Abstract
PURPOSE: The extraction with a surfactant, the fractionation of the extract with a zinc salt and other operations are effected to separate and purify the A Tase from tissues of human placenta.
CONSTITUTION: Tissues of human placenta are homogenized together with an aqueous solution containing an anionic or nonionic surfactant and centrifuged into the precipitate and the supernatant. The supernatant is fractionated by salting out with a zinc salt such as zinc acetate or zinc sulfate to recover the resultant supernatant. It is adjusted in pH to 6W8, brought into contact with an anion exchanger equilibrated with a buffer solution of 6W8pH, eluted with an aqueous solution containing 0.5W2M salt and the eluate is treated with a protease for decomposition. Then, the product is subjected to gel filtration to isolate the fraction in which A Tase is active.
