Abstract

PURPOSE: To make it possible to accomplish the discrimination and inspection highly sensitively, promptly and peculiarly with the use of the latex coaggulating reaction by coupling the adeno virus antigen to a special polystyrene latex.
CONSTITUTION: Adeno virus is cultivated in HeLa cell, and an antigen is extracted from the cell and the cultivating liquid. Then, the antigen is treated with fluorocarbon to eliminate any protein other than the antigen so that the purified adeno virus antigen is prepared. The polystyrene latex having a specific weight of 1.14 and a size larger than 0.91μ is made into 0.25% floating liquid with PBS, and the aforementioned purified antigen liquid is added thereto in the same quantity. The mixture is then treated at a room temperature for 2 to 4 hours and then is rinsed. The mixture thus rinsed is duspended in the free-drying medium, which is prepared by NPS to the PBS and glycine and dextran to the dilution from NaN3, followed by the free-drying process. The mixture, which is prepared by adding ten times the original quantity of the dilution to the freeze-dried latex, is used to discriminate the serum by the microtiter method. Then, the judgement can be accomplished for about 10 hours with the sensitivity of 160 to 200 times that of the conventional method.
