成形ベルトカレンダー
本発明は、活性成分を含む生成物の組成物(product composition)から扁平
な成形物、特に錠剤を製造するための装置および方法に関する。
錠剤を製造するための公知の方法は溶融カレンダー法である。このタイプの方
法は例えばドイツ国特許公開第1766546号明細書および米国特許第488
0585号明細書に記載されている。この方法の原理は活性成分をキャリアー、
例えば脂肪物質、または水溶性の熱可塑性ポリマーのメルト(溶融物)内に埋め
込むことである。メルトは活性成分、ポリマーおよび適当な場合には他の補助物
質の混合物によって、例えば押出し機内で溶融されて製造され、かつメルトとし
て下流の成形カレンダー内で錠剤に成形され、錠剤は冷却時に硬化する。成形カ
レンダーは1対の逆回転するロールを備え、ロールは表面に所望の錠剤の片半部
の形状に相当する彫刻(凹所)を有する。錠剤の成形は、2つのロールの接触領
域内で一方のロールの凹所内の錠剤組成物と他方のロールの対向凹所内の錠剤組
成物との組合せによって行われる。
しかし錠剤を製造するこの方法は欠点を持つ。すなわち例えばロールの構造が
きわめて複雑である。凹所
がロール内に切り込まれ、かつ最大の滑らかさを持つ錠剤表面を得るためにはた
いていの場合に引き続き研磨されなければならない。このようなロールの製作は
きわめて費用がかかるものである、それというのも凹所は大きな精度を有して形
成されなければならないからである。これは、2つの凹所が常にできる限り正確
に対向するようにロールが調整されなければならないためである。この条件は特
に凹所がきわめて小さい場合には達成するのは困難である。
更にこのような装置は柔軟ではない、なぜならば例えば同一の装置で異なる錠
剤形状およびまたはサイズのものを製造したい場合、ロールの完全な交換および
念入りな再調整なしでは可能ではないからである。しかし種々の錠剤形状および
サイズに関する大きな柔軟性は、特に薬剤分野の錠剤製造のための装置で望まれ
ることである。
スイス国特許第530944号(CH−A530944)から扁平な成形物を
製造するための装置が公知であり、この装置は生成物の組成物を供給するための
の装入手段少なくとも1つと、2つの逆回転可能なカレンダーロールから成る少
なくとも1対のロールとを備え、かつ循環する成形ベルト、有利に連続ベルトを
備え、ベルトは1対のロールの2つのロール間を通過するようになっており、成
形ベルトは生成物の組成物を受容するために表面に凹所を持つ。この公知の装置
は比較的大きな成形物を製造するために使用される。
本発明の目的は、一方で公知の装置よりもコストが小さく、他方でより大きな
柔軟性を与える、活性成分を含む生成物の組成物から扁平な成形物を製造するた
めの装置を提供することである。また錠剤製造の簡単で柔軟な方法の提供も本発
明の目的である。
この目的は、生成物の組成物を供給するための少なくとも1つの装入手段と、
2つの逆回転可能なカレンダーロールから成る少なくとも1対のロールとを備え
た、活性成分を含む生成物の組成物から扁平の成形物を製造するための装置であ
って、1対のロールの2つのロール間を通過する循環する成形ベルト、有利に連
続ベルトを備えており、成形ベルトが生成物の組成物を受容するために表面に凹
所を有する形式のものにおいて、凹所が成形ベルトに貫通アパーチャー(contin
uous aperture)として形成されていることによって達成される。
ロールは、これらがこれらの外面間にスリット様の通路を持つように配置され
る。更に1対のロールの外面間の最小距離がほぼ成形ベルトの厚みに相当する。
生成物の組成物は有利に広幅の生成物リボン(product ribbon)として成形ベル
トの運動方向に平行に2つのロール間のトラフ様の空所へ供給される。成形ベル
トおよび生成物の組成物は更にカレンダーロール間の次第に狭くなる領域内を通
過せしめられて成形ベルト
内の凹所内へ強制的に入れられる。更にロール間の間隔は生成物の組成物に作用
せしめられる圧力を、したがってこうして製造される成形物の厚さを規定する。
成形ベルトの使用は錠剤製造におけるかなり大きな柔軟性に導く。今や種々の
形状およびサイズの錠剤を、装置で単に成形ベルトを交換するだけで製造するこ
とができ、他方で異なる錠剤サイズに同一のプレッシャーロールを使用すること
ができる。更に装置は低コストで製造することができる、なぜならば今や錠剤に
つき凹所は1つ必要であるにすぎないので成形ベルト内の凹所を高精度で形成し
なくてもよいからである。互いに逆方向に可動である構成部材の綿密な調整は不
要である。
プラスティック製の成形ベルトは特に経済的であり、ベルトに成形によって凹
所を形成することが可能である。もちろんこの場合にはこのプラスティックが錠
剤製造中に生じる温度で寸法的に安定であるように注意する必要がある。
アパーチャーは有利に打ち抜きによって形成され、したがって使用される打ち
抜き工具に応じて任意の錠剤形状を製造することが可能である。この実施例では
錠剤製造の間に型内へ入れられた生成物の組成物は1対のカレンダーロールの両
外面と接触し、したがってこの場合には製造される錠剤は2つの平らな表面を持
つ。
成形ベルト内の凹所またはアパーチャーが、生成物の組成物が最小の損失で生
成物の成形物(product moldings)へ変換され得るように形成されていることは
自明である。
有利に使用される凹所またはアパーチャーは上から見て円形、楕円形、6角形
の形状を持つものである。
アパーチャーはまた例えば周面にバーを持っていてよく、バーはパンチの形状
における適当なノッチによって形成され、刻みを持つ錠剤を製造することができ
る。
成形ベルトは任意の所望の材料から製作することができるが、有利には金属、
特に鋼、プラスティックまたはゴムから製作される。比較的剛性の材料から成る
成形ベルトが使用される場合にはベルトの厚さは錠剤の所望の厚さにほぼ相当す
る。したがってより厚い錠剤形状はより厚いベルトを要求し、適当な場合にはベ
ルトは装置の変向ローラの周りを容易に通過することができるように関節で結合
されたベルトの形状であってよい。しかし撓み性の材料から成る成形ベルトの使
用も可能であり、この場合にはベルトは錠剤の所望の厚さよりも厚くすることが
できる。この場合には成形ベルトは特にカレンダーロール間のスリットよりも厚
い。ロール間へ供給されると、成形ベルトが次第に圧縮され、その結果カレンダ
ーロール間の距離に依存して異なる圧力を錠剤成形物に及ぼすことができる。
カレンダーロールは有利に滑らかな外面を持つ。しかし増大された表面領域を
持つ錠剤成形物が要求される場合には例えば表面が一定の粗さを持つカレンダー
ロールも考えられる。
温度制御可能なカレンダーロールが有利に使用される。これは、高温で可塑性
であり、冷時に固体となる生成物の組成物が使用される場合には冷却されたカレ
ンダーロールを使用することが有利であるからである。それに対して湿潤状態で
可塑性であり、乾燥状態で固体となる生成物の組成物も公知であり、この場合に
は加熱可能なロールが有利に使用される。したがってロールは生成物の組成物を
冷却または乾燥するために使用される。生成物の組成物はこれらのロールとロー
ルの直径に依存する異なる時間長さで熱接触する。プレッシャーロールは、有利
に熱伝導性で、圧縮力に対して安定で、かつ使用される物質に不活性である全目
的工具である。ロール間の距離は調整可能であり、異なる接触圧力を生ぜしめる
ことができる。
簡単な構成では装置は唯一の生成物の組成物用の装入手段を備える。しかし例
えば多層錠剤の製造が望まれよう、したがって他の構成は複数の生成物リボンを
製造する、複数の装入手段を持っていてよい。これらの生成物リボンは同時に1
対のロールへ供給することができる。しかし成形ベルトをして複数のロール対を
通過させることも可能であり、この場合には異なる生
成物リボンは個々のロール対内へ順次供給される。この場合のロール対間の距離
は有利には接触圧が第1のロール対では最低であり、かつ最後のロール対では最
高となるように調整されよう。こうしてこの方法で製造される錠剤は多層錠剤と
なる。
しかし錠剤を薄い保護皮膜、例えば拡散抵抗性のポリマーで被覆して活性成分
の特定の放出を達成することも考えられる。特定の環境条件下でのみ(例えばp
Hまたは温度に関し)溶けて、活性成分を放出する保護皮膜を製造することが可
能である。被覆はまた味覚マスクまたは光安定剤としての働きもする。完成錠剤
を保護皮膜で被覆すべき場合には、一般に別個の引き続く工程で公知の被覆方法
を用いて実施される。しかし、本発明によれば1枚または2枚のウエブをメルト
と一緒にメルトが2枚の皮膜ウエブ間に位置するようにロール装置内へ供給する
ことも可能である。これによれば上下面または上下のいずれかの面に適切な保護
皮膜を設けられた錠剤が得られる。上下面上の保護皮膜は同一である必要はない
。多層錠剤の場合には例えば保護皮膜は活性成分の種々の放出を与える。
押出し機およびまたは加熱可能な充填くさびは使用される優れた装入手段であ
る。
本発明による装置は錠剤またはペレット様の成形物を連続的に製造するために
使用することができる。装置はきわめて全目的型である、それというのも基本的
なロール装置は変更せずに残し、成形ベルトのみを交換すればよいからである。
製造できる生成物の範囲は使用される成形ベルトおよび凹所またはアパーチャー
の寸法に依存して極小のペレット様生成物から1g以上の重量を持つ錠剤にまで
及ぶ。成形物は90度以下の縁角度(edge angles)を持ち、これは特に脆い生
成物の組成物を持った錠剤のばり取りを助ける。成形ベルト内のアパーチャーは
有利には製造中に錠剤がつながることがないような相互距離を持つ。したがって
引き続く錠剤の個別化は不要である。
プレッシャーロール駆動装置との摩擦連結は別個の成形ベルトの駆動装置が不
要であることを意味する。成形ベルトの長さは広範囲内で変更することができる
。これは冷却されていない(未硬化の)状態の錠剤の取り出しが回避されるのに
必要なだけ型内での錠剤の滞在時間を延長することを可能にするので、特に有利
であると証明された。
カレンダーロールを出た後の成形ベルトからの生成物の取り出しは適当な場合
にはスパイクロールの使用を介してローラーの変向を通じて簡単な方法で可能で
ある。このことは特に貫通アパーチャーを持つ成形ベルトの場合に該当する、そ
れというのもこの場合には錠剤は上下両側からアクセス可能であり、容易にベル
トから押し出すことができるからである。
成形ベルトを用いて得られる扁平な成形物は錠剤と
して使用するのに特に好適である。この関係で特に、IR(瞬間放出)錠剤、す
なわち活性成分を迅速に放出する錠剤、SR(緩慢放出)錠剤、すなわち活性成
分をゆっくりと放出する錠剤、遅延作用錠剤、すなわち活性成分を遅れて放出す
る錠剤、活性成分が口腔粘膜から吸収され、したがって“舌下”で溶けるように
目された舌下錠剤、香錠または可溶錠剤が好適である。
このようにして製造された錠剤はもちろん薬品の分野または食品分野(例えば
ビタミン錠の形)で使用することができるが、農作物保護および工業分野での適
用も考えられる。
本発明はまた活性成分を含む生成物の組成物から扁平な成形物を製造するため
の方法にも関し、この方法においては、凹所を設けられた成形ベルトを1対のカ
レンダーロールの互いに逆に回転するロール間を通過させ、活性成分を含む組成
物から成る少なくとも1つの生成物ストランドを有利に広幅リボンとして形成し
、この生成物ストランドをロール対へ供給し、生成物の組成物をしてロール対の
ロール間へ供給された時に凹所内へ入り込ませ、かつロール対通過後に成形物を
凹所から取り除く。生成物の組成物圧縮時の所望の接触圧力は2つのロール間の
距離を調整することによって予め決められる。ロール間の距離、すなわちこれら
のロールの外面間の最小距離は有利には使用される成
形ベルトの厚みよりも大きくてはならない。特に撓み性である成形ベルトの場合
には、ロール間のスリットをベルトの厚さよりも明らかに狭くなるように選択す
ることが可能であり、その結果生成物の組成物に対してより高い圧力が及ぼされ
る。
本発明による方法の別の構成では、生成物リボンは複数の連続するロール対を
通過せしめられる。複数の生成物ストランドを形成することも可能であり、これ
らは1つのロール対へ同時にまたは複数のロール対へ順次供給される。この構成
の変更形として、後の錠剤の所望の順序の層をメルトにおいても持つ多層メルト
を同時押し出しによって製造することができる。
生成物の組成物は所望の錠剤に依存して熱時に、または湿潤時に可塑性である
。製造のモードに依存してロール対は固体の寸法安定な錠剤を得るために加熱さ
れるかまたは冷却されよう。
錠剤は、1つ以上の製薬学的活性成分および1つ以上の常用の補助物質を含み
、かつ少なくとも1成分の溶融または軟化のためにペースト状または粘稠になり
、したがって押し出し可能である混合物から製造される。
混合物は、特に製薬学的に認容性のポリマー(混合物のガラス転移温度は混合
物内の全成分の分解温度を下回る)、例えばポリビニルピロリドン(PVP)、
N−ブニルピロリドン(NVP)と酢酸ビニルのコポ
リマー、酢酸ビニルとクロトン酸のコポリマー、部分加水分解されたポリ酢酸ビ
ニル、ポリビニルアルコール、エチレン/酢酸ビニルコポリマー、ポリヒドロキ
シエチルメタクリレート、メチルメタクリレートとアクリル酸のコポリマー、セ
ルロースエステル、セルロースエーテル、ポリエチレングリコールまたはポリエ
チレンを含む混合物である。ポリマーのK値(フィケンチャー(H.Fikentscher
)による。セルロース化学(Cellulose-Chemie)13(1932)、58−64
および71−74)は10〜100、有利に12〜70、特に12〜35の範囲
、かつPVPについては有利に12〜35、特に12〜17の範囲内である。
重合結合材は、組成物が押し出し可能であるには50〜180、有利に60〜
130℃の範囲で全成分の完全な混合物中で軟化または溶融しなければならない
。そのため混合物のガラス転移温度は180、有利に130℃を下回らなければ
ならない。重合結合材は必要により、常用の製薬学的に認容性の可塑化補助物質
によって低減せしめられる。可塑化補助物質は例えば長鎖アルコール、エチレン
グリコール、プロピレングリコール、トリメチロールプロパン、トリエチレング
リコール、ブタンジオール、ペンタノール、ヘキサノール、ポリエチレングリコ
ール、シリコーン、芳香族カルボン酸エステル(例えばジアルキルフタレート、
トリメリット酸エステル、安息香酸エステル、テレフ
タル酸エステル)、または脂肪族ジカルボン酸エステル(例えばジアルキルアジ
ペート、セバシン酸エステル、アゼライン酸エステル、クエン酸エステルおよび
酒石酸エステル)または脂肪酸エステルである。
常用の製薬学的な補助物質(総量がポリマーの100重量%までであってよい
)の例は、特に混合物の総重量の0.02〜50、有利に0.20〜20%の濃
度で、エキステンダーまたは増量剤、例えば珪酸塩または珪藻土、酸化マグネシ
ウム、酸化アルミニウム、酸化チタン、ステアリン酸およびその塩、例えばマグ
ネシウムまたはカルシウム塩、メチルセルロース、ナトリウムカルボキシメチル
セルロース、タルク、しょ糖、乳糖、穀物澱粉またはとうもろこし澱粉、じゃが
いも粉、ポリビニルアルコール;
混合物の総重量の0.1〜5、有利に0.1〜3%の濃度で、滑剤、例えばステ
アリン酸アルミニウムおよびカルシウム、タルクおよびシリコーン;
染料、例えばアゾ染料、天然産の有機または無機の顔料または染料であり、無機
顔料は混合物の総重量の0.001〜10、有利に0.5〜3%の濃度で存在す
る。
流動助剤、例えば動物または植物脂肪、特に水素化形のものおよび室温で固体
であるもの。これらの脂肪は有利に50℃以上の融点を持つ。C12、C14、C16
およびC18脂肪酸のトリグリセリドが優れて
いる。またろう、例えばカルナウバろうを使用することも可能である。これらの
脂肪およびろうは有利に個々にまたはモノ−およびまたはジグリセリドまたはホ
スファチド、特にレシチンと一緒に添加混合することができる。モノ−およびジ
グリセリドは有利に上記の脂肪酸タイプから誘導される。脂肪、ろう、モノ−お
よびジグリセリドおよびまたはレシチンの総量は、各層に関して組成物の総重量
の0.1〜30、有利に0.1〜5%である;
安定剤、例えば酸化防止剤、光安定剤、ヒドロペルオキシド分解剤、ラジカル掃
去剤および微生物の攻撃に対する安定剤。
更に湿潤剤、防腐剤、崩壊剤、吸着剤、離型剤および噴射剤を添加することが
可能である(例えばズッカー (H.Sucker)他著“ファーマツォイティッシェ・
テヒノロギー(Pharmazeutische Technologie)”ティーメ出版社(Thieme-Verl
ag)。シュトッツガルト1978参照)。
補助物質はまた本発明の目的のためには製薬学的活性成分と一緒に固溶体を製
造するための物質を意味する。これらの補助物質の例はペンタエリトリトールお
よびペンタエリトリトールテトラアセテート、ポリマー、例えばポリエチレンオ
キシドおよびポリプロピレンオキシドおよびこれらのブロックコポリマー(ポロ
キサマー(poloxamers)、ホスファチド、例えばレシ
チン、ビニルピロリドンのホモ−およびコポリマー、界面活性剤、例えばポリオ
キシエチレン40ステアレート、およびクエンおよび琥珀酸、胆汁酸、ステロー
ルおよび他の、“Pharm.Acta Helv.” (フォード(J.L.Ford)著。61(1
986)69〜88)に示されているような物質である。
活性成分の溶解性を制御するための塩基および酸の添加も製薬学的な補助物質
として考えられる(例えばトーマ(K.Thoma)他著“Pharm.Ind.”51(198
9)98〜101参照)。
補助物質の適性に関する唯一の前提は十分な熱的な安定性である。
本発明の目的のための製薬学的活性成分は、処理条件下で分解しさえしなけれ
ば製薬学的作用および最小限の副作用を有する全ての物質を意味する。一服用単
位あたりの活性成分の量および濃度は、効能および放出速度に依存している幅広
い範囲内で変化しうる。唯一の条件は、これらが要求される効果を達成するため
に十分であることである。こうして、活性成分濃度は、0.1〜95重量%、有
利に20〜80重量%、殊に30〜70重量%の範囲内にあることができる。ま
た、活性成分の組合せ、例えばイブプロフェン/カフェインを使用することも可
能である。また本発明の目的のための活性成分は、ビタミンおよびミネラル、作
物処理剤および殺虫剤である。ビタミンは、ビタミン
A群、B群(これはB1、B2、B6およびB12ならびにニコチン酸およびニ
コチンアミドに加えて、ビタミンBの性質を有する化合物、例えばアデニン、コ
リン、パントテン酸、ビオチン、アデニル酸、葉酸、オロト酸、パンガム(panga
mic)酸、カルニチン、p−アミノ安息香酸、ミオイノシトールおよびリポ酸を意
味する)、ビタミンC、ビタミンD群、E群、F群、H群、IおよびJ群、K群
ならびにP群を包含する。また、本発明のための活性成分は、治療学のペプチド
を包含する。
本発明による方法は、例えば次の活性成分を処理するのに適当である:
アセブトロール、アセチルシステイン、アセチルサリチル酸、アシクロビア、
