Abstract

PURPOSE: To control the culturing conditions rapidly and accurately, by measuring the concentration of vaporizable components in a carrier gas diffused from the culturing medium through a hydrophobic open-cell micro-porous membrane separating the carrier gas and the medium.
CONSTITUTION: An opne-cell micro-porous flat or tubular membrane made of a hydrophobic material such as PTFE, PP, PVC, etc., is placed in a cultruing vessel. A culturing medium is brought into contact with one face of the membrane, while a carrier gas such as nitrogen, air, etc., is with the reverse side thereof. Concentrations of main carbon source or metabolized products diffused to the carrier gas through the micro-pores of the membrane, is measured. The temperature, pH value, rate of substrate flow, air flow, and dissolved oxygen concentration, etc., can be controlled to the optimum values according to the measured concentration or the rate of assimilation or production calculated therefrom. The process enables the automatic control of the culturing system, and improves the qualities and yield of the cultured product.
