Abstract

PURPOSE:To obtain an affinitive adsorbent for separation and purification of a specified enzyme or the like by combining[adenylyl-(3' 5')]n-adenosine oligomer used as ligand with a supporting body at an amino position of a base. CONSTITUTION:[Adenylyl-(3' 5')]n-adenosine oligomer (where n is a positive integer) is combined with an agarose derivative having an ester group used as a supporting body through the intermediary of a spacer -CO(CH2)4CH2 or the like, whereby an affinitive adsorbent (having combinations shown by the formula) for affinity chromatography is obtained. This adsorbent is used for specifically separating from other mixtures and purifying protein or enzyme such as polynucleotide phosphorylase capable of combining with[adenylyl-(3' 5')]n-adenosine oligomer which is obtained by adsorbing and separating by using an anion exchange substance from a liquid extracted from crushed bacterial body of Thermus thermophyllus. In this way, a specified enzyme or the like is obtained with high purity and high activity.
