Abstract

A substrate plasma, useful in the assay of blood coagulation factors in human beings and other species, is prepared by treating blood plasma from a first species of animal with an adsorbent (as hereinafter disclosed) to remove therefrom all but a small residual amount of one or more blood coagulation factors and adding to the treated plasma a small proportion of a serum from a second species of animal containing antiobodies immunologically specific for the said coagulation factors to remove the small residual amount of the said factor or factors from the plasma. The serum is obtained by injecting an animal of the second species with a concentrate containing the said one or more coagulation factors which has been prepared from the blood plasma or serum of an animal of the first species, drawing blood from the animal after an interval of time in which the antibodies have developed and separating the serum from the blood so obtained. The concentrate is prepared by treating plasma or serum from the first species with an adsorbent to adsorb one or more coagulation factors therefrom, separating the adsorbent from the plasma or serum, eluting the adsorbed factors and concentrating the eluate to obtain a concentrate of said coagulation factors. Specified adsorbents are calcium phosphate, barium sulphate, asbestos and an ion exchange cellulose derivative. Aqueous solution of sodium citrate may be used for elution. The substrate plasma mixed with thromboplastin, calcium chloride and a barbitone buffer is lyophilised for storage and is reconstituted with distilled water for use.
