Abstract

PURPOSE: To prepare an immobilized enzyme having excellent mechanical strength and water resistance, by mixing a specific isocyanate-terminated prepolymer with an aqueous solution of enzyme, adding a hydrophilic organic solvent to the mixture prior to the completion of the foaming reaction thereby breaking the foam, casting the composition to a film, and finally drying the film.
CONSTITUTION: An isocyanate-terminated prepolymer is prepared by reacting an isocyanate compound with a hydrophilic polyether comprising one or more EO/PO copolymers having an EO content of ≥50% and an average molecular weight of 500W10,000. 0.1W10parts of an aqueous solution of enzyme to be immobilized, is added to 1 part of the prepolymer, and made to react under agitation. When the degree of the foaming reaction reaches 50W70%, 0.1W3 times volume, based on the enzyme solution, of a hydrophilic organic solution such as acetone is added to the reaction mixture thereby reducing the viscosity and breaking the foams. The defoamed solution is cast to a film at room temperature, and made to gel by drying.
