Abstract
NEW MATERIAL:A 3-[ 4-( 4'-halogenobenzyloxy )phenoxy ]-propylphosphorylethanolamine of formula I (X is halogen), and its salt.
USE: Hypotensive drugs, having the hypotensive and the anti-renin actions, with a relatively low toxicity.
PROCESS: A 4-(4'-halogenobenzyloxy)phenol of formula II is reacted with a 3-halogeno-1-propanol of formula III (X3 is halogen) to give a propanol of formula IV, which is reacted with an 2-(N-substituted amino)ethylphosphoric acid or its derivative of formula V (X1 and X2 are halogens, etc.; Z1 and Z2 are amino groups) in an organic solvent in the presence of an acid binder, e.g. pyridine, or a condensing agent, e.g. dicyclocarbodiimide, to give an ethylphosphoric acid derivative of formula VI. The derivative is then hydrolyzed to form an ethylphosphoric acid derivative of formula VII, which is treated with an acid to afford the compound of formula I.
