(13)上記(9)から上記(12)のいずれかに記載の抗原タンパク質のポリユビキチン化促進剤を有効成分として含む、樹状細胞のCTL誘導能力増強剤。
なお、本明細書において引用された全ての先行技術文献は、参照として本明細書に組み入れられる。
外来抗原の細胞内局在を細胞分画法により検討した。抗原モデルとしてニワトリアルブミンOvalbumin (OVA) 、樹状細胞のモデルとして、マウス樹状細胞の培養細胞株であるDC2.4(ハプロタイプH-2Kb)を用いた。DC2.4細胞株は、樹状細胞由来であり、クロスプレゼンテーション等の樹状細胞の多くの性質を保ったまま株化されたものである。またコントロールとして、ハプロタイプがDC2.4と同じH-2KbであるBMDCを用いた。BMDCは、C57BL/6マウス(SLC)の骨髄細胞から5%FCSを添加したRPMI-1640中で5ng/ml GM-CSF (MBL)を用いて誘導し、培養5日後に細胞を磁性ビーズで精製したものを用いた。
上記仮説を立証するため、まず、蓄積されたOVAとERから細胞質への逆行性輸送関連物質との関連について共免疫沈澱法により検討した。2x107個のDC2.4またはBMDC細胞を10μM MG-132を含む培地中でbOVA 250μg ml-1存在下で4時間培養した。細胞を1%ジギトニンとプロテアーゼ阻害剤を含む20mM HEPES pH7.6で溶解し、ビオチン標識サンプルを回収した。サンプルはプロテインGセファロース(Amersham Pharmacia Biotech)であらかじめ不要物を除去し、沈降のための抗体(抗Sec61β、抗Sec61α、抗VCP、抗BiP、抗PDI、抗calreticulin、抗TAP1、抗TAP2)とインキュベートした後、プロテインGで免疫沈澱物を回収した。ニワトリ抗CHIP抗体を使った場合は、ウサギ抗IgYカラム(Mr.S.Seki,MBL)で免疫沈澱物を回収した。沈降サンプルは、SDS-PAGEおよびウエスタンブロッティングで解析した。
次に、ERADの関与をさらに確かめるため、ERAD阻害、例えば、Ca2+枯渇やthapsigargin処理によってbOVAの蓄積が影響を受けるか否かを検討した。ER内のCa2+を枯渇させるとERADが阻害されることが知られている。また、Thapsigargin(Tg)はER膜に結合しイオノフォアとして機能することで、ER内腔のCa2+濃度を低下させ、ERADを阻害することが知られており、Tg存在下ではbOVAのERADによる分解は抑制されると予想される。DC2.4細胞をMG-132存在下でbOVAとパルスし、続いてPBSまたは1mM EGTAを含むPBSで洗浄し、4時間にわたり追跡した。図4aに示すように、蓄積したbOVAはCa2+枯渇では減少しなかった。あらかじめEGTAに曝した細胞にCa2+を再添加すると、bOVA 分解が回復した(図4a)。このことは、Ca2+枯渇がDC2.4細胞に不可逆的ダメージを引き起こした可能性を排除する。thapsigarginもまた蓄積したbOVAの分解を阻害した(図4b)。

上記結果の全てが、DC2.4細胞に取り込まれたbOVAがERADを通じて分解されていることを強く支持している。しかしながら、最近になって、プロテオミクス解析や免疫電子顕微鏡観察によって、感染性バクテリアが取り込まれる場所であるファゴソームが、MHC Iといった抗原提示関連物質やTAP1/2等のERを構成する一群のタンパク質を含むことが明らかになった。このことから本発明者らは、DC2.4細胞やBMDC中の蓄積OVAの細胞内局在化について免疫蛍光コンフォーカル顕微鏡によって検討することとした。カルチャースライド(FALCON)上のDC2.4細胞とBMDCをMG-132で30分間前処理し、500μg ml-1OVAで2時間インキュベートした。培地で3回洗浄した後、細胞を3.7%ホルムアルデヒドで10分間固定し、0.1%TritonX-100 in PBSで室温10分間処理して透過処理し、5%BSA/2%FCSを含むPBS中で所定の抗体により37℃で1時間インキュベートし、染色した。二次抗体は、Alexa Fluor488ヤギ抗マウスIgG F(ab’)2フラグメント、Alexa Fluor488ヤギ抗ラットIgGおよびAlexa Fluor 546ヤギ抗ウサギF(ab’)2フラグメント(Molecular Probes)を使用した。画像は、蛍光装置のついたIX70(オリンパス)倒立顕微鏡を用いて取り込み、Deltavision deconvolution microscopy software(Applied Precision)によって処理して取得した。
次に本発明者らは、イオジキサノール勾配遠心によってDC2.4細胞の膜画分中の蓄積OVAの分布を検討した。密度勾配遠心のために、bOVA(500μg/ml)と3時間インキュベートしたDC2.4細胞を、PBSで2回洗浄し、homogenization medium(0.25Mスクロース、1mM EDTA、10mM Hepes-NAOH pH7.4)に懸濁し、Dounce homogenizerの10strokesで破壊した。未破壊細胞と核は、2,000xgで10分間遠心して除去した。核を除去した(postnuclear)上清を100,000xgで45分間遠心してペレットとし、homogenization mediumに再懸濁し、2.5-30または10-30%の不連続Optiprep(Gibco/Invitrogen)グラジエントに重層した。遠心は、Bechman SW 60Tiローターを用い、4℃で2.5-30%グラジエントの場合は200,000xgで2.5時間、10-30%グラジエントの場合は300,000xgで3時間行った。各遠心チューブの上層から取得した11画分をTCA沈殿させた後、SDS-PAGEおよびウエスタンブロッティングで解析した。
ERADに関与する細胞構成物質のノックダウンが外来性OVA抗原のクロスプレゼンテーションを低下させるか否かについて、DC2.4細胞とOT-IマウスのナイーブCD8+T細胞とを用いた抗原提示レベルの評価系によって検討した。樹状細胞のモデルとして用いたDC2.4細胞は、マウスMHCクラスIハプロタイプがH-2Kbである。OT-I(H-2Kb)マウスは、H-2Kb分子上に提示されたOVA由来の抗原ペプチド(SIINFEKL)(配列番号15)を認識するT細胞受容体(TCRα,β)を発現するトランスジェニックマウスである。OT-IマウスのCD8+細胞は、DC2.4細胞またはH-2Kbマウス由来のBMDCが提示するOVAペプチドを認識し、活性化される。DC2.4細胞をOVAまたはbOVAでインキュベートして固定した後、さらにOT-IマウスのナイーブCD8+T細胞とインキュベートしたときに、OT-I CD8+T細胞によって産生されるIL-2量は、DC2.4細胞によって提示されたOVA抗原によるT細胞刺激の指標となる。
ER内腔に局在するタンパク質のカルボキシル末端に小胞体局在配列であるKDEL配列(Lys-Asp-Glu-Leu)(配列番号3)を付加すると、ERとゴルジ体間を繰り返し輸送され、そのタンパク質のERADが促進されることが報告されている。そこで、OVAのC末端にKDEL(配列番号3)を付加したものをDC2.4細胞に取り込ませ、SDS-PAGEおよびウェスタンブロットにより、取り込み後にポリユビキチン化されたOVAを観察した。KDEL配列(配列番号3)付加OVAは、下記のようにして構築および発現させた。OVAをコードするDNA配列を含むプラスミドを鋳型とし,プライマーとしてoligo-1(配列番号10)とoligo2(配列番号11)を用いてPCR法を行なった。増幅した約1100 bpのPCR産物をpCR-Blunt vector(Invitrogen)に導入し、vector-1とした。oligo-3(配列番号12)とoligo-4(配列番号13)をアニールし、得られた約100 bpの遺伝子断片をpCR-Blunt vectorに導入し、vector-2とした。vector-2をSpe Iで消化し、得られた約130 bpの遺伝子断片を、Nhe Iで消化したvector-1に導入し、vector-3とした。vector-3をBamH I/Spe Iで消化したときに得られる約1250 bpの遺伝子断片(OVA-KDEL)を、遺伝子発現用ベクターに導入し、発現させた。KDEL配列(配列番号3)付加OVAアミノ酸配列を配列番号14に示す。
ブタ由来HSP90(10μg/ml)存在下または非存在下で、DC2.4細胞とOVA(0,50,250μg/ml)をRPMI1640/10%FCSで37℃6時間インキュベートした。抗原とHSPを洗浄後、OT-I CD8+T細胞を加えて37℃で一晩培養し、上清に含まれるIL-2量をELISA法で測定した。その結果、OVA単独とインキュベートした場合よりも、より強い抗原提示が認められた(図11-(a))。また、Hsp90濃度依存的に抗原提示能力が高くなった(図11-(b))。
マウス骨髄由来樹状細胞(BMDC)を次のとおり調製した。マウス骨髄から細胞を取り出し、GM-CSF,5%FCSを含むRPMI培地を用い、途中培地を一部交換しながら5日間培養した。この培養法によりマクロファージ、樹状細胞などが増えてくるので、CD11c磁気ビーズを用いて樹状細胞のみを回収し、これを骨髄由来樹状細胞(BMDC)として用いた。
上記調製した骨髄由来樹状細胞(BMDC)またはマウス樹状細胞株DC2.4細胞を96ウェル・プレートの1ウェルあたり5×104個ずつまいた。次に、エンドトキシン阻害剤であるポリミキシンB 50μg/ml存在下に、OVA(0,50,250μg/ml)、HSP90(0,10μg/ml)を添加して総液量を200μlとし、37℃で一晩培養した。エンドトキシンはDCを活性化することが知られているので、ポリミキシンBを添加し、試薬、器具などに夾雑した場合の影響を排除した。培養後、上記ウェル中の細胞を2回洗浄し、細胞に取り込まれずに残ったOVAとHSP90を除去した。そこにOT-IIトランスジェニック・マウスより調製したCD4+T細胞を1ウェルあたり5×104個加えて総液量を200μlとし、一晩培養した。ウェルを遠心して上清を回収し、上清中のIL-2量をELISA法により測定した。
OT-Iトランスジェニックマウスは、ovalbumin由来のペプチドがMHCクラスIに提示された状態を認識するT細胞レセプターを持つトランスジェニックマウスである。細胞上のペプチド-MHCクラスIとCD8+T細胞上のT細胞レセプターが反応することにより刺激が入り、T細胞がIL-2を産生する。一方、OT-IIトランスジェニックマウスは、ovalbumin由来のペプチドがMHCクラスIIに提示された状態を認識できるT細胞レセプター持つトランスジェニックマウスである。抗原提示細胞上のペプチド-MHCクラスIIとCD4+T細胞上のT細胞レセプターが反応することにより刺激が入り、T細胞がIL-2を産生する。OT-Iマウス由来のCD8+T細胞を用いた場合の結果(図11-(a))及びOT-II由来のCD4+T細胞を用いた場合の結果(図13)から、分子シャペロンHSP90は、樹状細胞(DC)による外部抗原OVAのMHCクラスI分子によるCD8+T細胞への提示を増強するが、MHCクラスII分子によるCD4+T細胞への提示は増強しないことが明らかである。つまり、HSP90はDCによる細胞傷害性T細胞(CTL)の活性化を増強するが、ヘルパーT細胞の活性化は増強しないことがわかった。
DC2.4細胞にHSP90(10μg/ml)とOVA(250μg/ml)を加えるタイミングを変えて、そのクラスI抗原提示に対する効果を見た。(i)HSP90を先に加え、DC2.4細胞を洗った後、OVAを添加した。(ii)HSP90とOVAをDC2.4細胞に同時に添加した。(iii)OVAを先に添加し、DC2.4細胞を洗ってからHSP90を添加した。これらの3条件の下で行った結果を比較した。また、全ての条件に対するコントロールとして、HSP90を添加せずにOVAのみを加えた実験も行った。
条件(i)では、DC2.4細胞にHSP90を添加して30分培養後、細胞を洗い、HSP90を除いた。次いで、OVAを加えて3時間培養した。その後、OVAを洗って除き、細胞培養液のみで3時間培養した。上記操作によりOVAで活性化されたDC2.4細胞を96穴プレートにまき、そこにOT-Iマウス由来のCD8+T細胞を加えて一晩培養後、上清に放出されたIL-2量をELISA法で測定した。
条件(ii)では、DC2.4細胞にOVAとHSP90を同時に添加して3時間培養後、細胞を洗って細胞培養液のみで3時間培養した。以後、OT-Iマウス由来のCD8+T細胞の活性化を調べる操作は、上記(i)と同様に行った。
条件(iii)DC2.4細胞にまずOVAを加えて3時間培養後、細胞を洗い、OVAを除いた。次いで、HSP90を添加してさらに3時間培養した。以後は(i)と(ii)に同様に行った。なお、条件(i)〜(iii)において、細胞がOVAと接触している段階とOVAを取り込んだ以降の段階との培養時間が同じになるようにタイミングを設定した。
HSP90のDC2.4細胞のOVAのクラスI抗原提示に対する効果は、HSP90がOVAと共存しないと発揮されないことが明らかとなった。また、一度取り込まれたOVAに対してもHSP90は効果を及ぼさないことも示された(図14カラム(O→H))。HSP90は抗原OVAの取り込みあるいは、そのプロセッシングを増強するということを予想させる。また、この結果は、HSP90がOVAの取り込みをいく分増強すること、および取り込まれたOVAのポリユビキチン化を顕著に増強するという結果(図10)とよく一致している。
Claims (13)
- 抗原タンパク質の樹状細胞内ポリユビキチン化を促進させるポリユビキチン化促進処理工程を含む、CTL誘導能力を獲得した樹状細胞の製造方法。
- 下記(a)および(b)の工程を含む、CTL誘導能力を獲得した樹状細胞の製造方法。
(a)哺乳動物由来の樹状細胞を単離する工程
(b)抗原タンパク質の樹状細胞内ポリユビキチン化を促進させるポリユビキチン化促進処理工程 - 下記(a)から(c)の工程を含む、CTL誘導能力を獲得した樹状細胞の製造方法。
(a)哺乳動物由来の樹状細胞を単離する工程
(b)抗原タンパク質の樹状細胞内ポリユビキチン化を促進させるポリユビキチン化促進処理工程
(c)樹状細胞内における抗原タンパク質のポリユビキチン化の促進を確認する工程 - ポリユビキチン化促進処理工程が単離された樹状細胞に抗原タンパク質とHSPとを接触させることを特徴とする工程である、請求項1から3のいずれかに記載のCTL誘導能力を獲得した樹状細胞の製造方法。
