Abstract

PURPOSE: Absorptivities of the first solution containing a standard substance and of the second standard solution containing the same amount of water instead of the standard substance are measured to determine enzymes simply and accurately.
CONSTITUTION: A mixture containing (A) a substrate that supports an enzymatic reaction in the presence of an enzyme to be determined, (B) a reagent that reacts with the reaction product to show absorptivity and (C) an enzyme catalyzing the reaction is combined with (D) the enzymatic reaction product from the substrate A as a standard substance to prepare the first standard solution. In the meantime, the same amount of water is added instead of the standard substance in the first standard solution to prepare the second standard solution. The absorptivities of the first and second solutions are measured and the device constant is calculated from the difference. Further, the absorptivities of the second standard solution and a sample containing the same amount of a solution containing the enzyme to be determined instead of water are measured and the enzyme is determined from the difference between them and the above device constant.
