Abstract
Description
Claims (1)
- 【特許請求の範囲】 1.下記工程: a)カベオレとGドメインを含有してなり、細胞膜のカベオレが発生する側とは 反対側に存在するコロイドシリカ粒子で被覆された精製細胞膜を製造する工程; b)コロイドシリカ粒子で被覆された該精製細胞膜を膜破砕法に供し、それによ り精製細胞膜からカベオレを取り出し、該精製細胞膜にもはや付着していないカ ベオレとGドメインを含む精製細胞膜断片を製造する工程; c)工程b)の産物を密度に基づく分離技術に供し、それによってカベオレが他 の精製細胞膜断片から分離される工程;および d)工程c)で分離したカベオレを、他の精製細胞膜断片から取り出し、それに より精製カベオレを製造する工程 を含む精製カベオレの製造方法。 2.膜破砕法が、約4℃〜8℃の温度で適当なディタージェントの存在下で行な われる請求項1記載の方法。 3.膜破砕法が剪断であり、ディタージェントの非存在下で行なわれる請求項1 記載の方法。 4.請求項1〜3いずれか記載の方法により調製される精製カベオレ。 5.下記工程: a)カベオレとGドメインを含有してなり、コロイドシリカ粒子で被覆された精 製細胞膜を製造する工程; b)工程a)で製造された精製細胞膜からカベオレを取り除き、それによりカベ オレを欠いたシリカ粒子で被覆された細胞膜を製造する工程; c)カベオレを欠いた該シリカ粒子で被覆された細胞膜を、細胞膜からのシリカ 粒子の分離を生ずる条件に供し、それによりカベオレを欠いた細胞膜を製造する 工程; d)カベオレを取り除いた細胞膜を、約4℃〜8℃の温度で適当なディタージェ ントの存在下で膜破砕法に供し、それによって細胞膜断片を製造する工程; e)工程d)の産物を密度に基づく分離技術に供し、それによってGドメインが 他の細胞膜断片から分離する工程;および f)工程e)で分離したGドメインを他の細胞膜断片から取り出し、それにより 精製Gドメインを製造する工程 を含む精製Gドメインの製造方法。 6.シリカ粒子の分離を生ずる条件が高塩濃度の条件である、請求項5記載の方 法。 7.請求項4〜6いずれか記載の方法により製造される精製Gドメイン。 8.下記工程: a)カベオレとGドメインを含有してなり、コロイドシリカ粒子で被覆された精 製細胞膜を製造する工程; b)該精製細胞膜を、細胞膜からのシリカ粒子の分離を生ずる条件に供し、それ により細胞膜を製造する工程; c)該細胞膜を、約4℃〜8℃の温度で適当なディタージェントの存在下で膜破 砕法に供し、それにより精製細胞膜断片を製造する工程; d)該精製細胞膜断片を密度に基づく分離技術に供し、それによってGPIアン カー蛋白質のマイクロドメインと関連するカベオレから本質的になる細胞膜ドメ インが、他の精製細胞膜断片から分離する工程;および e)GPIアンカー蛋白質のマイクロドメインと関連するカベオレから本質的に なる細胞膜ドメインを、他の精製細胞膜断片から取り出す工程 を含む、GPIアンカー蛋白質のマイクロドメインと関連するカベオレから本質 的になる細胞膜ドメインを、内皮細胞の細胞膜から単離する方法。 9.請求項8記載の方法により製造される、本質的にカベオレとGPIアンカー 蛋白質のマイクロドメインとからなる細胞膜。 10.精製細胞膜が内皮細胞の細胞膜である、請求項1、5または8いずれか記 載の方法。 11.膜破砕法が剪断である、請求項10記載の方法。 12.ディタージェントがTriton X−100であり、密度に基づく分離 技術がショ糖密度勾配遠心分離である、請求項2、5または8いずれか記載の方 法。 13.精製カベオレに特異的なモノクローナル抗体。 14.肺のカベオレに特異的なモノクローナル抗体。 15.精製カベオレを含有してなる薬物送達系。
