Abstract

PURPOSE: To prepare an optically active diol derivative which can be converted to an optically active β-blocker, economically, in an industrial scale, by reacting an optically active halohydrin derivative with an aromatic alcohol under basic conditions.
CONSTITUTION: An optically active diol derivative of formula III (Ar is aromatic group) can be prepared in a short step, without racemization, by reacting a halohydrin derivative of formula I (X is halogen; one of Y and Z is H and the other is acryl) with an alcohol of formula II in the presence of a base (e.g. alkali metal alkoxide. oxide or carbonate, alkaline earth metal hydroxide, etc.), pref. in an aqueous solvent. The optically active compound of formula I can be obtained easily by the asymmetric hydrolysis of 1,2-diacyloxy-3-halopropane with lipase, and the compound of formula II is also available easily as an industrial raw material.
