Abstract

PURPOSE: To enable measurement of good accuracy to be made because of low activity of coenzyme itself and solve problems such as short working life by covering the end of measuring electrode with the fixed coenzyme film comprising fixing coenzyme to a porous high polymer material.
CONSTITUTION: A reference electrode block 1 provided with a reference elecrode 15 consisting of a porous member 17, electrolyte 18, Ag-AgCl electrode 19 being disposed in an opening part 16 and a working electrode 21 consisting of a copper plate 22, glassy carbon plate 23, fixed coenzyme film 24 are provided and are opposed to the working electrode block 2. A voltage is then applied thereto. The measuring sample and substrate-containing buffer solution in tanks 9, 10 are passed through the sample chamber 3 by a metering pump 12. The enzyme in the sample promotes reaction of substrate and coenzyme and lets the coenzyme to change to oxidized or reduced form but this coenzyme changes again to the original form owing to electrochemical circulating action. The change in the current of this time is measured with a measuring instrument 26 connected to the electrodes 15, 21 by lead wires 27.
