Abstract

PURPOSE: To determine uric acid in blood serum using uricase/peroxidase/hydrogen-donor system, accurately, without interference of ascorbic acid, by removing ascorbic acid in the blood serum with Cu2+ and Fe2+ before the determination.
CONSTITUTION: Ascorbic acid is removed from blood serum containing uric acid by adding 0.2W0.5mmol/l of Cu2+ and 0.02W0.06mmol/l of Fe2+to the serum at pH3W5. After removing ascorbic acid, uric acid in the serum is determined by the use of a reaction system comprising uricase, peroxidase and a hydrogen donor. The method enables the accurate and reproducible determination of uric acid in a specimen containing a large amount of ascorbic acid which is a strong reducing substance, without the interference of ascorbic acid.
