Description
Claims (1)
- 【特許請求の範囲】 1.a.結合化合物をペプチド基質に接合させて修正ペプチド基質を形成し、 b.充分な量の修正ペプチド基質を選択されたタンパクキナーゼを含有する溶 液に添加し、 c.タンパクキナーゼが修正ペプチド生成物を形成するのに十分な時間活性で ある条件下でタンパクキナーゼを修正ペプチド基質と共に培養し、そして d.タンパクキナーゼの活性を測定する、 ことを含む選択されたタンパクキナーゼ活性の存在を測定する方法。 2.c段階の後に高い結合化合物親和性を有するマトリックスを用いて修正ペ プチド生成物を結合させることを更に含む特許請求の範囲1に記載の方法。 3.マトリックスがタンパク質を包含することを特徴とする特許請求の範囲2 に記載の方法。 4.タンパク質が実質的にアビジン及びストレプトアビジンよりなる群から選 択される特許請求の範囲3に記載の方法。 5.タンパクキナーゼ活性の測定に先立って、結合した修正ペプチド生成物を 洗浄することを更に含む特許請求の範囲2に記載の方法。 6.結合化合物がビオチンを包含する特許請求の範囲2に記載の方法。 7.ペプチド基質がビオチン化されている特許請求の範囲6に記 載の方法。 8.修正ペプチド基質及び修正ペプチド生成物が酵素活性を測定するためにビ オチン結合マトリックスに結合される特許請求の範囲6に記載の方法。 9.マトリックスがタンパク質被覆ディスクを包含する特許請求の範囲2に記 載の方法。 10.マトリックスがタンパク質被覆ビーズを包含する特許請求の範囲2に記載 の方法。 11.修正ペプチド基質及び標識の存在下でタンパクキナーゼを培養する特許請 求の範囲1に記載の方法。 12.標識が放射性標識である特許請求の範囲11に記載の方法。 13.放射性標識がγ32Pである特許請求の範囲12に記載の方法。 14.修正ペプチド生成物の量がシンチレーションスペクトロメータによって測 定される特許請求の範囲11に記載の方法。 15.修正ペプチド基質が、プロメガペプチドA(配列ID.1)、プロメガペ プチドB(配列ID.2)、プロメガペプチドC(配列ID.3)、プロメガペ プチドD(配列ID.4)、プロメガペプチドE(配列ID.5)、プロメガペ プチドF(配列ID.6)、プロメガペプチドG(配列ID.7)、プロメガペ プチドH(配列ID.8)並びにそれらの類似物及び組合せよりなる群から選択 される特許請求の範囲1に記載の方法。 16.タンパクキナーゼがセリン−トレオニンキナーゼ及びチロシンキナーゼよ りなる群から選択される特許請求の範囲1に記載の方法。 17.a.タンパクキナーゼに対して特異的反応性を有し定量化を可能ならしめ るために化学反応によって修正された修正ペプチド基質を包含する容器、と b.使用説明書、 とを含む選択されたタンパクキナーゼの存在又は活性を測定するためのキット。 18.修正ペプチド基質が結合化合物を含む特許請求の範囲17に記載のキット 。 19.結合化合物がビオチンである特許請求の範囲18に記載のキット。 20.タンパクキナーゼと相溶性である少なくとも1種の緩衝液を包含する容器 を更に含む特許請求の範囲17に記載のキット。 21.ビオチン結合マトリックスを包含する容器を含む特許請求の範囲17に記 載のキット。 22.修正ペプチド基質が、ビオチン化プロメガペプチドA(配列ID.1)、 ビオチン化プロメガペプチドB(配列ID.2)、ビオチン化プロメガペプチド C(配列ID.3)、ビオチン化プロメガペプチドD(配列ID.4)、ビオチ ン化プロメガペプチドE(配列ID.5)、ビオチン化プロメガペプチドF(配 列ID. 6)、ビオチン化プロメガペプチドG(配列ID.7)、ビオチン化プロメガペ プチドH(配列ID.8)並びにそれらの類似物及び組合せよりなる群から選択 される特許請求の範囲17に記載のキット。 23.タンパクキナーゼがセリン−トレオニンキナーゼ及びチロシンキナーゼよ りなる群から選択される特許請求の範囲17に記載のキット。 24.a.ビオチン化プロメガペプチドG(配列ID.7)並びにそれらの類似 物及び組合せよりなる群から選択される修正ペプチド基質を包含する容器、 b.ビオチン結合マトリックスを含有する容器、及び c.使用説明書、 を含むチロシンキナーゼの存在又は活性を測定するためのキット。 25.a.ビオチン化プロメガペプチドA(配列ID.1)、ビオチン化プロメ ガペプチドB(配列ID.2)、ビオチン化プロメガペプチドC(配列ID.3 )、ビオチン化プロメガペプチドD(配列ID.4)、ビオチン化プロメガペプ チドE(配列ID.5)、ビオチン化プロメガペプチドF(配列ID.6)、ビ オチン化プロメガペプチドG(配列ID.7)、ビオチン化プロメガペプチドH (配列ID.8)並びにそれらの類似物及び組合せよりなる群から選択される修 正ペプチド基質を有する容器、 b.ビオチン結合マトリックスを有する容器、及び c.使用説明書、 を含むセリン−トレオニンキナーゼの存在又は活性を測定するためのキット。
