Abstract

PURPOSE: The condensation reaction between a cytosine derivative and 3-deoxyribose derivative is effected in the presence a Lewis acid and protecting groups are removed to prepare title compound that is important as synthetic intermediate of pyrimidine-3'-deoxynucleoside derivatives with reduced steps in high yield.
CONSTITUTION: N4-Acylcytosine of formula I (R1 is acyl) is silylated with hexamethyldisilazane or trimethylchlorosilane and subjected to condensation reaction with a 3-deoxyribose derivative of formula II (R2 is aroyl; R3 is alkylcarbonyloxy, or halogen) in the presence of a Lewis acid as tin tetrachloride and the prorecting groups are removed by treating with an alkoxide to give 3'-deoxycitydine of formula III. The 3-deoxyribose derivative is obtained from cordycepin that is readily prepared by fermentation by only two steps.
