Abstract

PURPOSE:To estimate phenylthiohydantoic amino acid quickly and accurately in simple operation by generating fluorescent light by reacting phenylthiohydantoic amino acid with pyridoxamine and zinc acetate. CONSTITUTION:Pyridoxine and zinc acetate are reacted with phenylthiohydantoic (PTA) amino acid which is produced by Edman's decomposition of peptide or protein, and fluorescent light is generated, thereby PHT amino acid is estimated. For example, 250mug of pyridoxamine and 5ml of 2% pyridine methanol solution containing 250mg zinc acetate are added to 0.5ml of ethyl acetate containing 0.124mug of PHT-norleucine and churned. After the product is heated for 90 minutes at 70 deg.C, fluorescent light is generated. Then, it is cooled for 5 minutes by ice bathing, and measured by an excitation wavelength of 390nm and a fluorescent-light wavelength of 465nm, thereby a spectrum of PHT-amino-acid as shown by the pattern in the Figure is obtained.
