によって、この組換えベクターを所定の宿主細胞(例えばイースト、昆虫細胞、植物細胞)に導入し、所定の条件で当該宿主細胞又は該細胞を含む組織や個体を培養する。このことにより、目的とするペプチドを細胞内で発現、生産させることができる。そして、宿主細胞(分泌された場合は培地中)からペプチドを単離し、必要に応じてリフォールディング、精製等を行うことによって、目的のカルレティキュリン発現誘導ペプチドを得ることができる。
なお、組換えベクターの構築方法及び構築した組換えベクターの宿主細胞への導入方法等は、当該分野で従来から行われている方法をそのまま採用すればよく、かかる方法自体は特に本発明を特徴づけるものではないため、詳細な説明は省略する。
或いは、無細胞タンパク質合成システム用の鋳型DNA(即ちカルレティキュリン発現誘導ペプチドのアミノ酸配列をコードするヌクレオチド配列を含む合成遺伝子断片)を構築し、ペプチド合成に必要な種々の化合物(ATP、RNAポリメラーゼ、アミノ酸類等)を使用し、いわゆる無細胞タンパク質合成システムを採用して目的のポリペプチドをインビトロ(生体外、in vitro)合成することができる。無細胞タンパク質合成システムについては、例えばShimizuらの論文(Shimizu et al., Nature Biotechnology, 19, 751-755(2001))、Madinらの論文(Madin et al., Proc. Natl. Acad. Sci. USA, 97(2), 559-564(2000))が参考になる。これら論文に記載された技術に基づいて、本願出願時点において既に多くの企業がポリペプチドの受託生産を行っており、また、無細胞タンパク質合成用キット(例えば、日本の東洋紡績(株)から入手可能なPROTEIOS(商標)Wheat germ cell-free protein synthesis kit)が市販されている。
こうして得られるポリヌクレオチドは、上述のように、種々の宿主細胞中で又は無細胞タンパク質合成システムにて、カルレティキュリン発現誘導ペプチド生産のための組換え遺伝子(発現カセット)を構築するための材料として使用することができる。
カルレティキュリン発現誘導剤の形態に関して特に限定はない。例えば、典型的な形態として、液剤、懸濁剤、乳剤、エアロゾル、泡沫剤、顆粒剤、粉末剤、錠剤、カプセル、軟膏、水性ジェル剤等が挙げられる。また、使用直前に生理食塩水又は適当な緩衝液(例えばPBS、即ちリン酸緩衝生理食塩水)等に溶解して薬液を調製するための凍結乾燥物、造粒物とすることもできる。
なお、カルレティキュリン発現誘導ペプチド(主成分)及び種々の担体(副成分)を材料にして種々の形態の薬剤(組成物)を調製するプロセス自体は従来公知の方法に準じればよく、かかる製剤方法自体は本発明を特徴づけるものでもないため詳細な説明は省略する。処方に関する詳細な情報源として、例えばComprehensive Medicinal Chemistry, Corwin Hansch監修,Pergamon Press刊(1990)が挙げられる。この書籍の全内容は本明細書中に参照として援用されている。
例えば、インビトロで培養(継代)している多能性幹細胞(典型的にはiPS細胞若しくはES細胞)に対し、ここで開示されるカルレティキュリン発現誘導剤(即ち、カルレティキュリン発現誘導ペプチド)の適当量を、培養過程のいずれかの段階(好ましくはゲノム安定性の評価を行うより前の段階)で培地に添加するとよい。添加量及び添加回数は、多能性幹細胞の種類や状態、細胞密度(培養開始時の細胞密度)、継代数、培養条件、培地の種類、等の条件によって異なり得るため特に限定されない。典型的には、培地中のペプチド濃度が概ね0.1μM〜100μMの範囲内、好ましくは0.5μM〜20μM(例えば1μM〜10μM)の範囲内となるように、1〜複数回添加する(例えば培養開始時並びに細胞の継代時や培地交換時に合わせて追加添加する)ことが好ましい。
また、ここで開示されるカルレティキュリン発現誘導剤(即ち、カルレティキュリン発現誘導ペプチド)は、ゲノム安定性の評価に利用可能な多能性幹細胞の他の特徴(即ち、カルレティキュリン発現量ではない多能性幹細胞の特徴)を増強し得る他の化合物及び/又は該他の特徴を増強する方法と併用することも可能である。
ゲノム不安定であると判別されたiPS細胞等の多能性幹細胞を対象の多能性幹細胞培養物中から除去することで、ゲノム安定な多能性幹細胞を主体とする多能性幹細胞培養物(例えばiPS細胞培養物)を製造することができる。例えば、培養物中に含まれる多能性幹細胞の大部分(例えば80%以上、好ましくは90%以上、より好ましくは98%以上)をゲノム安定な多能性幹細胞が占める多能性幹細胞培養物を製造することができる。培養物中に含まれる実質的に全ての多能性幹細胞がゲノム安定な多能性幹細胞からなる(典型的には、ゲノム安定な多能性幹細胞が実質的に100%を占める)多能性幹細胞培養物は、好適な一態様である。特に、ヒト由来のiPS細胞は医療産業上の利用価値が高いため、上記対象の多能性幹細胞の好適例である。
また、上記多能性幹細胞のゲノム安定性評価方法で多能性幹細胞(典型的にはiPS細胞若しくはES細胞)のカルレティキュリン発現量を把握するために用いた目印(マーカー、標識、特徴)、例えばカルレティキュリン発現量を測定するために用いた抗体の標識等、を当該除去方法においてもそのまま利用することができる。ゲノム安定性評価方法で用いた目印をそのまま利用すれば、ゲノム不安定であると判別されたiPS細胞等の多能性幹細胞の除去に必要な操作を簡便化することができるため好ましい。
上記抗体の標識としては、例えば、蛍光色素、マグネティックビーズ、GSTタグやHisタグといった各種のアフィニティータグ等を利用することができる。典型的には、蛍光標識した抗カルレティキュリン抗体若しくは蛍光標識した二次抗体を用いた免疫蛍光抗体法によりカルレティキュリンを蛍光標識し、該蛍光標識を目印とすることで、ゲノム不安定であると判別されたiPS細胞等の多能性幹細胞を対象の多能性幹細胞培養物中から高精度かつ高効率に除去することができる。
例えば、カルレティキュリン発現量が所定レベルを上回った多能性幹細胞を対象の多能性幹細胞培養物中(典型的にはiPS細胞培養物中若しくはES細胞培養物中)から除去することで、ゲノム不安定であると判別された多能性幹細胞を除去することができる。ゲノム不安定な多能性幹細胞を対象の多能性幹細胞培養物中から除去する際に利用可能なカルレティキュリンの発現レベルは、ゲノム不安定であると判別された多能性幹細胞の除去が可能であれば特に限定されない。典型的には、ゲノム安定性評価方法におけるカルレティキュリンの発現レベルと同程度の発現レベルを利用することができる。例えば、ゲノム安定なiPS細胞等の多能性幹細胞(比較対象)におけるカルレティキュリン発現量に比べて好ましくは1.2倍以上(例えば1.5倍以上、より好ましくは2倍以上、さらに好ましくは5倍以上、例えば10倍以上)のカルレティキュリン発現量をゲノム不安定な多能性幹細胞を除去する基準とすることができる。なお、ゲノム不安定な多能性幹細胞を評価対象の多能性幹細胞培養物中から除去する際に利用可能なカルレティキュリンの発現レベルは、由来が同じ多能性幹細胞を用いて設定することが好ましい。
なお、上記ゲノム不安定であると判別されたiPS細胞等の多能性幹細胞の除去は、上記対象の多能性幹細胞培養物中の多能性幹細胞のゲノム安定性若しくはゲノム不安定性の判別と同時に行ってもよい。上記判別と上記除去を同時に行うと、ゲノム不安定な多能性幹細胞の除去に必要な操作を簡便化することができるため好ましい。
以下の方法で上記iPS細胞(201B7及び201B2)を継代維持することで、iPS細胞の培養物を準備した。詳細は以下のとおりである。
次に、各iPS細胞をそれぞれCTK溶液(0.1mg/mLのコラゲナーゼIV(Life Technologies社製品)、1mMの塩化カルシウム、20%のKSR(knockout serum replacement)を含む0.25%トリプシン溶液)を用いて、iPS細胞のコロニー辺縁部が剥離する程度に剥離処理した。CTK溶液をPBS(−)で洗浄除去し、ES培地を添加した後、セルスクレーパーを用いてiPS細胞コロニーを完全に剥離した。15mL容チューブ中にiPS細胞コロニーが懸濁した培養容器中のESC培地を移し、さらに、培養容器に残ったiPS細胞コロニーをESC培地で洗い込んで該15mL容チューブに回収した。上記15mL容チューブを5分間静置してiPS細胞コロニーを沈殿させ、上澄みを取り除いた後、新鮮なES培地中にピペッティングにてiPS細胞コロニーを分散懸濁し、iPS細胞懸濁液を用意した。
このようにして得られたiPS細胞の懸濁物の適量(例えば、同容量の培養容器を用いて継代培養する場合は細胞懸濁液の全量の1/3〜1/8量程度)を上記のとおり作製しておいた培養容器中のフィーダー細胞上に播種した。そして培養容器をインキュベータに入れ、5%CO2、37℃の条件下で培養した。1〜2日毎に培地交換を行い、iPS細胞が培養容器底面の80〜90%を占める状態(即ち、80〜90%コンフルエント)になるまで培養を継続した。上記80〜90%コンフルエントになったiPS細胞を上記の方法で継代培養することを繰り返した。このようにして、iPS細胞の培養物を準備した。
ゲノム安定なiPS細胞とゲノム不安定なiPS細胞について、カルレティキュリンの発現を調べた。供試細胞として、上記2種のiPS細胞(201B7及び201B2)を用いた。評価試験の詳細は以下のとおりである。
具体的には、まず、各iPS細胞の培養容器中の培地を除去し、PBS(−)で洗浄した。次いで、メタノール1容量とアセトン1容量とを混和した溶液(メタノール/アセトン=1:1溶液)を添加し、氷上に15分静置してiPS細胞を固定した。その後、メタノール/アセトン=1:1溶液を除去し、3%のBSA含有PBS(−)(3%のBSAを含むPBS(−))を添加して室温で1時間のブロッキングを行った。所定時間経過後、3%のBSA含有PBS(−)を除去し、PBS(−)にて洗浄した。
そして、一次抗体として抗カルレティキュリンモノクローナル[FMC75]抗体(マウス由来、Abcam社製、Cat No. ab22683、Lot No. GR56669-4)を1%BSA/PBS(−)(1%のBSAを含むPBS(−))で最終濃度:4×10−3mg/mLに調製した一次抗体希釈液を上記iPS細胞の培養容器中に添加し、4℃で一晩若しくは室温で2時間静置した。かかる抗原抗体反応の所定時間経過後、一次抗体希釈液を除去し、PBS(−)で洗浄した。次いで、二次抗体として蛍光色素(Alexa(登録商標)488)で標識した抗マウスIgG抗体(ヤギ:Life Technologies社製品、A11001)を1%BSA/PBS(−)で最終濃度:10×10−3mg/mLに調製した二次抗体希釈液を添加し、室温で2時間静置した。上記所定時間経過後、二次抗体希釈液を除去し、PBS(−)で洗浄した。その後、DAPI(4',6-diamidino-2-phenylindole)含有封入液(Life Technologies社製)とカバーガラスを用いて封入を行い、共焦点レーザー顕微鏡による蛍光観察を行った。
このことは、対象の多能性幹細胞の培養物中の多能性幹細胞についてカルレティキュリン発現量を調べ、該カルレティキュリン発現量の程度に基づいて該幹細胞のゲノム安定性若しくはゲノム不安定性を判別できることを示している。また、上記判別には免疫学的手法が好適に使用できることも示している。
配列番号10〜13のアミノ酸配列から成る合成ペプチドを後述のペプチド合成機を用いて製造した。なお、以下の説明では、当該合成ペプチドを配列番号の順番に対応してサンプル1〜4と呼称する。表1には、これら合成ペプチドのアミノ酸配列等の情報を列挙している。
サンプル1又はサンプル2に係るペプチド(配列番号10又は11)は、カルレティキュリン発現誘導ペプチド配列として配列番号1又は2に示すセントリン2siRNA関連配列をそれぞれ有する合計24アミノ酸残基から成るペプチドである。
サンプル3又はサンプル4に係るペプチド(配列番号12又は13)は、カルレティキュリン発現誘導ペプチド配列として配列番号3又は4に示すセントリン2siRNA関連配列の典型的な改変配列をそれぞれ有する合計22アミノ酸残基から成るペプチドである。
合成したサンプル1〜4に係るペプチドは、PBS(-)に溶かし、ペプチドストック液を調製した。
上記実施例3で得られたサンプル1〜4に係るペプチドについて、カルレティキュリン発現誘導活性を調べた。供試細胞として、上記iPS細胞(201B7及び201B2)を用いた。評価試験の詳細は以下のとおりである。
一方、201B7にサンプル1に係る合成ペプチドを添加して培養した場合(図4)は、当該細胞のペプチド無添加区(図6)と比較してカルレティキュリンを検出する蛍光標識の蛍光発色はほとんど増大しないことが確認された。即ち、ペプチド無添加区における201B7と201B2とのカルレティキュリン発現量を比較するよりも、サンプル1添加区における201B7と201B2とのカルレティキュリン発現量を比較した方が、ゲノム安定性の異なるiPS細胞間のカルレティキュリン発現量の差を明確に把握することができた。また、ここには図示していないが、サンプル2〜4に係る合成ペプチドもまた、サンプル1と同様に201B7におけるカルレティキュリンの発現量をほとんど増大させず、当該ペプチドを対象のiPS細胞培養物に添加して所定時間培養することでゲノム安定性の異なるiPS細胞間のカルレティキュリン発現量の差を明確にし得ることが確認された。
これらのことは、ここで開示されるカルレティキュリン発現誘導ペプチド(即ち該ペプチドを有効成分として含むカルレティキュリン発現誘導剤)がゲノム不安定な多能性幹細胞(典型的にはiPS細胞)のカルレティキュリン発現量を顕著に増加させ得るペプチド(組成物)であることを示している。さらに、ここで開示されるカルレティキュリン発現誘導ペプチド(即ち該ペプチドを有効成分として含むカルレティキュリン発現誘導剤)を対象の多能性幹細胞(典型的にはiPS細胞)に添加することで、多能性幹細胞のゲノム安定性の違いに起因するカルレティキュリン発現量の違いを増強し、より明確とし得ることも示している。以上より、ここで開示されるカルレティキュリン発現誘導ペプチド(即ち該ペプチドを有効成分として含むカルレティキュリン発現誘導剤)を対象の多能性幹細胞(典型的にはiPS細胞)に添加することで、該幹細胞におけるカルレティキュリン発現量の程度に基づいて該幹細胞のゲノム安定性若しくはゲノム不安定性を高感度かつ高精度に判別できることが示された。
上記iPS細胞(201B7及び201B2)を供試細胞として用いて、該iPS細胞の培養物中からゲノム不安定なiPS細胞を除去し得るか試験した。詳細は以下のとおりである。
抗カルレティキュリン抗体として、Phycoerythrin(以下、かかる蛍光色素を「PE」ともいう)で標識した抗体(即ち、蛍光PE標識抗体)である、抗カルレティキュリンモノクローナル[FMC75]−Phycoerythrin抗体(マウス由来、Abcam社製、Cat No. ab83220、Lot No. GR177933-2)を用いた。また、上記抗カルレティキュリンモノクローナル[FMC75]−Phycoerythrin抗体のアイソタイプコントロールとして、蛍光PE標識抗体である、Mouse IgG1(PE)-Isotype control(マウス由来、Abcam社製、Cat No. ab81200、Lot No. GR183088-1)を用いた。かかる抗カルレティキュリン抗体(或いは上記アイソタイプコントロール)をY-27632(10μM)含有インキュベーションバッファーで最終濃度:10μg/mLに調製した抗体希釈液を各試験管に添加し、4℃の暗所に1.5時間静置した。かかる所定時間の抗原抗体反応を行った後、130×gで5分間の遠心処理を行い、抗体希釈液を除去し、各iPS細胞を1mLのインキュベーションバッファーで2回洗浄(遠心処理条件:130×g、5分間)した。次いで、各iPS細胞を300μLのOn-Chip Sample Buffer(オンチップバイオテクノロジーズ社製)で1回洗浄(遠心処理条件:260×g、5分間)した。そして、各iPS細胞を、On-Chip Sample Buffer中に分散し、細胞密度が少なくとも5×105細胞/100μL以上の細胞分散液を調整した。
上述のとおり、上記の蛍光免疫染色では、細胞の固定および透過処理を行わなかった。即ち、抗カルレティキュリン抗体によって、細胞表面のカルレティキュリンを特異的に蛍光免疫染色した。
また、対照試験区(ネガティブコントロール区)として、蛍光免疫染色を行っていない各iPS細胞(Unstained cells)を、上記Phycoerythrinの蛍光強度測定と同様の条件で分析した。かかるUnstained cellsの分析結果を、上記抗カルレティキュリン抗体を用いて蛍光免疫染色を行った試験区の蛍光強度を示すヒストグラム(図7および図8)に重ねて示す。図7および図8中では、かかる試験区(ネガティブコントロール区)の結果を、「Unstained cells (Negative control)」として示す。
なお、ここでは詳細なデータは示していないが、上記カルレティキュリンの発現量の程度を調べるより前にカルレティキュリン発現誘導ペプチド(即ちペプチドを有効成分として含むカルレティキュリン発現誘導剤)を該iPS細胞培養物中に添加し所定時間培養することで、ゲノム不安定な細胞の細胞表面でのカルレティキュリンの発現を増大することができ、ゲノム不安定なiPS細胞の除去を高精度かつ高い信頼性に実現し得ることを確認した。
上記サンプル1〜4に係る合成ペプチド(カルレティキュリン発現誘導ペプチド)50mgと結晶化セルロース50mg及び乳糖400mgとを混合した後、エタノールと水の混合液1mLを加え混練した。この混練物を常法に従って造粒し、ここで開示されるカルレティキュリン発現誘導ペプチドを主成分とする顆粒剤(顆粒状組成物)を得た。
Claims (19)
- 対象となるヒト又は他の哺乳動物由来の多能性幹細胞のゲノム安定性を評価する方法であって、
前記多能性幹細胞の培養物を準備すること、および
該培養物中の多能性幹細胞についてカルレティキュリンの発現量を調べ、該カルレティキュリンの発現量の程度に基づいて該多能性幹細胞のゲノム安定性若しくは不安定性を判別すること、
を包含し、
ここで、前記判別において、カルレティキュリン発現量が所定レベルを上回った多能性幹細胞をゲノム不安定性であると判別する、評価方法。 - 前記カルレティキュリンの発現量の程度に基づいた多能性幹細胞のゲノム安定性若しくは不安定性の判別は、カルレティキュリン又はその断片と特異的に反応する抗体を用いた免疫学的手法で行う、請求項1に記載の評価方法。
- 前記判別の前に、前記対象の多能性幹細胞培養物中に以下のアミノ酸配列:
CRAKAGDPC(配列番号1);および
CEQKQEIRC(配列番号2);
のうちのいずれか若しくは該アミノ酸配列において1個、又は2個、又は3個のアミノ酸残基が置換、欠失及び/又は付加されて形成された改変アミノ酸配列のいずれかからなるカルレティキュリン発現誘導ペプチド配列を含む合成ペプチドを少なくとも1回供給することをさらに含み、該合成ペプチドを少なくとも一回供給した当該多能性幹細胞培養物を所定時間培養した後に前記判別を行うことを特徴とする、請求項1又は2に記載の評価方法 - 前記改変アミノ酸配列は、配列番号1又は2のアミノ酸配列のN末端及びC末端のシステイン残基(C)を欠失したものである、請求項3に記載の評価方法。
- 前記合成ペプチドは、前記カルレティキュリン発現誘導ペプチド配列のN末端側若しくはC末端側に膜透過性ペプチド配列を有する、請求項3又は4に記載の評価方法。
- 前記合成ペプチドは、前記膜透過性ペプチド配列として、以下のアミノ酸配列:
KKRTLRKNDRKKR(配列番号5);
を有する、請求項5に記載の評価方法。 - 前記合成ペプチドを構成する全アミノ酸残基数が30以下である、請求項3〜6のいずれか一項に記載の評価方法。
- 前記合成ペプチドは、以下のアミノ酸配列:
KKRTLRKNDRKKRGGCRAKAGDPC(配列番号10);
KKRTLRKNDRKKRGGCEQKQEIRC(配列番号11);
KKRTLRKNDRKKRGGRAKAGDP(配列番号12);および
KKRTLRKNDRKKRGGEQKQEIR(配列番号13);
のうちのいずれかを有する、請求項7に記載の評価方法。 - 対象となるヒト又は他の哺乳動物由来の多能性幹細胞を含む培養物中からゲノム不安定な多能性幹細胞を除去する方法であって、
請求項1〜8のいずれか一項に記載の評価方法によって該培養物中の多能性幹細胞のゲノム安定性若しくは不安定性を判別すること、および
該判別方法によりゲノム不安定性であると判別された多能性幹細胞を該培養物中から除去すること、
を包含する、除去方法。 - 前記ゲノム不安定性であると判別された多能性幹細胞を、細胞選別機(セルソーター)を使用して除去する、請求項9に記載の除去方法。
- 前記ゲノム不安定な多能性幹細胞が、染色体異常を有する多能性幹細胞である、請求項9又は10に記載の除去方法。
- ヒト又は他の哺乳動物由来の多能性幹細胞を含む培養物を製造する方法であって、
該多能性幹細胞の培養過程において、請求項9〜11のいずれか一項に記載の除去方法によってゲノム不安定な多能性幹細胞を該多能性幹細胞の培養物中から除去することを包含する、製造方法。 - ゲノム不安定な多能性幹細胞に対してカルレティキュリン発現誘導活性を奏する人為的に合成されたペプチドであって、以下のアミノ酸配列:
CRAKAGDPC(配列番号1);および
CEQKQEIRC(配列番号2);
のうちのいずれか若しくは該アミノ酸配列において1個、又は2個、又は3個のアミノ酸残基が置換、欠失及び/又は付加されて形成されたアミノ酸配列のいずれかからなるカルレティキュリン発現誘導ペプチド配列を含む合成ペプチド。 - 前記改変アミノ酸配列は、配列番号1又は2のアミノ酸配列のN末端及びC末端のシステイン残基(C)を欠失したものである、請求項13に記載の合成ペプチド。
- 前記カルレティキュリン発現誘導ペプチド配列のN末端側若しくはC末端側に膜透過性ペプチド配列を有する、請求項13又は14に記載の合成ペプチド。
- 前記膜透過性ペプチド配列として、以下のアミノ酸配列:
KKRTLRKNDRKKR(配列番号5);
を有する、請求項15に記載の合成ペプチド。 - 前記合成ペプチドを構成する全アミノ酸残基数が30以下である、請求項13〜16のいずれか一項に記載の合成ペプチド。
- 以下のアミノ酸配列:
KKRTLRKNDRKKRGGCRAKAGDPC(配列番号10);
KKRTLRKNDRKKRGGCEQKQEIRC(配列番号11);
KKRTLRKNDRKKRGGRAKAGDP(配列番号12);および
KKRTLRKNDRKKRGGEQKQEIR(配列番号13);
のうちのいずれかを有する、請求項17に記載の合成ペプチド。 - ゲノム不安定な多能性幹細胞のカルレティキュリン発現量を増大するために用いられるカルレティキュリン発現誘導剤であって、請求項13〜18のいずれか一項に記載の合成ペプチドを含む、カルレティキュリン発現誘導剤。
