Abstract
PURPOSE: To prepare highly active heparin in simple procedures efficiently, by adsorbing heparin (H) on an immobilized protamine, and by treating H with salt solutions in a low and high concentrations in order.
CONSTITUTION: Heparin in a solution (A) is adsorbed on an immobilized protamine, and then treated with a salt solution (B) in a low concentration or a buffer having the ionic strength corresponding to (B) to elute and remove the low active heparin fraction. The heparin is then eluted with a salt solution(C) in a higher concentration or a buffer having the ionic strength corresponding to (C) to elute the heparin.The solution (A) is obtained by partly purifying heparin extracted from the livers, lungs, or intestinal mucous membranes of swine, cows, or sheep, or by dissolving a commercial heparin standard sample, preferably in a buffer. The protamine is immobilized on Sepharose, granular starch, or synthetic resin by covalent bonding with a cyanogen bromide or hydrazine.
