Abstract
1475151 Derivatives of piperidin-4-ol SANDOZ Ltd 18 June 1974 [22 June 1973 28 June 1973 29 June 1973 2 July 1973] 26850/74 Heading C2C The invention comprises compounds of formula and their acid addition salts, wherein R 1 is Ph(CH 2 ) 2 or PhCO(CH 2 ) 3 ; H 2 is OR 5 or NR 6 R 7 where R 5 is C 1-4 alkyl, C 3-5 alkenyl, or Ph, and R 6 , R 7 are each H, C 1-4 alkyl, C 4-6 cycloalkyl-C 1-4 -alkyl, C 4-6 cycloalkyl, Ph or Ph-C 1-4 alkyl, orNR 6 R 7 constitutes a saturated heterocyole having 5-6 ring members; R 3 is C 1-4 alkyl or C 4-6 cycloalkyl-C 1-4 -alkyl, C 4-6 cycloalkyl or Ph, or [when R 1 is Ph(CH 2 ) 2 ] C 2-4 alkenyl, and R 4 is H or C 1-4 alkyl, or CR 3 R 4 constitutes a C 4-6 cycloalkane ring; and the phenyl group Ph may be substituted by F, Cl, Br or C 1-3 alkyl or alkoxy. In examples, these compounds are prepared by (1) reacting the corresponding piperide-4-one with LiCR 3 R 4 COR 2 , (2) introducing R 1 into the corresponding N - unsubstituted piperidin - 4 - ol (which is itself made by hydrogenolysis of the N-benzyl derivative), (3) [for R 1 = PhCO(CH 2 ) 3 ] hydrolysing the analogue where R 1 = 3-(2-phenyl- 1,3-dioxolan-2-yl)propyl, (4) [for R 2 =NH 2 ] reacting the corresponding nitrile with H 2 O 2 , (5) [for R 2 = NR 6 R 7 ] reacting the phenyl ester with HNR 6 R 7 . Therapeutic compositions having analgesic activity comprise compounds of the above formula, and may be administered orally, parenterally or rectally.
