Abstract

PURPOSE: To measure a nonelectron pair bond fatty acid quantitatively, by treating a nonelectron pair bond fatty acid, coenzyme A (Co A), etc. with an enzyme and decomposer, followed by converting the remaining Co A to 2-nitro-5-mercaptobenzoic acid, which is determined optically.
CONSTITUTION: Acyl coenzyme A synthetase (ACS) and a decomposer for adenosine-5'-triphosphate (AMP), e.g., myokinase, etc. are added to and brought into contact with a specimen containing a nonelectron pair bond fatty acid (NEFA), coenzyme A (Co A) and AMP in pH7W9 at room temperature to 40°C. Consequently, the above-mentioned substances which is treated in the specimen are converted enzymically to acyl coenzyme A, adenosine-5'-phosphate (AMP), and pyrophosphoric acid, and simultaneously AMP and pyrophosphoric acid are transformed or decomposed. The remaining Co A in the treatment is reacted with 5,5'-dithiobis- (2-nitrobenzoic acid) to give 2-nitro-5-mercaptobenzoic acid (TNB-), which is determined optically to measure a nonelectron pair bond fatty acid. The reactions are shown by the formulas I and II.
