新規化合物
本発明は、ある種のIgE介在アレルギー性疾患、T細胞介在自己免疫症状お
よび感染剤に対する不当な免疫応答などの、IL4および/またはIL13の望
ましくない作用に由来の症状を治療するためのヒトインターロイキン4(IL4
)および/またはヒトインターロイキン13(IL13)のアンタゴニストに関
する。
インターロイキンは免疫応答の分泌性ペプチドメディエーターである。公知の
インターロイキンは、各々、免疫系の細胞の発生、活性化、増殖および分化にお
いて多くの効果を有する。IL4はかかる機能において生理学的役割を有するが
、また疾患の発病の一因ともなりうる。特に、IL4は、外因性喘息、鼻炎、ア
レルギー性結膜炎、アトピー性皮膚炎およびアナフィラキシーなどのアレルギー
性疾患の顕著な特徴であるIgE抗体の形成に至るBリンパ球の発生の経路に関
連している。IL4はまた、インスリン依存性糖尿病、多発性硬化症およびリウ
マチ様関節炎などのある種の自己免疫症状における、および移植拒絶反応におけ
る組織損傷の一因となるTリンパ球に対する一般的な増殖および分化のファクタ
ーとしても作用しうる。IL4はさらに、感染症を制御するのに要求される細胞
介在応答の形成を抑制しうる。したがって、IL4のTまたはBリンパ球に対す
る効果の拮抗作用は、かかる疾患に対して有効な作用があると考えられる。IL
13は、最近になって同定されたものであり、レセプター構造/機能の態様(A
versa,G.ら(1993)、J.Exp.Med.178、2213〜2218)を含
め、IL4の生物学的特性と多くの類似性を共有している(Minty,A.ら(19
93)、Nature 362、248〜250)。
ヒトIL4は、2つのN−グリコシル化可能な部位および3個のジスルフィド
結合に関連する6個のシステインを有する129個のアミノ酸のポリペプチド単
鎖からなる(Le,H.V.ら(1988)、J.Biol.Chem.263、10817
−10823)。IL4のアミノ酸配列およびこれらジスルフィド結合の位置は
知られている(Carr,C.ら、(1991)Biochemistry 30、1515−1
523)。
ジスルフィド結合は、残基3と127、24と65、および46と99の間に
ある。IL4の分子量は、グリコシル化度で15kDa(グリコシル化せず)か
ら60kDa(過グリコシル化のIL4)まで変化する。
ヒトIL4についてのDNA配列はまた、Yokota,T.らにより、P.N.A.S
.1986 83 5894−5898に記載されている。
WO93/10235は、IL4アンタゴニストまたは部分アンタゴニストで
ある、IL4のある種の変異株を記載する。
EP−A−0464533は、種々の免疫グロブリン分子の不変領域部と、別
のヒト蛋白質またはその部分とからなる融合蛋白質を開示する。
本発明は、最低1つの免疫グロブリン不変領域またはそのフラグメントに融合
したIL4変異体または変異株からなる、IL4および/またはIL13アンタ
ゴニストまたは部分アンタゴニスト活性を有する可溶性蛋白質を提供するもので
ある。
「変異体または変異株」なる語は、ヒトに内投与した後、IL4および/また
はIL13を拮抗する能力を保持するIL4蛋白質の不完全な誘導体または他の
誘導体などのいずれの分子をも包含する。かかる他の誘導体は、アミノ酸を付加
、欠失、置換または転位することで、またはその化学的修飾で調製することがで
きる。
IL4の変異体または変異株をコードするDNAポリマーは、常法により、例
えば、G.Winterらにより、Nature 1982、299、756−758に、ま
たはZollerおよびSmith 1982;Nucl.Acids Res.、10、6487−6
500に記載されているように、IL4をコードするcDNAの部位定方向性突
然変異、またはChanおよびSmithによりNucl.Acids Res.、1984、12
、2407−2419にて、またはG.WinterらによりBiochem.Soc.Trans.
、1984、12、224−225に記載されているような欠失突然変異、ある
いはMikaelianおよびSergeantによりNucleic Acids Research、1992、
20、376に記載されているようなポリメラーゼ連鎖反応により調製できる。
本明細書にて用いる場合、「IL4および/またはIL13アンタゴニストま
たは部分アンタゴニスト活性を有する」とは、SpitsらによりJ.Immunology
139、1142(1987)にて記載されている検定にて、IL4刺激性T細
胞増殖が用量依存的にて阻害されることを意味する。
120から128までのいずれか1つの位置で、野生型IL4にて天然に存在
する少なくとも1個のアミノ酸が異なる天然アミノ酸と置換されている、適当な
IL4変異体がWO93/10235に開示されている。特に、124位に天然
に存在するチロシンは、グリシン、さらに好ましくはアスパラギン酸などの異な
る天然アミノ酸で置換されていてもよい。
免疫グロブリンはいずれのサブクラス(IgG、IgM、IgA、IgE)であっ
てもよいが、好ましくはIgG1、IgG3またはIgG4のようなIgGである。
該不変領域またはそのフラグメントは重鎖または軽鎖あるいはその両方から誘導
される。本発明は、Fcレセプター結合のような天然免疫グロブリンの望ましく
ない特性を削除する、および/または安定性のような望ましい特性を導入する、
免疫グロブリン成分における突然変異を包含する。例えば、Angal S.、King
D.J.、Bodmer M.W.、Turner A.、Lawson A.D.G.、Roberts G.、P
edley B.およびAdair R.は、Molecular Immunology、vol.30、105−
108頁、1993において、残基241(Kabat番号付け)がセリンからプロ
リンに変わっているIgG4分子を記載する。この変化は、IgG4分子の血清半
減期を増加させる。Canfield S.M.およびMorrison S.L.はJournal of E
xperimental Medicine、 vol.173、1483−1491頁において、IgG
3にてロイシンからグルタメートへの、およびマウスIgG2bにてグルタメー
トからロイシンへの残基248(Kabat番号付け)の変更を記載する。前者にお
けるロイシンのグルタメートとの置換はFcγRIレセプターに関連する免疫グ
ロブリン分子のアフィニティーを減少させ、後者でのグルタメートのロイシンと
の置換はそのアフィニティーを増加させる。EP0307434は、IgGの残
基248(Kabat番号付け)のLからEへの突然変異を包含する種々の突然変異
を開示する。
不変領域またはそのフラグメントは、ヒトIgGの重鎖の不変領域の全体また
は置換部であることが好ましく、最も好ましくはIgG4である。一の態様にお
いて、IgG成分はCH2およびCH3領域ならびに重鎖間のジスルフィド結合
に寄与するシステイン残基を含むIgG1のヒンジ領域、例えばIgG1ヒンジ領
域の残基11と14からなる(Frangione B.およびMilstein C.、Nature、
vol216、939−941頁、1967)。好ましくは、IgG1成分は、Ell
ison J.、Berson B.およびHood L.E.、Nucleic Aids Research、vol1
0、4071−4079頁、1982に記載のIgG1のヒンジの残基1−4お
よび6−15、CH2の1−110およびCH3の1−107に対応するアミノ
酸からなる。ヒンジの残基5は、ヌクレオチド配列のTGTからGCCへの
変更により、公開されたIgG1配列におけるシステインからアラニンへと変わ
っている。別の態様において、IgG成分は、CH2およびCH3領域と、重鎖
間のジスルフィド結合に寄与するシステイン残基を包含するヒンジ領域、例えば
IgG4ヒンジ領域の残基8および11とからなるIgG4より誘導される(Pin
ck J.R.およびMilstein C.、Nature、vol216、941−942頁、19
67)。好ましくは、IgG4成分は、Ellison J.、Buxbaum J.およびHood
L.、DNA、vol1、11−18頁、1981に記載のIgG4のヒンジの残基
1−12、CH2の1−110およびCH3の1−107に対応するアミノ酸か
らなる。IgG4における適当な変異の一例として、ヒンジの残基10(残基2
41、Kabat番号付け)を野生型のセリン(S)からプロリン(P)に変え、C
H2の残基5(残基248、Kabat番号付け)を野生型のロイシン(L)からグ
ルタメート(E)に変える。
IL4変異体または変異株のIg不変領域またはフラグメントへの融合は、一
の成分のC−末端を他の成分のN−末端に融合させることでなされる。好ましく
はIL4変異体または変異株をそのC−末端を介してIg不変領域またはフラグ
メントのN−末端に融合させる。
好ましい態様において、本発明の融合蛋白質のアミノ酸配列は、配列番号:4
、配列番号:7または配列番号:10で表される。
さらなる態様において、本発明は、本発明の化合物の製法であって、組換え宿
主細胞にて該化合物をコードするDNAを発現させ、その産物を回収することか
らなる方法を提供する。
該化合物をコードするヌクレオチド配列からなるDNAポリマーもまた本発明
の一部を形成する。
好ましい態様において、DNAポリマーは、配列番号:3、配列番号:6また
は配列番号:9の配列からなる。
本発明の方法は、Maniatisら、Molecular Cloning−A Laboratory Manu
al:Cold Spring Harbor、1982およびDNA Cloning、第I巻、第II巻
および第III巻(D.M.Glover編、IRI Press Ltd.)に記載されてい
るような慣用的組換え技法により行うことができる。
特に、該方法は:
i)宿主細胞中、本発明の化合物をコードするヌクレオチド配列からなるDNA
ポリマーの発現能を有する複製可能な発現ベクターを調製し;
ii)宿主細胞を該ベクターで形質転換し;
iii)該形質転換された宿主細胞を該DNAポリマーの発現を可能とする条件下
で培養し、該化合物を産生し;および
iv)該化合物を回収する
工程からなる。
本発明はまた、適当なモノ−、ジ−またはオリゴマーヌクレオチドユニットを
融合することでDNAポリマーを調製する方法を提供する。
該調製は、化学的に、酵素学的に、または2つの方法を組み合わせることによ
り、適宜、in vitroまたはin vivoにて行うことができる。かくして、DNAポ
リマーは、D.M.Rovertsら、Biochemistry、1985、24、5090−5
098に記載されているような常法に従って、適当なDNAフラグメントを酵素
的に連結することで調製される。
DNAフラグメントは、必要なヌクレオチド配列を有するDNAを、化学的合
成により、DNAまたはRNA鋳型での酵素的重合により、またはこれらの方法
の組み合わせにより、適当な制限酵素で消化することで得られる。
制限酵素を用いる消化は、20〜70℃の温度で、一般に、50μlまたはそ
れ以下の容量の適当なバッファー中、0.1〜10μgのDNAで行うことがで
きる。
DNAの酵素的重合は、10〜37℃の温度で、一般に50μlまたはそれ以
下の容量の、要すればヌクレオシド三リン酸dATP、dCTP、dGTPおよ
びdTTPを含有する適当なバッファー中、DNAポリメラーゼI(クレノーフ
ラグメント)などのDNAポリメラーゼを用い、in vitroにおいて行うことがで
きる。
DNAフラグメントの酵素的連結は、4℃ないし外界温度で、一般に50μl
またはそれ以下の容量の適当なバッファー中、T4DNAリガーゼなどのDNA
リガーゼを用いて実施することができる。
DNAポリマーまたはフラグメントの化学的合成は、'Chemical and Enzyma
tic Synthesis of Gene Fragments−A Laboratory Manual'(H.G.Gass
enおよびA.Lang編)、Verlag Chemie、Weinheim(1982)または他の科
学刊行物、例えば、M.J.Gait、H.W.D.Matthes、M.Singh、B.S.Spr
oatおよびR.C.Titmas、Nucleic Acids Research、1982、10、62
43;B.S.SproatおよびW.Bannwarth、Tetrahedron Letters、1983

うな固相合成法を用い、通常のホスホトリエステル、亜リン酸またはホスホルア
ミダイト化学により行うことができる。好ましくは、自動式DNA合成装置を用
いる。
DNAポリマーは、好ましくは、一緒になって化合物をコードするDNA配列
を構成する2またはそれ以上のDNA分子を連結することで調製される。本発明
による個々の方法は、そのIL4変異体または変異株をコードする第1のDNA
分子およびその免疫グロブリン領域またはそのフラグメントをコードする第2の
DNA分子を連結することからなる。
DNA分子は、必要なコーディング配列を有するベクターの適当な制限酵素で
消化することにより、またはポリメラーゼ連鎖反応合成法を用いることにより得
られる。
DNA分子の正確な構造およびそれを得る方法は、所望の産物の構造に依存す
る。化合物をコードするDNA分子を構築するための適当な方法を設計すること
は当該分野における当業者にとって慣用的なことである。
組換え宿主細胞にて化合物をコードするDNAポリマーの発現は、宿主細胞中
、DNAポリマーの発現能を有する複製可能な発現ベクターで行ってもよい。該
発現ベクターは新規であり、さらに本発明の一部を形成する。
複製可能な発現ベクターは、宿主細胞と適合するベクターを切断し、無傷のレ
プリコンを有する線状DNAセグメントを得、該線状セグメントをその線状セグ
メントと一緒になって連結反応条件下で化合物をコードする1またはそれ以上の
DNA分子を合することにより、本発明に従って調製することができる。
線状セグメントと1以上のDNA分子の連結は、同時にまたは所望により連続
的に行ってもよい。
すなわち、DNAポリマーは、予め形成されていても、または所望によりベク
ターの構築の間に形成されてもよい。
ベクターの選択は、原核細胞、例えばイー・コリ(E.coli)、または真核細
胞、例えばマウスC127、マウス骨髄腫、チャイニーズハムスター卵巣または
ヒーラ細胞、菌類、例えば糸状菌または単細胞酵母または昆虫細胞、例えばドロ
ソフィラである宿主細胞によりいくらかは決定されるであろう。宿主細胞はまた
形質転換した生物であってもよい。適当なベクターは、プラスミド、バクテリオ
ファージ、コスミドおよび例えば、バキュロウイルス、ワクシニアまたはセムリ
キ森林熱ウイルスに由来の組換えウイルスを包含する。
複製可能な発現バクターの調製は、例えば、Maniatisら、前掲の操作により
DNAの制限、重合および連結についての適当な酵素を用いて慣用的に行われる
。重合および連結は、DNAポリマーの調製について前記したように行うことが
できる。制限酵素を用いる消化は、20〜70℃の温度で、一般に50μlまた
はそれ以下の容量の適当なバッファー中、0.1〜10μgのDNAで行うこと
ができる。
組換え宿主細胞は、本発明によれば、宿主細胞を、形質転換条件下、本発明の
複製可能な発現ベクターで形質転換することにより調製される。適当な形質転換
条件は慣用的なものであり、例えば、Maniatisら、前掲に、または“DNA
Cloning”第II巻、D.M.Glover編、IRL Press Ltd,1985に記載さ
れている。
形質転換条件の選択は宿主細胞により決定される。すなわち、イー・コリのよ
うな細菌宿主をCaCl2の溶液(Cohenら、Proc.Nat.Acad.Sci.、1973
、69、2110)で、またはRbCl、MnCl2、酢酸カリウムおよびグリセロ
ールの混合物からなる溶液で処理し、ついで3−[N−モルホリノ]−プロパン
−スルホン酸、RbClおよびグリセロールで処理してもよい。培養物中の哺乳動
物細胞は、ベクターDNAの細胞上へのカルシウム共沈殿により形質転換されて
もよい。
本発明はまた本発明の複製可能な発現ベクターで形質転換された宿主細胞にま
で及ぶ。
形質転換された宿主細胞の、DNAポリマーの発現を可能とする条件下での培
養は、例えば、前記したManiatisら、“DNA Cloning”に記載されているよ
うに、慣用的に行われる。かくして、好ましくは、細胞に栄養素を供給し、45
℃以下の温度で培養す。
発現産物は宿主細胞に応じた常套手段により回収される。すなわち、宿主細胞
がイー・コリのようなバクテリアであるなら、それを物理的、化学的または酵素
学的に溶菌させ、蛋白質産物を得られた溶菌液より単離してもよい。産物が細菌
細胞より分泌されるものであるならば、それを周辺腔または栄養培地より回収し
てもよい。宿主細胞が哺乳動物細胞であるならば、産物は栄養培地より単離され
るのが一般的である。
DNAポリマーを、産物を発現する安定な形質転換された哺乳動物細胞系を単
離するように設計されたベクター;例えば、ウシパピロマウイルスベクターまた
はチャイニーズハムスター卵巣細胞の増幅ベクターに組み入れてもよい(DNA
Cloning、第II巻、D.M.Glover編、IRL Press 1985;Kaufman,R.
J.ら、Molecular and Cellular Biology 5、1750−1759、198
5;Pavlakis G.N.およびHamer,D.H.、Proceedings of the National
Academy of Science(USA)80、397〜401、1983;Goeddel,
D.V.ら、欧州特許出願第0093619号、1983)。
本発明の化合物はIL4および/またはIL13アンタゴニスト活性を有し、
したがってIL4および/またはIL13の望ましくない作用に由来する症状、
例えばIgE介在アレルギー性疾患およびT細胞介在自己免疫症状または慢性微
生物感染症の治療にて有用であるかもしれない。
したがって、本発明はさらに、本発明の化合物と、医薬上許容される担体とか
らなる医薬組成物を提供する。
使用において、該化合物は、通常、ヒト医薬用担体、希釈体および/または賦
形剤と組み合わせた医薬組成物の形態にて用いられるが、組成物の正確な形態は
投与方法に依存する。化合物は、例えば、吸入用のエアロゾルまたは噴霧溶液ま
たは非経口投与用の滅菌溶液の形態にて用いられる。
本発明の化合物を投与する場合の投与量は、IL4および/またはIL13介
在症状に対して所望の効果を付与し、それによりIgE抗体介在徴候が減少する
か、または自己免疫疾患の進行が停止または後退する量である。その投与量は、
一般に、年齢、医学的症状、たとえあるとしても禁忌の程度または重篤度で変化
する。単位投与量は1mg以下から300mgまで変化しうるが、典型的には用
量当たり1〜20mgの範囲であり、一日の投与量が0.02〜40mg/kg
の範囲にあるように、一日に1回またはそれ以上、例えば1〜6回投与する。
注射に適する組成物は、溶液、懸濁液またはエマルジョン、あるいは使用前に
適当なビヒクルに溶かすかまたは懸濁させる乾燥散剤の形態である。
流体単位投与形は、化合物および発熱物質不含の滅菌ビヒクルを用いて調製さ
れる。用いるビヒクルおよび濃度に応じて、化合物をビヒクルに溶かすかまたは
懸濁させることができる。溶液は非経口投与用のあらゆる形態にて用いることが
でき、とりわけ静脈内感染用に用いられる。溶液の調製においては、化合物をビ
ヒクルに溶かし、必要ならば塩化ナトリウムを添加することで溶液を等張にし、
適当な滅菌バイアルまたはアンプルに充填し、かつ密封する前に無菌技法を用い
、滅菌フィルターを介して濾過することで滅菌処理する。別法として、溶液の安
定性が十分であるならば、密封容器中の溶液をオートクレーブで滅菌処理しても
よ
い。有利には、緩衝化剤、可溶化剤、安定化剤、保存剤または殺菌剤、沈殿防止
剤または乳化剤および/または局所麻酔剤のような添加剤をビヒクルに溶かして
もよい。
使用前に適当なビヒクルに溶かすかまたは懸濁させることができる乾燥散剤は
、予備滅菌処理した薬および他の成分を滅菌分野における無菌技法を用いて滅菌
容器に充填することにより調製してもよい。また、薬および他の成分を水性ビヒ
クルに溶かし、該溶液を濾過により滅菌処理し、滅菌分野における無菌技法を用
いて適当な容器に分配してもよい。ついで、産物を凍結乾燥させ、容器を無菌状
態で密封する。
筋肉内、皮下または皮内注射に適する非経口用懸濁液は、滅菌化合物を滅菌ビ
ヒクルに溶かす代わりに懸濁させ、滅菌処理を濾過で達成することができないこ
とを除いて、実質的に同じ方法で調製する。化合物を滅菌状態で単離するか、ま
たは別法として単離後に、例えば、γ線照射により滅菌処理してもよい。有利に
は、沈殿防止剤、例えば、ポリビニルピロリドンを組成物中に配合し、化合物の
均一な分配を促進する。
気道を介する投与に適する組成物は、吸引用のエアロゾル、噴霧性溶液または
微細状粉末を包含する。後者の場合、50ミクロン未満、特に10ミクロン未満
の粒径が好ましい。そのような組成物は、常法にて製造され、通常の投与装置と
一緒に用いることができる。
さらなる態様において、IL4および/またはIL13の望ましくない作用に
由来する症状の治療法であって、有効量の本発明の化合物を患者に投与すること
からなる方法を提供する。
本発明はさらには活性治療物質として用いるための、特にIL4および/また
はIL13の望ましくない作用に由来する症状を治療するための化合物を提供す
る。
本発明はまたIL4および/またはIL13の望ましくない作用に由来する症
状を治療するための医薬の製造における本発明の化合物の使用を提供する。
本発明の化合物を本発明に従って投与する場合、予期せぬ毒性作用は考えられ
ない。
以下に実施例を用いて本発明を説明する。
実施例1 IL4.Y124D/IgG1融合蛋白質
IL4.Y124D/IgG1キメラcDNAの構築、哺乳動物の発現系におけ
る対応する蛋白質の発現およびその活性を記載する。
1. 融合蛋白質をコードするDNAの構築
(a) IL4.Y124Dコーディング領域の構築
蛋白質の残基124を野生型のチロシンからアスパラギン酸に変異させたと記
載されている(Kruse,N、Tony,H−PおよびSebald,W.EMBO Journal
11:3237[1992])、ヒトIL4遺伝子の変異体を、ヒトIL4c
DNA(British Biotechnologyより購入)をPCR突然変異誘発に付して産
生した。IL4.Y124D cDNAを、HindIIIおよびBg1 II部位を用いて
発現ベクターpTR312に挿入し(M J Browne、J E Carey、C G Ch
apman、A W R Tyrrell、C Entwise、G M P Lawrence、B Reavy、I
Dodd、A Esmail&J H Robinson.Journal of Biochemistry263:1
599、[1988])、プラスミドpDB906を形成した。
IL4.Y124D分子を増幅し、都合のよい制限部位を、各々、サブクロー
ニングの終わりで加えるために、PCR反応を基質として20ngのpDB90
6プラスミドを用いて行った。PCRプライマーを制限酵素部位を有するように
設計し、側面に10−15個のヌクレオチド塩基対を付し、終わりにプライマー
を「アンカー」した。プライマー配列は以下のとおりであった:
プライマーを最終濃度5ng/μlで用い、dNTPを合計100μlの反応
容量中0.2mMの最終濃度で加えた。31サイクルのPCRを行った。サイク
ルは94℃で1分間の変性工程と、50℃で1分30秒間のアニーリング工程と
、72℃で1分30秒間の延長工程とからなる。サイクル1で、変性を5分間に
延ばし、最終サイクルで延長を7分間に延ばした。Advanced Biotechnologies
由来の2.5ユニットのTaqポリメラーゼ酵素をPCR反応に用いた。587b
pのPCR産物を産生した。これをPromega“マジックPCRクリーンアップ(
Magic PCR cleanup)”キットを用いて精製し、ついで反応バッファー4中
、EcoRIおよびKpnI(制限酵素はすべてGibcoBRLより購入した)で消化
し、「付着末端」を生成した。37℃で4時間30分経過した後、反応物を70
℃で10分間加熱し、ついでエタノール沈降に付した。得られたDNAをアガロ
ースゲル電気泳動に付して分析し、約570bp、463bpおよび100bp
の3つのバンドの存在が認められた。消化物は不完全であるため、存在した57
0bpフラグメントはIL4の全長IL4.Y124D変異体を示す。2個のよ
り小さなフラグメントは、IL4.Y124D cDNA中にEcoRI部位がある
ために産生された。570bpバンドをGenecleanTM操作により精製し、EcoR
IおよびKpnIで、つづいてGenecleanTMで消化することで調製されたBluescr
iptKS+TMに連結した。BluescriptKS+/IL4.Y124D組換え体をこう
して形成した。この多量の組換えDNAをPromega“マジック・マキシプレプ(
Magic Maxiprep)”法を用いて産生した。IL4.Y124D挿入体を、Sma
IおよびKpnIを用い、Bluescript組換え体より切除した。20μgの組換え
DNAを、反応バッファー4中、25ユニットのSmaIと一緒に30℃で一夜イ

(b)IgG1コード領域の構築
COSFcLinkベクター(表1)は、Ellison J.、Berson B.およびHood
L.E.、Nucleic Acids Research、第10巻、4071−4079頁、19
82に記載の、ヒンジのアミノ酸1−4および6−15、CH2の1−110お
よびCH3の1−108をコードするヒトIgG1 cDNAを含有する。ヒンジ
の残基5を、ヌクレオチド配列のTGTをGCCに変更することで、公開されて
いるIgG1配列のシステインからアラニンに変える。これを、RT−PCRを
用い、ヒトIgG形質細胞白血病ARH−77(アメルシャム型組織収集(Amer
ican Type Tissue Collection))よりクローンし、完全に配列決定し、公開
されている配列[特許出願公開WO92/00985]との同一性を確認した。
COSFcの構築を前記のヒトIgG1 cDNAを含有するpUC18ベクタ
ー(pUC18−Fc)で始め、それをKpnIおよびSacIIで消化し、CH1、
ヒンジおよびCH2の一部を欠失させた。欠失領域を、以下のヒンジ−CH2領
域を含有するPCR増幅フラグメントで置き換えた。次のPCRプライマーを用
いた:

ヒンジ−CH2領域含有のDNAフラグメントをpUC18-Fcより増幅し、
KpnIおよびSacIIで消化し、ゲル精製し、KpnI/SacII消化pUC18-Fc
ベクター中にクローンした。IgG1重鎖の230位に存する(Kabat番号付け
;Kabatら、“Sequence of Proteins of Immunological Interest”、第5
版、USDepartment of Health and Human Services、NIH出版、No.9

3242(1991))Cysを、ヌクレオチド配列のTGTをGCCに置換する
ことでAlaに変更した。PCRプライマーの一部の変更DNA配列をヒンジの5
'末端の独特KpnI部位に導入した。得られたプラスミドはpUC18Fcmodと
称され、結合およびPCR増幅領域を確認のために配列決定した。
pUC18-Fcmod中の完全なヒンジ−CH2−CH3挿入物を、KpnIおよ
びXbaIで単一のDNAフラグメントとして除去し、ゲル精製し、KpnIおよび
XbaIで切断したSFcR1Cos4に連結し、COSFcを形成した。
SFcR1Cos4はpST4DHFR(Deen,K、McDougal,JS、Inacker
,R、Folena-Wasserman,G、Arthos,J、Rosenberg,J、Maddon,PJ、Ax
el,RおよびSweet,RW、Nature 331:82[1988])の誘導体であり
、サイトメガロウイルス(CMV)プロモーターとウシ成長ホルモン(BGH)
ポリアデニレーション領域の間に挿入される可溶性FcレセプターI型(sFc
R1)を含有し、さらにβ−グロビンプロモーターとSV40ポリアデニレーシ
ョン領域の間に挿入されるジヒドロ葉酸レダクターゼ(DHFR)cDNA、S
V40複製起点および細菌における成長のためのアンピシリン耐性遺伝子を含有
する。ベクターをKpnIおよびXbaIで切断することで、COSFcベクターが
CMVプロモーターとBGHポリA領域の間に挿入されるヒンジ−CH2−CH
3領域を含有するように、sFcR1コード領域が除去される。
このEcoRI部位を再び形成し、またBstEII、PstIおよびEcoRVをクロ
ーンする部位を導入する、ベクターの独特EcoRI部位でオリゴヌクレオチドリ
ンカーを挿入することにより、COSFcLinkベクターをCOSFcより調製し
た。
結合を配列決定し、ベクターにおける配向を確認した。最終ベクターの大きさ
は6.37kbである。
(c)融合蛋白質をコードするDNAの構築
IL4.Y124DcDNAを挿入するために、COSFcLinkベクターをEc
oRVおよびKpnIで以下のように消化することで調製した:5μgのDNAを
反応2にて37℃で5時間、15ユニットのEcoRVと一緒にインキュベートし
、つづいてエタノール沈降に付した。得られたDNAを反応4にて37℃で3時
間、KpnIで消化し、エタノール沈降に付した。IL4.Y124D/SmaI/
KpnIおよびCOSFcLink/EcoRV/KpnIフラグメントを連結し、IL4
.Y124D/IgG1融合蛋白質をコードする、プラスミドpDB951を形成
した。Amersham DNA連結キット、製品コードRPN 1507を用い、反応
物を16℃で一夜インキュベートし、連結を達成した。連結反応産物をPromega
JM能力細胞(高能力)に形質転換し、アンピシリンを50μg/mlで含有
するLuria Broth寒天にプレートした。形質転換物をLuria Broth(アンピシ
リンを50μg/mlで含有)中で培養し、DNAをPromega“マジック・マキ
シプレプ”を用いて調製した。IL4.Y124D/COSFcLink組換えDN
Aの産生を制限消化物およびDNA配列決定により確認した。完全IL4.Y1
24D配列およびCOSFcLinkDNAとの結合をDNA配列決定により確認し
た(表2)。組換えIL4.Y124D/IgG1DNAのコード配列を表3に示
し、融合蛋白質のアミノ酸配列を表4に示す。IL4.Y124D/COSFcL
ink組換えDNAを調製し、塩化セシウム勾配を用いて精製し、そのDNAを用
いてヒーラ細胞を一時的にトランスフェクトした。
2.融合蛋白質の発現
ヒーラ細胞を、10%ウシ胎児血清および1%グルタミンを含むMEMα培地
(Gibco)にて培養した。検定のために、トランスフェクションの4日前、1x
106個のヒーラ細胞を、75cm2のフラスコ中、10%ウシ新生児血清、1%
グルタミンを含む15mlのRPMI−1640培地(「シード培地」)にて接
種した。トランスフェクションの前日に、さらに12.5mlのシード培地を各
フラスコに加えた。トランスフェクションの当日に、培地を、15mlの「トラ
ンスフェクション培地」(10%ウシ新生児血清および1%非必須アミノ酸を有
するエール(Earle's)塩を含むMEM培地)に変えた(時間0)。+3時間の
時に、0.125M CaCl2、1xHBS(HEPES緩衝セイライン)中の適
当なDNA(25μg)をその細胞に加えた。+7時間の時に、細胞をグリセロ
ールショック(15%v/v)に付し、ついで5mMの酪酸ナトリウム含有の1
2.5mlのシード培地中で一夜放置してインキュベートした。その翌日、細胞
をPBS(ダルベッコ(Dulbecco)のリン酸緩衝セイライン)で洗浄し、12.
5mlの「収集培地」(7.5%のストック炭酸水素ナトリウム溶液を2%有す
るRPMI−1640)を加えた。さらに124時間インキュベートした後、上
澄を取り出し、1000rpmで5分間遠心分離に付し、細胞残骸を除去し、4
℃または−20℃のいずれかで貯蔵した。
3.生物学的活性
IL4アンタゴニスト活性について上澄を検定するために:Spitsら、J.I
mmunology 139,1142(1987)に記載の方法を用い、ヒト末梢血液リ
ンパ球をフィトヘマグルチニン、T細胞マイトジェンと一緒に3日間インキュベ
ートし、IL4レセプターをアップレギュレーションさせた。ついで、得られた
芽細胞をIL4でさらに3日間刺激した。増殖を3Hチミジンの組み込みにより
測定した。
IL4.Y124D/IgG1キメラは、用量依存法における133pMのIL
4で刺激されたヒト末梢血液Tリンパ球による3Hチミジンの組み込みを阻害し
た。
実施例2 IL4.Y124D/IgG4融合蛋白質
1.融合蛋白質をコードするDNAの構築
PCRを行って、IL4.Y124Dコード領域を増幅し、XhoI部位を形成
する3'末端でサイレントヌクレオチド置換を導入した。PCR反応のための基
質として、20ngの線状化pDB951プラスミド(実施例1.1(c))を
用いた。用いたオリゴヌクレオチドプライマーを以下のとおりであった:
第2のPCR反応を行って、ヒトIgG4重鎖のヒンジ−CH2−CH3フラ
グメントを増幅した。このための基質は合成ヒトIgG4重鎖cDNAであり、
その配列は表5に記載されており、Genbank配列GB:HUMIGCD2に基づ
いている(Ellison J.、Buxbaum J.およびHood L.E.、DNA 1:11
−18、1981)。多くのサイレント置換を公開されたヌクレオチド配列に対
して行った。2つの0.5kb合成DNAフラグメントを合することで遺伝子を
組み合わせた。各0.5kbのフラグメントは、一連の重複オリゴヌクレオチド
をアニーリングし、ついでギャップをPCRで充填することで調製した。2個の
0.5kbフラグメントをSacII部位で合し、pCR2ベクターに挿入した。完
全な不変領域を含有する1.0kbのApaI-BglIIフラグメントを単離し、ヒト
化IL4特異的可変領域を含有する発現ベクター、pCDに連結した。この構築
物をPCR基質として用い、IgG4のヒンジ−CH2−CH3領域を増幅した
。
IgG4ヒンジ-CH2-CH3領域を増幅するのに用いたオリゴヌクレオチド
プライマーは次のとおりであった:
両方のPCR反応についての条件はpDB951の誘導について記載したとお
りであった。簡単には、プライマーを5ng/μlで用い、dNTPを合計10
0μlの反応容量中0.2mMの最終濃度で加えた。2.5ユニットのAdvanced
BiotechnologiesからのTaqポリメラーゼ酵素を用いて、31サイクルのPCR
反応を行った。サイクルは94℃で1分間の変性工程、50℃で1分30秒間の
アニーリング工程、および72℃で1分30秒間の延長工程からなった。サイク
ル1で変性を5分間に延ばし、最終サイクルで延長を7分に延ばした。
約700bp(IgG4のヒンジ−CH2−CH3)および400bp(IL
4.Y124D)のPCR産物を得、Promega“マジック・PCR・クリーンア
ップ”キットを用いて精製した。ついで、精製したPCR反応物を次の酵素で消
化し、「付着末端」を形成した:IgG4ではXhoIおよびXbaIならびにIL
4.Y124DではEcoRVおよびXhoI。消化物を37℃で3時間インキュベ
ートし、ついでエタノール沈降に付した。得られたDNAをゲル電気泳動法によ
り分析し、約690bp(IgG4のヒンジ−CH2−CH3)および370b
p(IL4.Y124D)の大きさを得た。
pDB951(COSFcLinkにおけるIL4.Y124D)をEcoRVおよ
びXbaIで消化することによりIgG4 PEのヒンジ−CH2−CH3およびI
L4.Y124Dフラグメントを連結させ、大部分のIL4.Y124D/IgG
1融合分子を除去するためのベクターを調製する。わずかに残っている部分はI
L4の5'末端の約75bpであり、それはPCR増幅で産生したIL4.Y12
4D EcoRV/XhoIフラグメントには存在しなかった。5μgのpDB95
1DNAを反応バッファー2(GibcoBRL)を用いて合計30μlの容量にて
消化した。得られた5.8kbのDNAフラグメントをGenecleanTM操作を用い
て精製した。
3種の記載されているフラグメント(IL4.Y124D EcoRV/XhoI、
IgG4 XhoI/XbaIのヒンジ-CH2-CH3およびpDB951のEcoRV
/XbaI消化により得られる5.8kbフラグメント)を一緒に連結して、IL
4.Y124D/IgG4融合蛋白質をコードする、プラスミトpDB952を形
成した。連結反応をAmersham製のDNA連結キット(製品コードRPN150
7)を用い、反応物を16℃で一夜インキュベートすることで行った。連結反応
産物をPromega JM109能力細胞(高能力)に形質転換し、アミピシリンを
50μg/mlで含有するLuria Broth寒天上にプレートした。形質転換体を
Luria Broth(アミピシリンを50μg/mlで含有する)で培養し、DNA
をPromega“マジック・ミニプレプス”を用いて調製した。IL4.Y124D
/IgG4組換えDNAの産生を制限消化物で確認し、完全IL4.Y124Dお
よびヒンジ-CH2-CH3 IgG4領域をDNA配列決定により確認した。表6
はIL4.Y124D/IgG4融合分子のコード領域の配列だけを記載し、表7
は融合蛋白質のアミノ酸配列を含む。IL4.Y124D/IgG4組換えDNA
を調製し、塩化セシウム勾配で精製し、そのDNAを用いてヒーラ細胞を一時的
にトランスフェクトさせた。
2.融合蛋白質の発現

エール塩を含むMEM培地)に変えた(時間0)。+3時間の時に、0.125
M CaCl2、1xHBS(HEPES緩衝セイライン)中の適当なDNA(25
μg)をその細胞に加えた。+7時間の時に,細胞をグリセロールショック(1
5%v/v)に付し、ついで5mM酪酸ナトリウム含有の12.5mlのシード
培地にて一夜放置してインキュベートした。その翌日、細胞をPBS(ダルベッ
コ・リン酸緩衝セイライン)で洗浄し、12.5mlの「収集培地」(7.5%の
ストック炭酸水素ナトリウム溶液を2%含むRPMI−1640)を加えた。
さらに24時間インキュベートした後、上澄を取り出し、1000rpmで5分
間遠心分離に付し、細胞残骸を除去し、4℃または−20℃のいずれかで貯蔵し
た。
3.生物学的活性

実施例3 IL4.Y124D/IgG4 PE融合蛋白質

PCRを行い、IL4.Y124Dコード領域を増幅し、実施例2に記載する
ようにXhoI部位を形成するその3’末端でサイレントヌクレオチド置換を導入
する。
第2のPCR反応を行い、ヒトIgG4重鎖PE変異体のヒンジ−CH2−C
H3フラグメントを増幅させる。IgG4 PEにおいて、ヒンジの残基10(K
abat番号付け、残基241)を野生型のセリン(S)からプロリン(P)に変え
、CH2の残基5(Kabat番号付け、残基248)を野生型のロイシン(L)か
らグルタミン酸塩(E)に変える。Angal S.、King D.J.、Bodmer M.W.
、Turner A.、Lawson A.D.G.、Roberts G.、Pedley B.およびAdair
R.、Molecular Immunology、第130巻、105〜108頁、1993は、
241残基(Kabat番号付け)がセリンからプロリンに改変されているIgG4
分子を記載する。この変化はIgG4分子の血清半減期を増加させる。
IgG4 PE変異体を、表5に記載の合成ヒトIgG4重鎖cDNAをPCR
突然変異誘発に付して形成させ、ついでpCD発現ベクターに連結させた。これ
はIgG4PEのヒンジ−CH2−CH3フラグメントを増幅させるPCR反応
の基質として用いるブラスミドであった。IgG4 PE変異体の配列を表8に記
載する。PE変異体を調製するのに改変されるIgG4 ヌクレオチド配列の残基
は以下のとおりである:

オリゴヌクレオチドのプライマーを、pDB952の誘導化について記載する
ように、IgG4 PE変異体のヒンジ−CH2−CH3領域を増幅するのに用い
る。
約700bp(IgG4 PE変異株のヒンジ−CH2−CH3)および400
bp(IL4.Y124D)のPCR産物を得、Promega“マジックPCRクリ
ーンアップ”キットを用いて精製する。ついで、その精製したPCR反応物を次
の酵素で消化し、「付着末端」:IgG4 PEではXhoIおよびXbaIならびに
IL4.Y124DではEcoRVおよびXhoIを形成する。消化物を37℃で3
時間インキュベートし、ついでエタノール沈降に付す。得られたDNAは約69
0bp(IgG4 PEのヒンジ−CH2−CH3)および370bp(IL4.
Y124D)の大きさである。
多量のIgG4 PE変異体のヒンジ−CH2−CH3フラグメントおよびIL
4.Y124Dフラグメントを得るために、精製し、かつ消化したPCR産物を
、IgG4 PEフラグメントのヒンジ−CH2−CH3ではXhoIおよびXbaI
またはIL4.Y124DフラグメントではEcoRVおよびXhoIのいずれかで
消化することで調製されるBluescript KS+TMに、つづいてGenecleanTMに連
結する。こうして、Bluescript KS+/IgG4 PE組換え体のヒンジ−CH
2−CH3およびBluescript KS+/IL4.Y124D組換え体を生成する。
多
量のこれらのDNAがPromega“マジック・マクシプレプ(Magic Maxiprep)
”法を用いて産生される。IgG4 PEヒンジ−CH2−CH3フラグメントを
XhoIおよびXbaIを用いてBluescript組換え体より切除する。得られた約6
90bpのフラグメントをGenecleanTMで精製し、多量のIgG4 PEヒンジ−
CH2−CH3XhoI/XbaIフラグメントを生成する。IL4.Y124Dフ
ラグメントをEcoRVおよびXhoIを用いてBluescript組換え体より切除し、
得られた約370bpのフラグメントをGenecleanTMで精製する。
pDB952の誘導化で記載したように、pDB951をEcoRVおよびXba
Iで消化することによりIgG4 PEのヒンジ−CH2−CH3およびIL4.
Y124Dフラグメントを連結させるためのベクターを調製する。
3種の記載されているフラグメント(IL4.Y124D EcoRV/XhoI、
IgG4 PE変異体XhoI/XbaIのヒンジ-CH2-CH3およびpDB951
のEcoRV/XbaI消化により得られる5.8kbフラグメント)を一緒に連結
して、Amerscham製のDNA連結キット(製品コードRPN1507)を用い、
16℃で一夜反応体をインキュベートして、プラスミドpDB953を形成する
。連結反応産物をPromega JM109能力細胞(高能力)に形質転換し、50
μg/mlのアミピシリンを含有するLuria Broth寒天上にプレートする。形質
転換体をLuria Broth(アミピシリンを50μg/mlで含有する)で培養し
、DNAをPromega“マジック・ミニプレプス”を用いて調製する。IL4.Y
124D/IgG4PE変異体組換えDNAの産生を制限消化物で確認し、完全
IL4.Y124Dおよびヒンジ−CH2−CH3 IgG4 PE変異体領域をD
NA配列決定により確認する。表9はIL4.Y124D/IgG4 PE融合分
子のコード領域の配列だけを記載し、表10は融合蛋白質のアミノ酸配列を含む

実施例4.IL4.Y124D/IgG4 PEをコードするDNA含有の哺乳動
物発現ベクター
1.DNAの構築
pCDNベクター(Aiyar,N.、Baker,E.、Wu,H-L.、Nambi,P.、Edw
ards,R.M.、Trill,J.J.、C.Bergsma,D.Molecular and Cellular Bi
ochemistry 131:75−86、1994)は、CMVプロモーター、ポリリ
ンカークローニング領域、およびBGHポリアデニル化領域を含有する。このベ
クターは、GeneticinTMを選択するためにβ−グロビンプロモーターとSV40
ポリアデニル化領域の間に挿入される細菌性ネオマイシンホスホトランスフェラ
ーゼ遺伝子(NEO)、メトトレキセート(MTX)を増幅するためにβ−グロ
ビンプロモーターとBGHポリアデニル化領域の間に挿入されるDHFR選択カ
セット、細菌を増殖するためのアンピシリン耐性遺伝子および複製のSV40起
点を含有する。
IL4.Y124D/IgG4 PEcDNAを挿入するために、pCDNベク
ターを以下のようにNde1およびBstX1で消化することで調製した:15μg
のDNAを反応2にて30ユニットのBstX1(Gibco-BRL)と55℃で1
時間インキュベートし、エタノール沈降に付した。得られたDNAを反応2にて
Nde1を用い、37℃で1時間消化し、エタノール沈降に付した。pDB953
(実施例3.1)をBstX1およびNde1で以下のように消化することでIL4
.Y124D/IgG4 PEフラグメントを調製した:15μgのDNAを反応
2にて30ユニットのBstX1と55℃で1時間インキュベートし、エタノール
沈降に付した。得られたDNAを反応2にてNde1を用い、37℃で1時間消化
し、エタノール沈降に付した。
IL4.Y124D/IgG4 PE Nde1/BstX1およびpCDN Nde1
/BstX1フラグメントを連結し、プラスミドpCDN−IL4.Y124D/
IgG4 PEを形成させた。連結を2ユニットのT4 DNAリガーゼ(Gibco-
BRL)とT4DNAリガーゼバッファーを一緒に用いて行った。反応物を16
℃で一夜インキュベートした。連結反応産物をGibco-BRL DH5a能力細胞
(サブクローニング能)に形質転換し、75μg/mlアンピシリン含有のLur
ia Broth寒天上にプレートした。形質転換体をLuria Broth(50μg/
mlのアンピシリン含有)中で培養し、アルカリ性溶菌によりDNAを調製した
。pCDN−IL4.Y124D/IgG4 PE DNAの産生を制限消化物で確
認した。組換えIL4.Y124D/IgG4 PE DNAの完全な配列を配列決
定により確認した。pCDN−IL4.Y124D/IgG4 PE組換えDNA
を調製し、Qiagenカラムを用いて精製し、DNAを用いて一時的にCOS細胞
