Abstract

PURPOSE: The glucose in serum is previously removed by the hexokinase reaction system, then starch is added as a substrate to measure free glucose, thus determining the activity of the titled substance rapidly and accurately.
CONSTITUTION: The glucose in serum is previously removed by the hexokinase reaction system and the hexokinase is deactivated with an anionic surfactant. A modified starch is added as a substrate and the glucose liberated by the action of α-amylase is measured colorimetrically using an enzymatic reaction system consisting of glucose oxidase, peroxidase, 4-aminoantipyrin and a coupler to determine the activity of the α-amylase in serum.
