NEW MATERIAL:A bicyclooctane derivative of formula I: (R1 is H or lower alkyl group; R2 is ≤7C straight or branched chain alkyl group; Y is H or OH-protecting group).
EXAMPLE: 2β- ( 3'-Oxo-1'-trans-octenyl ) -3α-hydroxy-7E- (4'-methoxycarbonyl-butylidene)-bicyclo[3.3.0]octne.
USE: Intermediates for the synthesis of stable Prostaglandin I2 analogs and medicines having blood platelet aggregation inhibitory actions, useful for treating arteriosclerosis, heart failure, and thrombosis.
PROCESS: An alcohol derivative of formula II is oxidized with an oxidizing agent to give an aldehyde derivative of formula III, which is reacted with a Wittig reagent of formula IV (R3 is lower alkyl group) or formula V (R4 is aryl group) and if necessary the ester is hydrolyzed or the carboxylic acid is esterified to form the bicylooctane derivative of formula I.
