Abstract

PURPOSE: To synthesize peptide derivatives using protease.
CONSTITUTION: Peptide derivatives S-A-B-Y, X-A-B-C-OH ro X-A-B-C-Y (X is terminal amino-protecting group, amino acid residue or peptide residue having protected terminal amino group; A is amino acid residue having protease specificity; Y is terminal carboxyl-protecting group; B is amino acid residue having protease specificity; C is amino acid residue, peptide residue) are prepared by reacting (a) an L-amino acid or a peptide derivative X-A-OH with (b) excess or deficient amount of an L-amino acid or a peptide derivative H-B-Y, H-B-C-OH, or H-B-C-Y, in the presence of protease at 20-50°C, pref. 30-40°C, and at a pH optimum to the protease.
