Claims (1)
- 【特許請求の範囲】 1. 抗体の全てまたは1部をコードする外因性ヌクレオチド配列を含む遺伝 子修飾細胞の移植組織片であって、上記外因性ヌクレオチド配列がその発現およ び上記抗体の分泌に必要な諸要素の制御下に置かれている移植組織片。 2. 上記抗体が: − 天然の抗体、 − キメラ抗体 − 抗体フラグメント、特にフラグントFab、F(ab’)2、Fcまたはs cFv、および −二特異性抗体 からなる群から選択されることを特徴とする、請求項1に記載の移植組織片。 3. 上記抗体が毒性または免疫強化物質により修飾されることを特徴とする 、請求項1または2に記載の移植組織片。 4. 上記抗体がリボヌクレアーゼ、特にBacillus amyloli quefaciensからのリボヌクレアーゼ、リシン、ジフテリヤ毒素、コレ ラ毒素、単純ヘルペスウイルスチミジンキナーゼ、Escherichia c oliまたはSaccharomyces属酵母からのシトシンデアミナーゼ、 Pseudomonasからの外毒素およびヒトアンギオゲニン、または上記物 質の類似体から選択された毒性物質により修飾されていてもよいことを特徴とす る、請求項3による移植組織片。 5. プラスミドから、レトロウイルスからもしくはヘルペスウイルスから、 アデノウイルスから、発現および上記抗体の分泌に必要な諸要素の制御下に置か れた上記外因性ヌクレオチド配列を含むアデノウイルス−会合ウイルスから誘導 されたベクターのトランスフエクションにより細胞が遺伝子的に修飾されること を特徴とする、請求項1ないし4のいずれか1項記載の移植組織片。 6. 上記ベクターがジシストロンであることを特徴とする、請求項5に記載 の移植組織片。 7. 上記ベクターがレトロウイルスであり、次の5’ないし3’: (a)レトロウイルスから誘導された5’LTR、 (b)エンカプシデーション領域、 (c)次の: −内部プロモーター、 −抗体の重鎖をコードする1番目配列、 −リボソームエントリー開始部位、 −抗体の軽鎖をエンコートする2番目配列、 を含む外因性ヌクレオチド配列、および (d)レトロウイルスから誘導される3’LTR を含むことを特徴とする、請求項6に記載の移植組織片。 8. 上記外因性ヌクレオチド配列が、2番目配列の下流に、かつ操作可能な ように融合された毒性または免疫強化物質をコードする3番目配列を追加的に含 むことを特徴とする、請求項7に記載の移植組織片。 9. 遺伝子修飾されたオートローガス細胞を含む、請求項1ないし8のいず れか1項記載の移植組織片。 10. 遺伝子修飾された繊維芽細胞を含む、請求項9に記載の移植組織片。 11. 106ないし1012、好ましくは107ないし1011の遺伝子修飾され た細胞を含むことを特徴とする、請求項1ないし10のいずれか一項記載の移植 組織片。 12. 遺伝子修飾された細胞および細胞外マトリックスを接触させることを 特徴とする、請求項1ないし11のいずれか1項記載の移植組織片の調製方法。 13. 後天性疾患の治療または予防を意図した薬剤組成物の調製のための、 請求項1ないし11のいずれか1項記載の移植組織片の使用。 14. 感染性疾患、および特にAIDS、または癌の治療または予防を意図 した薬剤組成物の調製のための、請求項13に記載の移植組織片の使用。 15. 宿主細胞中に多重体を形成し得る、1種または2種以上の関心事タン パクの全てまたは一部をコードする外因性ヌクレオチド配列を含む組換えアデノ ウイルスベクターであって;上記外因性ヌクレオチド配列がその発現に必要な諸 要素の抑制下に置かれる組換えアデノウイルスベクター。 16. 上記宿主細胞中にダイマーを形成し得る、1種または2種以上の関心 事タンパクの全てまたは1部をコードする外因性ヌクレオチド配列からなる、請 求項15に記載の組換えアデノウイルスベクター。 17. 上記宿主細胞中にテトラマーを形成し得る1種または2種以上の関心 事タンパクの全てまたは1部をコードする外因性ヌクレオチド配列を含む、請求 項15に記載の組換えアデノウイルスベクター。 18. 抗体の全てまたは1部をコードする外因性ヌクレオチド配列を含む組 換えアデノウイルスベクターであって;上記外因性ヌクレオチド配列がその発現 に必要な諸要素の制御下に置かれる組換えアデノウイルスベクター。 19. 上記抗体が天然抗体、キメラ抗体、抗体フラグメントおよび特にフラ グメントF(ab’)2、Fc またはscFvおよび二重特異性抗体からなる群 から選択されることを特徴とする、請求項18に記載の組換えアデノウイルスベ クター。 20. 上記抗体が毒性または免疫強化物質により修飾されることを特徴とす る請求項18または19に記載の組換えアデノウイルスベクター。 21. 上記抗体が、リボヌクレアーゼ、および特にBacillus am yloliquefaciensからのリボヌクレアーゼ、リシン、ジフテ リヤ毒素、コレラ毒素、単純ヘルペスウイルス・チミジンキナーゼ、Esche richia coliからのまたはSaccharomyces属酵母からの シトシンデアミナーゼ、Pseudomonasからの外毒素およびヒトアンギ オゲニンまたは上記物質の類似体から選択された毒性物質により修飾されていて もよいことを特徴とする、請求項20による組換えアデノウイルスベクター。 22. 上記抗体が免疫強化物質により修飾されることを特徴とする、請求項 20に記載の組換えアデノウイルスベクター。 23. ヒト、イヌ、トリ、ウシ、ネズミ、ヒツジ、ブタまたは猿起原のアデ ノウイルスから誘導されたまたは、異なった起原のアデノウイルスゲノムフラグ メントを含むハイブリッドから誘導された、請求項15ないし22のいずれか1 項記載の組換えアデノウイルスベクター。 24. 複製の場合に欠陥であることを特徴とする、請求項15ないし23の いずれか1項記載の組換えアデノウイルスベクター。 25. El領域の全てまたは1部および、任意にE3 領域の全てまたは1部 を少なくとも欠失していることを特徴とする、請求項24に記載の組換えアデノ ウイルスベクター。 26. 2F5 抗体の重鎖、IRES要素および2F5抗体の軽鎖をコードす る外因性ヌクレオチド配列を含んでなり;上記外因性ヌクレオチド配列がその発 現に必要な諸要素の制御下に置かれる、請求項24または25に記載の組換えア デノウイルスベクター。 27. 2F5 抗体重鎖の定状部γ3 領域(ヒンジ領域−CH2 およびCH3 )に操作可能なように融合されたCD4 タンパクのシグナル配列ならびに細胞外 IおよびIIドメインをコードする外因性ヌクレオチド配列を含んでなる、請求 項24または25に記載の組換えアデノウイルスベクター。 28. 2F5 抗体の重鎖の定状部γ3 領域(ヒンジ領域−CH2 およびCH 3)に操作可能なように融合し、かつ成熟ヒト・アンギオゲニンに操作可能なよ うに融合しているCD4 タンパクのシグナル配列ならびに細胞外IおよびIIド メインをコードする外因性ヌクレオチド配列を含んでなる、請求項24または2 5に記載の組換えアデノウイルスベクター。 29. 発現に必要な諸要素が、アデノウイルス初期プロモーターE1A、後 期プロモーターMLP(Major Late Promoter)、ネズミま たはヒトPGK(ホスホグリセレートキナーゼ)プロモーター、SV40ウイル ス初期プロモーター、RSV(ラウス肉種ウイルス)ウイルスプロモーター、腫 瘍細胞中で特に活性なプロモーターおよび最後に感染細胞中で特に活性なプロモ ーターからなる群から選択されたプロモーターを含んでなることを特徴とする、 請求項15ないし28のいずれか1項記載の組換えアデノウイルスベクター。 30. 請求項15ないし29のいずれか1項記載の組換えアデノウイルスベ クターを含んでなる感染性ウイルス粒子。 31. 請求項15ないし29のいずれか1項記載の組換えアデノウイルスベ クターまたは請求項30に記載の感染性ウイルス粒子を含んでなる真核宿主細胞 。 32. 薬学的に許容される担体を伴った、請求項15ないし29のいずれか 1項記載の組換えアデノウイルスベクター、請求項30に記載の感染性ウイルス 粒子または請求項31に記載の真核宿主細胞を含んでなる薬剤組成物。 33. 104ないし1014pfuを含んでなる、請求項32に記載の薬剤組 成物。 34. 注射可能な形態をなすことを特徴とする、請求項32または33に記 載の薬剤組成物。 35. 遺伝子治療によるヒトまたは動物体の治療および/または予防を意図 した薬剤組成物の調製のための、請求項15ないし29のいずれか1項記載の組 み換えアデノウイルスベクター、請求項30に記載の感染性ウイルス粒子、また は請求項31に記載の真核宿主細胞の使用。 36. 後天性疾患ならびに特に癌およびAIDSの治療および/または予防 を意図した薬剤組成物の調製のための、請求項35に記載の使用。 37. 静脈内または腫瘍内ルート経由で投与できる薬剤組成物の調製のため の、請求項36記載の使用。
