Abstract

PURPOSE:To make it possible to separate and purify with excellent purity a substance (enzyme or the like) capable of combining with adenosine diphosphate, by combining adenosine diphosphate used as ligand with a supporting body at specified positions of ribose. CONSTITUTION:Adenosine diphosphate used as ligand is combined with a supporting body such as an agarose derivative through the intermediary of a spacer -(CH2)-6 at positions 2'-C and 3'-C of ribose, whereby an affinitive adsorbent (shown by the formula) for affinity chromatography is prepared. This adsorbent is used, for instance, for separating and purifying, with high purity and without contaminating of other similar proteins or the like, polynucleotide phosphorylase which is an enzyme capable of combining with adenosine diphosphate obtained from a liquid extracted from crushed bacterial body of Thermus thermophyllus through adsorption and separation by using an anion exchange substance.
