Abstract

PURPOSE: To measure the CF values of an antigen and an antibody with high sensitivity by stacking an agar plate wherein a complement and an antigen or an antibody is mixed and an agar plate wherein sensitized blood corpuscles are mixed to cause hemolysis and measuring diameters of produced annular bodies.
CONSTITUTION: A purified antigen or antibody and a complement such as guinea pig blood serum of appropriate concentrations are added to an agar solution for electrophoresis to prepare a plate A, and then a hole for pouring in specimen blood serum or an antigen is made. In the same way, an agar plate B including sensitized blood corpuscles is prepared. Standard blood serum or antigen and specimen blood serum or antigen are poured into the hole of the agar plate A and they are put into a closed vessel to cause an antigen-antibody reaction. If the plate A and the plate B are stacked to cause a hemolysis, an annular body of insoluble blood whose diameter varies with a difference between the CF value of the standard blood serum or the standard antigen and the CF value of the specimen blood serum or the specimen antigen is produced and therefore the CF value of the specimen can be determined by measuring the diameter.
