Claims (1)
- 【特許請求の範囲】 1.(a)配列番号:1のヌクレオチド490もしくは634から1011ま でのヌクレオチド; (b)配列番号:3のヌクレオチド813もしくは1002から1376まで ;および (c)厳密な条件下で(a)または(b)にハイブリダイゼーションし、軟骨 および/または骨および/または他の結合組織の形成能を示す蛋白をコードして いる配列 からなる群より選択されるDNA配列を含む、BMP−15関連蛋白をコードし ている単離DNA配列。 2.(a)配列番号:2のアミノ酸−48もしくは1から125までをコード しているヌクレオチド; (b)配列番号:4のアミノ酸−63もしくは1から125までをコードして いるヌクレオチド;および (c)厳密な条件下で(a)または(b)にハイブリダイゼーションし、軟骨 および/または骨および/または他の結合組織の形成能を示す蛋白をコードして いる配列 からなる群より選択されるDNA配列を含む、BMP−15蛋白をコードしてい る単離DNA配列。 3.発現制御配列に作動するように結合している請求項1のDNA分子を含む ベクター。 4.発現制御配列に作動するように結合している請求項2のDNA分子を含む ベクター。 5.請求項3のベクターで形質転換された宿主細胞。 6.請求項4のベクターで形質転換された宿主細胞。 7.軟骨および/または骨および/または他の結合組織の形成誘導能により特 徴づけられる蛋白をコードしている配列を有し、 (a)配列番号:1のヌクレオチド634から1008まで;および (b)配列番号:3のヌクレオチド1002から1376まで;および (c)(a)または(b)の天然に存在する対立遺伝子配列および等価な縮重 コドン配列 からなる群より選択されるDNA配列を含む単離DNA分子。 8.発現制御配列に作動するように結合している請求項7のDNA分子を含む ベクター。 9.請求項8のベクターで形質転換された宿主細胞。 10.BMP−15蛋白をコードしており、配列番号:3のヌクレオチド10 02から1376を含むDNA分子。 11.読み枠を合わせてDNAコーディング配列に連結している5'側の適当 なプロペプチドをコードしているヌクレオチド配列をさらに含む、請求項10記 載の単離DNA分子。 12.発現制御配列に作動するように結合している請求項11のDNA分子を 含むベクター。 13.請求項12のベクターで形質転換された宿主細胞。 14.下記工程: (a)BMP−15関連蛋白をコードしているヌクレオチド配列を含む請求項 1記載のDNA配列で形質転換された宿主細胞を培養すること;次いで (b)培地から該BMP−15関連蛋白を回収し精製すること を特徴とする、精製BMP−15関連蛋白の製造方法。 15.下記工程: (a)BMP−15関連蛋白をコードしているヌクレオチド配列を含む請求項 2記載のDNA配列で形質転換された宿主細胞を培養すること;次いで (b)培地から該BMP−15関連蛋白を回収し精製すること を特徴とする、精製BMP−15関連蛋白の製造方法。 16.下記工程: (a)BMP−15関連蛋白をコードしているヌクレオチド配列を含む請求項 7記載のDNA配列で形質転換された宿主細胞を培養すること;次いで (b)培地から該BMP−15関連蛋白を回収し精製すること を特徴とする、精製BMP−15関連蛋白の製造方法。 17.配列番号:1のDNA配列によりコードされるアミノ酸および配列番号 :3のDNA配列によりコードされるアミノ酸からなる群より選択されるアミノ 酸配列を含む精製BMP−15関連ポリペプチド。 18.ポリペプチドがダイマーであり、各サブユニットが配列番号:2のアミ ノ酸1から125までのアミノ酸配列および配列番号:4のアミノ酸1から12 5までのアミノ酸配列から選択されるアミノ酸配列を含むものである請求項17 の精製BMP−15ポリペプチド。 19.ポリペプチドがダイマーであり、1のサブユニットが配列番号:2のア ミノ酸1から125までおよび配列番号:4のアミノ酸1から125までからな る群より選択されるアミノ酸配列を含み、もう1つのサブユニットがBMP−1 、BMP−2、BMP−3、BMP−4、BMP−5、BMP−6、BMP−7 、BMP−8、BMP−9、BMP−10、BMP−11、BMP−12および BMP−13からなる群より選択される骨形態形成蛋白に関するアミノ酸配列を 含むものである請求項17の精製BMP−15ポリペプチド。 20.下記工程: (a)配列番号:3に示すヌクレオチド1014から1388までのヌクレオ チド配列を含むDNAで形質転換された細胞を培養すること;次いで (b)配列番号:4に示すアミノ酸1からアミノ酸125までのアミノ酸配列 を含む蛋白を培地から回収し精製すること により製造される精製BMP−15蛋白。 21.治療量の請求項17記載の少なくとも1種のBMP−15関連蛋白を含 んでなる組成物。 22.該組成物を支持し、骨および/または軟骨および/または他の結合組織 の成長のための表面を提供するマトリックスをさらに含んでなる請求項21の組 成物。 23.該マトリックスがヒドロキシアパタイト、コラーゲン、ポリ乳酸および リン酸三石灰からなる群より選択される材料を含むものである請求項22の組成 物。 24.骨および/または軟骨および/または他の結合組織の形成の誘導が必要 な患者に有効量の請求項21の組成物を投与することを特徴とする、骨および/ または軟骨および/または他の結合組織の形成の誘導方法。 25.BMP−15関連ポリペプチドをコードしているDNA配列に正しい読 み枠で結合しているTGF−βスーパーファミリーのメンバー由来のプロペプチ ドをコードしているDNA配列を含むキメラDNA分子。 26.配列番号:4のアミノ酸1から125までのアミノ酸配列を含む精製B MP−15蛋白。 27.配列番号:2のアミノ酸1から125までのアミノ酸配列を含む精製B MP−15関連蛋白。 28.モノマー形態で約10〜17kdの分子量を有し、配列番号:10のア ミノ酸配列を含み、ローゼン−改変サムパス−レディアッセイにおいて軟骨およ び/または骨および/または他の結合組織の形成を誘導する能力を有する精製B MP−15関連蛋白。 29.請求項17記載の精製BMP−15関連蛋白に対する抗体。
