Claims (1)
- 【特許請求の範囲】 1.相同組替えによる形質転換DNAの該プラスチドゲノム中への挿入を行う 標的セグメント、選択可能表現型を該形質転換プラスチドを含有する植物細胞へ 与える選択マーカー遺伝子、および目的とする外来遺伝子をコードする付加的な 発現可能DNAの挿入のためのクローニング部位を有する多細胞植物のプラスチ ドを安定に形質転換するためのDNA構築物において、核コードプラスチドRN Aポリメラーゼによって認識および転写される5’プロモーターエレメントを特 徴とする改良されたDNA構築物。 2.構築物がプラスチドの形質転換に適したベクター中へ組み込まれる請求項 1記載のDNA構築物。 3.該5’プロモーターエレメントがプラスチドコードプラスチドRNAポリ メラーゼによって認識および転写される請求項1記載のDNA構築物。 4.該プロモーターエレメントがPrrn-62、PORF2280-1577、PatpB-289、PORF1 901-41、Prbs2-152、Prps16-107、PatpI-207、Pclp-511、Pclp-173、Pclp-53お よびPaccD-129よりなる群から選択されるプラスチド遺伝子のプロモーターエレ メントから選択される請求項1記載のDNA構築物。 5.該プロモーターエレメントがPclp-95である請求項2記載のDNA構築物 。 6.植物細胞のプラスチドを安定に形質転換するための、および少なくとも1 個の付加的遺伝子産物をそこに発現するためのDNA構築物であって: a)形質転換するためのプラスチドを有する予め決定されたプラスチドゲノム 配列に対して実質上相同なDNA配列を含む標的セグメントであって、予め決定 されたプラスチドゲノム配列との相同組替えが可能な標的セグメント;および b)非致死選択可能表現型を、該DNA構築物を有するプラスチドを含有する 細胞へ与える、該標的セグメント内に配置された選択マーカー遺伝子;および c)タンパク質またはその前駆体をコードする遺伝子の転写単位を含む、付加 的なDNAセグメント;および d)該転写単位への結合が可能なプロモーター配列、核コードポリメラーゼに よって認識されている該プロモーター配列、該プロモーターによって調節されて いる該タンパク質をコードする該遺伝子 を含むDNA構築物。 7.該転写単位が選択マーカー遺伝子をコードする請求項6記載のDNA構築 物。 8.該選択マーカー遺伝子が核コードプラスチドポリメラーゼによって転写さ れたプロモーターによって調節される請求項7記載のDNA構築物。 9.該転写単位がレポーター遺伝子をコードする請求項6記載のDNA構築物 。 10.該構築物がプラスチドの形質転換に適したベクター中へ組み込まれる請 求項6記載のDNA構築物。 11.請求項1記載のDNA構築物を用いて安定に形質転換された多細胞植物 。 12.請求項2記載のDNA構築物を用いて安定に形質転換された多細胞植物 。 13.プラスチド細胞が少なくとも1個の目的とする外来遺伝子によって安定 に形質転換された、植物細胞または多細胞植物を得る方法であって: a)形質転換するためのプラスチドを有する予め決定されたプラスチドゲノム 配列に対して実質上相同なDNA配列を含む標的セグメントであって、予め決定 されたプラスチドゲノム配列との相同組替えが可能な標的セグメント;および b)選択可能プラスチド表現型を、該DNA構築物を有するプラスチドを含有 する細胞へ与える、該標的セグメント内に配置された選択マーカー遺伝子;およ び c)核コードプラスチドRNAポリメラーゼによって認識されるプロモーター により調節されている目的とする外来遺伝子 を含むDNA構築物を植物細胞へ投与すること; d)該表現型を発現する細胞を選択すること;および e)安定に形質転換されたプラスチドを含有する該細胞から植物を再生するこ とを特徴とする方法。
