Abstract
PURPOSE: To prepare a saccharification solution with high concentration, by subjecting starch of corn to direct enzymatic liquefaction and saccharification without separating and purifying.
CONSTITUTION: Corns, e.g., maize, milo, wheat, etc. are ground so that skins and embryo-buds are removed, and they are powdered into granules ≤100 mesh. Water or warm water is added to the powdered corns to prepare slurry of crude starch with 30W35g/dl. 50W100 Unit (AU) pergstarch of an enzyme to liquefy starch is added to the corn slurry, crude starch is added to the slurry continuously or in batch way in such a way that stirring is carried out at a temperature < the paste temperature of starch and the viscosity of the slurry is in a range lower than 3,000 cp, to prepare low-viscosity slurry of crude starch comprising substantially ≥35wt% starch. The slurry is then subjected to enzymatic liquefaction and enzymatic saccharficiation conventionally.
