Abstract

NEW MATERIAL:The free acid of a monotetrazolium salt of formula (when R1 is sulfone, R2 is H; when R1 is H, R2 is carboxy; X is Cl or Br), wherein its 5-phenyl group is substituted at 2- or 4-position; or its alkali metal salt.
EXAMPLE: 3-(4-Iodophenyl)-2-(4-nitrophenyl)-5-(4-carboxyphenyl)-2H tetrazolium chloride (R1=H and R2=COOH in the formula).
USE: Useful as a hydrogen-acceptor in absorption photometry of a dehydrogenase. For example, lactic acid dehydrogenase in blood is determined for the diagnosis of a disease of heart muscle, liver, etc.. It has improved solubility and gives reproducible results.
PREPARATION: For example, a 2- or 4-substituted benzaldehyde is reacted with 4-nitrophenylhydrazine in an organic solvent. The product is mixed with 4-iodophenyldiazonium chloride under basic conditions, and oxidized under acidic conditions to give the compound expressed by the formula.
