Description
【発明の詳細な説明】
本発明は新規2−シクロプロピルアミノ−4−
ホルミルアミノ−6−アミノ−s−トリアジン−
誘導体とその塩、及びその製造方法並びに該化合
物を含有する有害生物防除剤に関するものであ
る。
2−シクロプロピルアミノ−4−ホルミルアミ
ノ−6−アミノ−s−トリアジン−誘導体は次式
:

R1は水素原子、未置換もしくは低級アルコキ
シ基で置換された炭素原子数1ないし4のアルキ
ル基、炭素原子数3ないし4のアルケニル基、炭
素原子数3ないし4のアルキニル基、シクロプロ
ピル基を表わし、
R2は水素原子、メチル基もしくはエチル基を
表わすか、
R1及びR2はこれらが結合している窒素原子と
一緒になつて、モルホリニル基、ピロリジニル基
及びイミダゾリル基を表わし、
R3は水素原子を表わす。)
で表わされる構造及びその酸付加塩の構造を有す
る。
R1のアルキル基に関しては、直鎖もしくは枝
分れ鎖の未置換もしくは、例えば炭素原子数1な
いし4のアルコキシ基等で置換された基であつて
もよい。そのようなアルキル基の例としては、メ
チル基、エチル基、n−プロピル基、イソプロピ
ル基、n−ブチル基、イソブチル基、第2及び第
3ブチル基並びに2−メトキシエチル基が挙げら
れる。
炭素原子数3ないし4のアルケニル基と炭素原
子数3ないし4のアルキニル基は各々アリル基及
びプロパギル基から成る。
前記式で表わされる2−シクロプロピルアミ
ノ−4−ホルミルアミノ−6−アミノ−s−トリ
アジン−誘導体の「酸付加塩」という概念は、強
い鉱酸による塩たとえば塩酸もしくは硫酸による
塩を意味すると理解されるべきである。
前記式で表わされる化合物のうち、防除作用
の点で好ましい化合物は、式中R1が水素原子、
未置換もしくは低級アルコキシ基で置換された炭
素原子数1ないし4のアルキル基、炭素原子数3
ないし4のアルケニル基、炭素原子数3ないし4
のアルキニル基、シクロプロピル基を表わし、
R2が水素原子、メチル基もしくはエチル基を表
わすか、R1とR2がこれらが結合している窒素原
子と一緒になつて、1−ピロリジニル基、4−モ
ルホリニル基もしくは1−イミダゾリル基を表わ
し、R3が水素原子を表わす化合物である。
そのうち特に有利な化合物は、前記式中R1
が水素原子、メチル基、エチル基、イソプロピル
基、2−メトキシエチル基、アリル基、プロパル
ギル基もしくはシクロプロピル基を表わし、R2
が水素原子もしくはメチル基を表わすか、R1と
R2がこれらが結合している窒素原子と一緒にな
つて1−ピロリジニル基、4−モルホリニル基、
もしくは1−イミダゾリル基を表わし、R3が水
素原子を表わすものである。
さらに最も好ましい化合物は、前記式中R1
が水素原子、エチル基、アリル基、もしくはプロ
パルギル基を表わし、R2が水素原子を表わし、
R3が水素原子を表わすものであり、特に2−シ
クロプロピルアミノ−4−ホルミルアミノ−6−
エチルアミノ−s−トリアジン、
2−シクロプロピルアミノ−4−ホルミルアミ
ノ−6−アリルアミノ−s−トリアジン、
2−シクロプロピルアミノ−4−ホルミルアミ
ノ−6−プロパルギルアミノ−s−トリアジン、
及び
2−シクロプロピルアミノ−4−ホルミルアミ
ノ−6−アミノ−s−トリアジン
の4化合物である。
前記式で表わされる化合物は、次の方法で製
造され、それらはたとえば
(a) 次式:

R1、R2及びR3は前記式で表わした意味を
有し、R4及びR5は同一もしくは異なつて、メ
チル基もしくはエチル基を表わす。)
で表わされる2−シクロプロピルアミノ−4−
ホルムアミジノ−6−アミノ−s−トリアジン
を、加水分解によつて相当する2−シクロプロ
ピルアミノ−4−ホルミルアミノ−6−アミノ
−s−トリアジンとする方法、または
(b) 次式:
(式中
R3は前記式で表わした意味を表わし、R1
及びR2は前記式で表わした意味のうち、水
素原子を除いたものの意味を有する。)
で表わされる2−シクロプロピルアミノ−4・
6−ジアミノ−s−トリアジンを、ホルミル化
剤と反応させる方法である。
(a)の加水分解は塩酸のような酸を添加して0゜な
いし100℃で常圧下に実施することもできる。有
利には、溶媒もしくは希釈剤として水を用い、も
し望むならば、ジオキサン、アルコール等の有機
溶媒を添加する。そうすることによつて反応生成
物が沈殿する。
(b)の反応は、好ましくはホルミル化試薬として
蟻酸及び無水酢酸を使用するか、もしくは触媒量
の強酸の存在下で、前記式で表わされる化合物
とオルト蟻酸エステルとを縮合させ、その後部分
的に加水分解することによつて行う。前者の場合
には、反応体に対して不活性な溶媒もしくは希釈
剤を用いることができ、後者の場合には、溶媒と
してそのオルトエステルを用いる。
前記式で表わされる出発原料は、次式:
(式中R1、R2及びR3は前記式で表わした意味
を有する)
で表わされるトリアジン誘導体を次式:
(式中、R4及びR5は、前記式中で表わした意
味を有する。)
で表わされるアセタールと反応させることによつ
て得られる。
前記式で表わされる出発原料は、塩素を1個
もしくは数個の相当するアミノ基で置き換えるこ
とにより、塩素−置換s−トリアジン誘導体か
ら、通常の方法によつて製造できる。
ジアミノ−s−トリアジン及びトリアミノ−s
−トリアジンは、合衆国特許第3189521号明細書
中に、イエバエ(Musca domestica)の成虫に
対する化学不妊剤として記述されている。2・
4・6−トリアミノ−s−トリアジン誘導体(メ
ラミン誘導体)の昆虫に対する化学不妊作用もま
たエス・ナガサワ等による防虫科学39(4)、105
(1974)に記述されている。また、アー・ベー・
ボルコベツク(A・B・Borkovec)及びアー・
ベー・デミロ(A・B・DeMilo)〔J.Med.Chem.
10(5)、457(1967)参照〕及びゲー・ツエー・ラブ
レツケ(G.C.LaBrecque)、エル・エル・フエ
(R.L.Fye)、アー・ベー・デミロ及びアー・ベ
ー・ボルコベツク、〔J.Econ.Entomol.61、(6)、
1621(1968)参照〕は、他の化合物の中で2−シ
クロヘキシルアミノ−4・6−ジアミノ−s−ト
リアジン、2−シクロヘキシルアミノ−4・6−
ジヘキシルアミノ−s−トリアジン及び2・4・
6−トリス−シクロヘキシルアミノ−s−トリア
ジン及びその塩類のイエバエ(Musca
domestica)の生虫に対する化学不妊作用を述べ
ている。アセチルメラミンの化学不妊作用は、合
衆国特許第3520974号明細書中に記述されている。
本発明においては、前記式で表わされる化合
物が、昆虫の幼虫好ましくは双翅目(Diptera)
の幼虫に対し、顕著な殺幼虫作用を示すことが見
い出された。上記の殺虫性化学不妊剤に比べて、
前記式で表わされる化合物は、昆虫の発育段階
に対して作用する。上記作用は、新しくかえつた
幼虫を殺すかもしくは、さなぎからかえつて成虫
になるのを妨害する作用から成る。前記式で表
わされる化合物の作用の型は、今までの殺虫剤、
化学不妊剤もしくは類似した幼虫ホルモンと比較
されるべきではない。
前記式で表わされる有効成分は、特に双翅目
すなわちカ科(culicidae)、シムリイダエ
(Simuliidae)、ガガンボ科(Tipulidae)、イエバ
エ科(Muscidae)、及びクロバエ科
(Calliphoridae)の、衛生上の有害生物、及び動
物に付く体外寄生虫の防除に使用できる。
前記式で表わされる化合物は、純粋な有効成
分として、もしくは適当な担体及び/または添加
剤と共に使用できる。適当な担体及び添加剤は、
固体もしくは液体でもよく、それは配合の分野で
慣例的に使用される物質に相当し、たとえば天然
もしくは再生物質である溶媒及び/または分散剤
である。
本発明による組成物は、前記式で表わされる
有効成分(化合物)を適当な担体と、場合によつ
ては有効成分に対して不活性な分散剤もしくは溶
媒を加えて均一に混合及び/または磨砕すること
による公知の方法で製造される。
有効成分は下記のような剤形にして使用するこ
とができる。
固体製剤:
粉剤、散布剤、顆粒(被覆顆粒、含浸顆粒及び
均質顆粒)プレミツクス(飼料添加剤)
液体製剤:
(a) 水中で分散する有効成分:
水和剤、ペースト、エマルジヨン
(b) 溶液:
噴霧剤(エーロゾル)
固体の剤形(粉剤、散布剤)を製造するため
に、有効成分を固体の担体と混合する。適当な担
体としては、たとえばカオリン、タルク、膠灰粘
土、黄土、チヨーク、石灰石、重質炭酸カルシウ
ム、アタクレイ(attaclay)、ドロマイト、ケイ
ソウ土、沈殿シリカ、アルカリ土類シリケート、
ケイ酸ナトリウム、ケイ酸カリウム、ケイ酸アル
ミニウム(長石及び雲母)、硫酸カルシウム、硫
酸マグネシウム、酸化マグネシウム、粉砕合成物
質、肥料たとえば硫酸アンモニウム、リン酸アン
モニウム、硝酸アンモニウム、尿素など、またコ
ーンミール、樹皮粉、のこ屑、堅果殻ミール、セ
ルロース粉のような植物性産物の粉砕物、また植
物抽出の残留物、活性炭等が挙げられる。これら
の物質はそれのみで使用するか、もしくは互いに
添加して使用する。
顆粒は、前記式で表わされる有効成分を、有
機溶媒中に溶解し、得られた液をアタパルジヤイ
ト(attapulgite)、二酸化ケイ素、顆粒カルシウ
ム、ベントナイト等の顆粒状の鉱物に適用し、溶
媒を蒸発させることによつて、非常に容易に製造
することができる。
重合体顆粒もまた、前記式で表わされる有効
成分と、重合可能な化合物(尿素/ホルムアルデ
ヒド、ジシアンジアミド/ホルムアルデヒド、メ
ラミン/ホルムアルデヒド等)とを混合し、ゲル
状態のうちに粒状化を行いながら有効成分に影響
を与えない程度で穏やかに重合させることによ
り、製造することができる。また、比表面積と前
もつて決定づけられた好ましい吸着/脱色率を有
する加工処理した多孔性重合体顆粒(尿素/ホル
ムアルデヒド、ポリアクリロニトリル、ポリエス
テル等)に、有効成分をたとえば低沸点溶媒での
その溶液として含浸させ、その後溶媒を除去する
ことによりより有利に行える。好ましくは嵩密度
が300g/ないし600g/である微細顆粒の形
状の、この種の重合体顆粒は、またアトマイザー
の助けにより、製造することができる。
また、担体を有効成分及び添加剤と共に圧縮
し、その生成物を微粉砕することによつても、顆
粒を得ることができる。
これらの混合物に、有効成分を安定化させる添
加剤及び/または非イオン界面活性剤、陰イオン
界面活性剤、及び陽イオン界面活性剤たとえば良
好な水和性を確実にするもの(水和剤)、良好な
分散性を確実にするもの(分散剤)を加えること
もできる。適当な物質の例としては、オレイン/
チヨーク混合物、セルロース誘導体(メチルセル
ロース、カルボキシメチル セルロース)、1分
子につきエチレンオキシド基5ないし15個を有
し、アルキル部分の炭素原子数が8ないし9であ
るモノアルキルフエノール、及びジアルキルフエ
ノールのヒドロキシエチルグリコールエーテル、
リグニンスルホン酸、それらのアルカリ金属塩及
びアルカリ土類金属塩、ポリエチレングリコール
エーテル(カーボワツクス)、1分子につき、エ
チレンオキシド基を5ないし20個有し、脂肪アル
コール部分の炭素原子数が8ないし18である脂肪
アルコールポリエチレングリコールエーテル、エ
チレンオキシドの縮合生成物、プロピレンオキシ

び乳化性濃厚物は、水で希釈することによつて所
望の濃度とすることができる組成物である。
上記濃厚物は有効成分、担体、場合によつては
有効成分及び界面活性物質を安定させる添加剤、
また消泡剤、もし望むならば溶媒、とから成つて
いる。
水和剤及びペーストは有効成分を適当な装置内
で分散剤及び粉状担体と共に、均一になるまで混
合、粉砕することによつて得られる。適当な担体
は、たとえば前述のもののうち適用時に固体の剤
形のものである。いくらかの場合においては、異
なつた担体を混合して用いると有利である。分散
剤の例としては、スルホン化ナフタレン及びスル
ホン化ナフタレン誘導体とホルムアルデヒドとの
縮合生成物、ナフタレンもしくはナフタレンスル
ホン酸とフエノール及びホルムアルデヒドとの縮
合生成物、並びにリグニンスルホン酸のアルカリ
金属塩、アンモニウム塩、及びアルカリ土類金属
塩、加えてアルキルアリールスルホネート、ジブ
チルナフタレンスルホン酸のアルカリ金属及びア
ルカリ土類金属塩、また硫酸化ヘキサデカノー
ル、硫酸化ヘプタデカノール、硫酸化オクタデカ
ノールのような脂肪アルコール硫酸塩、及び硫酸
化脂肪アルコールグリコールエーテル、オレイル
メチルタウリドのナトリウム塩、ジ−第3−エチ
レングリコール、ジアルキルジラウリルアンモニ
ウムクロライド、及び脂肪酸のアルカリ金属塩、
及びアルカリ土類金属塩が挙げられる。
適当な消泡剤としてはたとえばシリコーン油が
挙げられる。
有効成分は、上記添加剤と混合粉砕、篩分けさ
れ、変形して、水和剤においては固体の粒径が
0.02ないし0.04mmペーストでは0.03mmとなり、そ
れ以上の大きさとはならない。乳化性濃厚物及び
ペーストは、前記のような分散剤、有機溶媒、及
び水を使用して製造する。適当な溶媒の例として
は、アルコール、ジメチルスルホキシド、及び沸
点が120゜及び350℃の間にある鉱油留分が挙げら
れる。上記溶媒は実際には無臭で、有効成分に対
し不活性でなければならない。
さらに、本発明による組成物は、溶液の形にお
いてでも適用できる。この目的のために、有効成
分もしくは通常前記式で表わされるいくつかの
有効成分を、適当な有機溶媒もしくは溶媒の混合
物中に溶解する。有機溶媒としては、脂肪族及び
芳香族炭化水素、その塩化物誘導体、アルキルナ
フタレン及び鉱油を単独でもしくは(互いに)混
合して用いることができる。
前記式で表わされる新規有効成分は、下記の
剤形とすることができる。部は、重量部を表わ

家禽飼料のような動物飼料 95部と均一に混合す

2%の噴霧剤を製造するため、次の物質を使用

他の殺生物作用を有する有効成分及び他の殺生
物剤を、上記組成物に混合してもよい。このよう
に通常前記式で表わされる引用化合物に加え
て、本発明による組成物はたとえば、広い活性ス
ペクトルを有する殺虫剤を含有することができ
る。
上記組成物もしくは有効成分は、好ましくは双
翅目の昆虫の、主に幼虫の発育もしくはさなぎに
対する防除作用を有するものである。
実施例 1
2−シクロプロピルアミノ−4−ホルミルアミ
ノ−6−モルホリノ−s−トリアジン
2−シクロプロピルアミノ−4−N′・N′−ジ
メチルホルムアミジノ−6−モルホリノ−s−ト
リアジン 25gを、水1000ml中に溶解し、得られ
た溶液を1N塩酸100mlで処理した。室温で15分間
撹拌した後、融点288℃の2−シクロプロピル−
アミノ−4−ホルミルアミノ−6−モルホリノ−
s−トリアジンが沈殿し始めた。粗生成物をジメ
チルホルムアミドから再結晶した。融点291−292
℃。
前記式で表わされる以下の化合物は、上記製
造方法により製造される。

実施例 2
イエバエ(Musca domestica)に対する作用
新しく製造したうじ用CSMA培地をビーカー
に入れた。各々の有効成分の1%アセトン酸溶液
を、2.5mlずつビーカー中の培地にピペツトで入
れた。上記培地を完全に混合し、その後溶媒を留
去した。
生後1日目、2日目及び3日目のイエバエのう
じ25ひきと、イエバエの卵約50ぴきを、それぞれ
の有効成分を試験するための、処理済み培地を含
有するそれぞれのビーカーに入れた。うじがさな
ぎとなつた後に、そのさなぎを水で洗い流すこと
によつて培地から分離し、数を数えた。さなぎか
らかえつたハエの数を、10日後に数え、変態にす
る何らかの影響を観察した。
上記試験において、実施例1の化合物は、イエ
バエに対して良好な作用を示した。
実施例 3
ルシリア・セリカータ(Lucilia sericata)に
対する作用
0.5%の有効成分を含有する水性溶液1mlを50
℃で培地9mlに加えた。その後、新しくかえつた
ルシリア・セリカータの幼虫約30ぴきを培地に入
れ、それぞれ48及び96時間後の死虫率を測定する
ことによつて、その殺虫作用を決定した。上記試
験において、前記式で表わされる化合物はルシ
リア・セリカータに対し良好に作用した。
実施例 4
ネツタイシマカ(Ae¨des aegypti)の幼虫に対
する作用
一定数のビーカーのそれぞれに入つている150
mlの水の中に、有効成分の0.1%アセトン溶液の
特定量をピペツトで入れることにより、有効成分
の濃度がそれぞれ10、5及び1ppmの溶液を得た。
アセトンを蒸発させた後、生後2日目のネツタイ
シマカの幼虫30ないし40ぴきを有効成分の溶液の
入つたそれぞれのビーカー内へ入れた。1日後、
2日後及び5日後にそれぞれ死虫率を測定した。
上記試験において、前記式で表わされる化合
物は、ネツタイシマカに対して良好な作用を示し
た。
実施例 5
試験A:双翅目(Order Diptera)のイエバエ
(Musca comeotica)の成長抑制
試験物質:アセトン中0.1%の試験化合物
試験対照物:イエバエ
試験材料:ウジ用培地;製造方法:酵母100g
を水9に懸濁する。この懸濁液をぬか/ム
ラサキウマゴヤシ(1:1)の混合物5Kgに
添加して充分に混合する。5日後に試験に適
する材料が得られる。〕

試験法:培地50gを200mlのポリエチレン製ビ
ーカーに入れる。各試験化合物の0.1%アセ
トン溶液5mlをビーカー内の培地上にピペツ
トで添加する。この培地を充分に混合した
後、少なくとも20時間かけてアセトンを蒸発
させる。その後、イエバエの2日たつた卵25
個を各有効成分で処理された試験用培地を含
むビーカー及び対照群としての未処理培地を
含むビーカーに入れる。各試験あたり2個の
ビーカーを使用する。5日後、サナギを培地
から水で洗い出し、ビーカーに入れ、銅製の
網の蓋でおおう。
評価方法:培地から分離した後、正常及び奇形
のサナギの数を数える。また、試験開始後10
日間にサナギから羽化した成虫の数を数え
る。
