Abstract

PURPOSE: To prepare ribonucleic acid in high efficiency, by extracting specific novel yeast with hot neutral or alkaline aqueous solution of NaCl.
CONSTITUTION: Pure ribonucleic acdi (RNA) is prepared by (1) dispersing 5- 10wt% of yeast obtained by the cultivation of Candida tropicalis C-1151 (FERM- P No. 4173), in water, (2) adding about 1-15 wt% of NaCl to the dispersion, (3) adding an alkaline aqueous solution to the dispersion with stirring to adjust the pH value between 7 and 8 (pref. 7.5 and 7.8), (4) heating the dispersion at about 70-100°C for 30-200 minutes to extract the RNA, (5) removing the extraction residue by centrifugal separation, (6) adding about 0.5-5 wt% of CaCl2 to the extract containing RNA, (7) adjusting the pH of the solution to ≤ 2, by adding HCl and (8) separating the precipitate
