哺乳動物系における表現型ベースの遺伝スクリーニングの主要な制限は、ゲノムの二倍体性質である。ブルーム症候群遺伝子(Bml)の喪失は、ヘテロ接合の喪失(LOH)の増大した割合を示す(German,J.、Dermatol.Clin.13:7〜18(1995)、Groden,J.,Nakamura,Y.およびGerman,J.、Proc.Natl.Acad.Sci.USA 87:4315〜4319(1990)およびLuo,G.ら、Nature Genet.26:424〜429(2000)を参照のこと)。

上記のような改変を設計する際に、アミノ酸の疎水性指数が考慮され得る。タンパク質における相互作用的な生物学的機能を与える際の疎水性アミノ酸指数の重要性は、一般に当該分野で認められている(Kyte.JおよびDoolittle,R.F.J.Mol.Biol.157(1):105−132,1982)。アミノ酸の疎水的性質は、生成したタンパク質の二次構造に寄与し、次いでそのタンパク質と他の分子(例えば、酵素、基質、レセプター、DNA、抗体、抗原など)との相互作用を規定する。各アミノ酸は、それらの疎水性および電荷の性質に基づく疎水性指数を割り当てられる。それらは:イソロイシン(+4.5);バリン(+4.2);ロイシン(+3.8);フェニルアラニン(+2.8);システイン/シスチン(+2.5);メチオニン(+1.9);アラニン(+1.8);グリシン(−0.4);スレオニン(−0.7);セリン(−0.8);トリプトファン(−0.9);チロシン(−1.3);プロリン(−1.6);ヒスチジン(−3.2);グルタミン酸(−3.5);グルタミン(−3.5);アスパラギン酸(−3.5);アスパラギン(−3.5);リジン(−3.9);およびアルギニン(−4.5))である。
本明細書において「スクリーニング」とは、目的とするある特定の性質をもつ生物または物質(例えば、遺伝子)などの標的を、特定の操作/評価方法で多数を含む集団の中から選抜することをいう。スクリーニングのために、本発明の細胞を使用することができる。
(条件的なBlm ES細胞の作製方法)
マウスBlm遺伝子(配列番号1および2)を含むゲノムDNAをR1−ESゲノムライブラリーから単離した。ターゲティングベクターをR1−ES細胞に導入し、続けてG418選択および/またはピューロマイシン選択(シグマ)を行った。標的化したクローンをPCRによってスクリーニング(Expand High Fidelity PCR System(Roche))し、そしてサザンブロット解析(Rapid−hyb buffer(アマシャムファルマシア))で確認した。本研究で使用した標的化したクローンは全て、正常な核型を保有した。
Blmtet/tetES細胞を1.0μg/mlのドキシサイクリン(dox)(シグマ)を含む培地中で培養し、適切な時点で収集した。dox中止後のBlm発現を調べるために、doxを含む培地中で培養した細胞をPBSで1回洗浄し、収集するまで、さらにdoxの不在下で細胞を培養した。Blmタンパク質を抗BLM抗体(ab476、abcam)で検出した。ウェスタンブロットは、ECL Western Blotting Detection Reagents(アマシャムファルマシア)を用いた。
適切な期間、doxの存在下または不在下で培養したBlmtet/tetES細胞を、3μg/mlの5-ブロモデオキシウリジン(BrdU)(シグマ)で20時間標識し、0.1μg/mlのコルセミド(KaryoMax−Colcemid(インビトロジェン))で45分間処理した。広がった染色体を0.1mg/mlのアクリジンオレンジ(シグマ)で染色した。
変異neo遺伝子(Koikeら、EMBO Rep.3、433〜437(2002)を参照のこと)(配列番号4および5)を有するFasl遺伝子座に対するターゲティングベクターを、Blmtet/tetES細胞に導入した。二多型の変異誘発率を、前述(Koike,H.ら、EMBO Rep.3:433〜437(2002)を参照のこと)のLuria−Delbruck機能解析を用いて測定した。詳細には、Blmtet/tetES細胞を1.0μg/mlのdoxの存在下または不在下で24時間培養し、doxを伴ってかまたは伴わない純系濃度の培養条件で100mm培養皿に撒いて、単一細胞クローンを得た。次いで、doxを伴わない高用量G418(1.0mg/ml)の補助により、広がった単一細胞クローンを選択した。選択の10日後に、高用量G418耐性クローンの数を数えた。
ENU変異誘発およびHprt遺伝子座の変異頻度の算出は、Chen,Y.ら、Nature Genet.24,314〜317(2000)に記載された方法で行った。変異誘発されたES細胞を、doxの存在下または不在下で4日間培養し、変異ES細胞ライブラリーを作製した。この変異ライブラリーおよびコントロールライブラリーを、1mlあたり1.0×106細胞の懸濁液中、10nMのプロアエロリジン(Protox Biotech)で処理し、ゼラチンコーティングした100mm培養皿に、培養皿1枚あたり5〜8×106細胞で撒いた。次の日、死細胞を洗い出し、生細胞をさらに5nMのプロアエロリジンで8時間処理し、マイトマイシンC(協和発酵(株))で処理したフィーダー細胞を添加し、GPIアンカー欠損変異体コロニーを作製した。結果として得られたコロニーを収集して拡大させ、相補性アッセイのために、GFP−GPI発現ベクターおよびGPIアンカーの生合成に関するcDNAを用いてトランスフェクトした(TransFast transfection reagent(プロメガ)、DNA 2mg、TransFast 12μlを1.0×105個のES細胞に加える)。
図1aは、テトラサイクリンで条件的に調節されるBlm対立遺伝子(Blmtet)の概略図(配列番号3)である。テトラサイクリン系ベースの調節カセット(tetカセット)(Bond,C.T.ら、Science 289:1942〜1946(2000)を参照のこと)を、Blmの両対立遺伝子の翻訳開始コドンの直上流に挿入し、これらをBlmtetと交換した。ターゲティングは、サザンブロット解析で確認した(図1b)。本発明者らは、Blmの持続的な欠損は、ゲノム上での二対立遺伝子変異の持続的な集積を引き起こし、結果として長期の培養の間に表現型を変化させると仮定した。その一方、BlmtetによってもたらされるBlmの一過性の喪失は、表現型における変化を最小限にする。Blm発現の調節は、テトラサイクリンアナログであるドキシサイクリン(dox)を使用することによって調べた。doxの添加は、結果としてBlmタンパク質の急速な減少をもたらした(図1cおよび1d)。より重要には、Blmタンパク質は、doxの中止後にその本来のレベルを回復した(図1c)。ブルーム症候群細胞(German,J.、Dermatol.Clin.13:7〜18(1995)を参照のこと)の代表的な細胞遺伝現象である、姉妹染色分体交換(SCE)の高いレベルが観察された一方で(図1e)、Blmタンパク質は検出されなかった(図1c)。SCEは、脊椎動物細胞において相同組換えと密接に関連する(Sonoda,E.ら、Mol.Cell.Biol.19;5166〜5169(1999)を参照のこと)。それゆえ、dox処理は、相同染色体間の組換えを誘導すると予想され得、組換えが4N期に起こる場合、一対立遺伝子変異が細胞分裂の後に二対立遺伝子変異となる(図2a)。二対立遺伝子変異の割合における一過性のBlm欠損の影響を調べるために、「一対立遺伝子変異」モデルとしての遺伝子ターゲティングによって、変異neo遺伝子をFasリガンド(Fasl)遺伝子座に導入した。本発明者らは以前に、「二対立遺伝子変異」を意味する、この遺伝子座における変異neo遺伝子の重複が、高用量G418を使用して選択され得ることを証明した(Koike,H.ら、EMBO Rep.3:433〜437(2002)を参照のこと)。Luria−Delbruck周期的変動解析(Luria,S.E.およびDelbruck,M.、Genetics 28:491〜510(1943)を参照のこと)によって決定された重複率は、doxの不在下で1世代あたりの1細胞あたり8.5×10−6事象であるのに対し、この比率は、doxの存在下で1世代あたりの1細胞あたり2.3×10−4事象に増加した(図2b)。それゆえ、Blmの一過性の喪失は、二対立遺伝子変異の比率に27倍の増加をもたらした(図2b)。本研究で得られたこの比率は、野生型ES細胞(2.3×10−5)およびBlm欠損ES細胞(4.2×10−4)におけるLOHについて以前に報告されたもの(Luo,G.ら、Nature Genet.26:424〜429(2000)を参照のこと)と類似する。

Claims (29)
- 対立遺伝子の両方の鎖に変異が挿入された、幹細胞。
- 前記幹細胞は、胚性幹細胞である、請求項1に記載の幹細胞。
- 前記幹細胞は、ブルーム症候群(Blm)遺伝子が喪失されているかまたは機能しないように改変されている、請求項1に記載の幹細胞。
- 前記ブルーム症候群遺伝子は、配列番号1に記載される配列またはその改変体を含む、請求項3に記載の幹細胞。
- 対立遺伝子の両方の鎖に変異が挿入された、幹細胞のライブラリーであって、該ライブラリーに含まれる幹細胞は、ゲノム全体にわたって該変異が導入されている、ライブラリー。
- 前記幹細胞は、胚性幹細胞である、請求項5に記載のライブラリー。
- 前記幹細胞は、ブルーム症候群遺伝子が喪失されているかまたは機能しないように改変されている、請求項5に記載のライブラリー。
- 前記ブルーム症候群遺伝子は、配列番号1に記載される配列またはその改変体を含む、請求項7に記載のライブラリー。
- 対立遺伝子の両方の鎖に変異が導入された幹細胞を生産する方法であって、
A)幹細胞を提供する工程;
B)該幹細胞におけるブルーム症候群遺伝子が機能しないようにする工程;および
C)該幹細胞における変異を誘発する工程、
を包含する、方法。 - 前記ブルーム症候群遺伝子は、一過的に機能しないように処理される、請求項9に記載の方法。
- 前記ブルーム症候群遺伝子は、薬剤の存在下で、一過的に機能しないように処理されている、請求項10に記載の方法。
- 前記薬剤は、テトラサイクリン、ドキシサイクリン、エストロジェン誘導体およびプロジェストロン誘導体からなる群より選択される、請求項11に記載の方法。
- 前記変異の誘発は、変異原への暴露、トランスポゾン遺伝子の利用、紫外線への暴露および放射線への曝露からなる群より選択される、請求項9に記載の方法。
- 相同組換えを誘発する工程をさらに包含する、請求項9に記載の方法。
- 相同組換えを前記細胞の4N期に誘発させ、該誘発後に細胞分裂をさせる工程をさらに包含する、請求項14に記載の方法。
- 前記幹細胞は、胚性幹細胞である、請求項9に記載の方法。
- 前記胚性幹細胞は、哺乳動物の胚性幹細胞である、請求項9に記載の方法。
- 請求項9に記載の方法によって得られる幹細胞。
- 胚性幹細胞である、請求項18に記載の幹細胞。
- 請求項9に記載の方法によって得られる幹細胞から得られる、組織。

- 前記Blm遺伝子は、前記幹細胞において喪失されるかまたは機能しないように改変される、請求項27に記載の使用。
- 前記ブルーム症候群遺伝子は、配列番号1に記載される配列またはその改変体を含む、請求項27に記載の使用。
