Abstract
Description
Adhesion Molecule)とは、細胞と細胞間または細胞と細胞骨格の接着を司る分子である。そして、その歴史は1976年にEdelmanらが鶏の神経細胞接着分子(Neural
Cell Adhesion Molecule)を研究したことに始まる。
つまり上記のように以前作成した抗CD166抗体は、特異的な吸着の度合いが十分でないことが考えられた。

鶏CD166(別名SC1、BEN,ALCAM)蛋白質のN末端から第4番目のイムノグロブリン様ループの一部を構成する、下記表1に記載する配列番号1および2のアミノ酸の合成ペプチドを抗原とし、オス6週齢のオスBALB/cマウスに免疫した。
実施例1では、本願発明の抗体AおよびBが、in vitroでCD166に特異的に吸着することを確認した。そこで次にin vivoでの特異的な吸着を確認する。
次に本発明の抗体AおよびBの増殖抑制作用を、確認した。ヒト肺癌細胞(A549細胞)106個をヌードマウスの頚部皮下に移植し、5、10、14日目に正常マウスIgG、上記2種の抗CD166モノクローナル抗体(抗体Aおよび抗体B)をそれぞれ100μgを腹腔内に接種した。
本発明の抗体AおよびBは、ELISA(Enzyme Linked Immuno Sorbent Assay)法にも応用できる。
次に実施例4のELISA法で血液検査によって癌の進行度の測定ができる点を説明する。CD166は上述のように、癌細胞の表面に多く発現する分子であるが、癌の進行によって血液中のCD166の含有量が増加してくる。これをELISA法で確認することで、癌の進行度合いを相対的に判断することができる。
本実施例では、ヒトの血液中のCD166を測定することで、癌の進行度を測定した。
Claims (8)
- 細胞接着分子CD166/ALCAMに対し特異的に結合するモノクローナル抗体。
- 配列番号1に示すアミノ酸配列の少なくとも1部に特異的に結合する請求項1記載のモノクローナル抗体。
- 配列番号2に示すアミノ酸配列の少なくとも1部に特異的に結合する請求項1記載のモノクローナル抗体。
- サブクラスが免疫グロブリンGであることを特徴とする請求項1乃至3のいずれかの1に記載のモノクローナル抗体。
- 請求項1乃至3のいずれかの1に記載のモノクローナル抗体を産生するハイブリドーマ。
- キジ科動物のCD166蛋白質で免疫したマウスから得た抗体をモノクローナル化するモノクローナル抗体の製造方法。
- 請求項1乃至3のモノクローナル抗体を用いたCD166の含有量の測定方法。
- 血液中のCD166の含有量から癌の進行状態を推定する癌診断方法。
