Abstract

PURPOSE: After the first solution containing oxidase, peroxidase, phenolic derivative, aniline derivative or naphthol derivative is added to a sample, the second solution of coupler is added thereto to determine the substrate or enzyme activity.
CONSTITUTION: The first solution is prepared using an oxidase such as cholesterol oxidase, glycerol oxidase or pyruvic oxidase, peroxidase, a phenolic derivative such as phenol or p-chlorophenol, an aniline derivative such as aniline or N,N-dimethylaniline or a naphthol derivative such as 1-naphthol or 2-naphthol and the second solution is prepared using a coupler. After the first solution is added to a sample, the second solution is added thereto to effect exact determination of the objective substrate or enzyme activity without overdetermination of free cholesterol and free glycerol.
