Abstract

PURPOSE:To set an accurate standard curve without complicated operations by subjecting the coordinates of the quantity of antigen to log transformation, by estimating values of radioisotope (RI) at times when the quantity of antigen is 0 and infinite, and by subjecting the coordinates of a measured value of RI to logit transformation based on the measured and estimated values. CONSTITUTION:In setting a standard curve at the time when a linked antibody-labelled- antigen substance is measured by utilizing the competitive reaction between antigen in a known quantity and RI-marked antigen in a prescribed quantity so as to analyze the antigen in a sample, the quantity of antigen is made to be x and this is subjected to log transformation. Next, a measured value y of RI corresponding to x is found, and the measured values y of RI at the times when the quantity of antigen is 0 and infinite are supposed from a plurality of known quantities (x) of antigen and the measured values (y) of RI corresponding thereto by using a specific relational expression, and are made to be B0 and N, respectively. Based on these supposed values B0 and N and (y), the measured values (y) of RI are subjected to logit transformation, and the measured values of RI are represented in the standard curve on the coordinates of the respective transformed values by a method of least squares. By using this standard curve, the quantity of antigen in the sample can be determined with little error from the measured values of the RI of the sample.
