Abstract

PURPOSE: To obtain excellent reproducibility of elution time of component such as amino acid of protein hydrolysate, by passing a specific buffer into the passage upstream of separation column before supplying the first stage buffer after supplying regenerating liquid into the separation column.
CONSTITUTION: The first stage eluate B1 runs through a T-pipe 3, to which are connected a solenoid valve V1, manifold 1, pump 2, damper 4 and pressure gauge 5, sampler 6 and separation column 7, and the sample solution of which components are separated is mixed with a coloring reagent 8 which is sent through a pump 9. The mixture develops color in a reaction vessel 10, and the optical characteristics are measured by a photometer 11. Eluates B1 to B3 are successively changed over, and when the elution of amino acid from the column 7 is over, regenerating liquid RG is passed in, so that the column filler is regenerated. After the second eluate B2 of which power to swell the filler is smaller than the eluate B1 which contains the substance to swell the said filler is passed for a certain time, the eluate B1 is permitted to flow so as to balance the column 7. Thus, pressure elevation may be avoided, and the liquid RG may not be left in the pipe 3.
