Abstract
PURPOSE: A specific strain in Bacillus stearothermophilus is cultured and pyruvate kinase is collected from the culture mixture, thus permitting the practical production of heat-resistant pyruvate kinase in a large scale.
CONSTITUTION: Bacillus stearothermophilus UK788 strain is cultured and heat-resistant pyruvate kinase is collected from the resultant culure mixture. The culture medium for the UK788 strain is preferably liquid and the strain is cultured at 20W 80°C batchwise or continuously. In the batchwise operation, the cultivation is preferably continued till the late stage in the logarithmic growth. In the continuous cultivation, the chemostat-type continuous cultivation is conducted near the maximum specific growth rate of the UK788 strain, thus producing cell bodies containing the heat-resistant pyruvate kinase in a high concentration.
