ゲル化された水性外相、非水性中間相及び水性内相
からなる乳濁液
本発明は乳濁液、その製造方法及びその使用、そして特に活性成分がその中に
分散されているゲル化された外相を有する乳濁液に関する。
ゲランガムのようなゲルの中へ活性成分を封じ込めることはよく知られている
。例えば、JP 62125850は食品、油脂、薬物などの成分のゲランガムビーズへの
封じ込めを開示している。実施例では、サラダ油乳濁液を0.5mlサイズの液滴と
して1%ゲランガム溶液に添加しており、得られるビーズは厚さ0.35mmのスキン
を有し、各球の中に0.3mlの油を含んでいる。
US4563366は野菜、果物、食肉、魚、砂糖、及び/又は牛乳を含む少なくとも
1つの分散された食品成分を含むマトリックスからなるゲル化食品製造物を開示
している。
GB2219803はゲラン、カッパ-カラゲナン及びマンナンのブレンドからなるゲル
化用組成物を開示している。このゲル化用組成物は食品製造物例えばペットフー
トなどのゲル化マトリックスとして有用である。
JP 63267361はゲランガム及びそのカラゲナン、ゼラチン、寒天、ローカスト
ビーンガム、キサンタンガム、カルボキシメチルセルロースなどとの組合せから
選ばれるゲル化剤の他に香水、殺菌剤、殺虫剤などを含むことができるゲルを開
示している。
しかしながら、ゲランガムのようなアニオン性ヒドロコロイドを含むゲルマト
リックスにある種の活性成分を組み入れることは、活
性成分とゲルとの間で後者の崩壊又は沈殿を引き起こす不適合性があるため問題
のあることが判明している。
この不適合性は、例えば制汗薬を作る試みにおいてアルミニウムクロロハイド
レートをゲランガムと配合する場合にも認められ、それにより望ましくないゲラ
ンガムの沈殿が起こる。同様に、他のカチオン性成分の不適合性、例えばカチオ
ン性薬物(塩酸ベラパミル、マレイン酸クロロフェニラミンなど)及びカチオン性
界面活性剤(例えば塩化ベンザルコニウムなど)と上述のアニオン性ゲルマトリ
ックスとのそれも問題があることが判っている。
本発明は上の問題を軽減するものであり、この場合、ゲルマトリックスと適合
しない活性成分はゲルの性質に悪い影響を与えることなくその中に組み入れるこ
とができる。
本発明によれば、水性のゲル化された外相、非水性の中間相及びゲル化された
外相とはなじみ合わない(incompatible)活性成分を含有する水性の内相からな
り、この場合活性成分を含有する内相は中間相により封じ込められている乳濁液
が提供される。
従って本発明の乳濁液は、水性のゲル化された外相を水性内相に存在する活性
成分から中間相が引き離すことにより上述の問題を解消するものである。
適切にはゲル化外相は1つ又はそれより多くのゲル化されたアニオン性ヒドロ
コロイドからなる。本発明にしばしば使用されるヒドロコロイドはゲランガムで
あるが、その他の適当なヒドロコロイドはアルギネート、ペクチン、カラゲナン
、寒天、ローカストビーンガムなどである。
ゲランガムは微生物シュードモナス・エロデア(Pseudomonas
elodea)を適当な栄養培地中で好気発酵させることにより得られる細胞外多糖で
ある。種々な形のゲランガムが知られており、例えば産生したそのままのもの、
脱アセチル化したもの、脱アセチル化し清澄化したもの、部分的脱アセチル化し
たもの、及び部分的脱アセチル化し清澄化したものの諸型である。
本発明のゲルに使用するゲランガムは「低度アセチル」ゲランガムが好ましい
。本明細書で使用する用語「低度アセチル」は0〜0.3重量%のゲランガムのア
シル化の水準を意味する。
本発明に有用な種々のアルギネートはDavidson編集,Handbook of Water-Solu
ble Gums and Resins(1980)の2章に所載のI.W.Cottrell及びP.Kovacsによ
る”Alginates”に詳細に記述されている。
アルギネートには「バイオアルギン」及び「アルガル」アルギネートがある。
バイオアルギンはシュードモナス及びアゾトバクター両族の菌株が生産する微
生物生産性多糖類であり、例えばJarmanらの、米国特許4,235,966に記述されて
いる。これらのアルギネートはD−マンヌロン酸及びL−グルウロン酸残基の一
部分がアセチル化された可変性のブロックコポリマーからなる多糖類である。Ja
rman等は生産される多糖は分子が一部分アセチル化されていることを除いて海藻
が生産するものと似ていると述べている。
用語「アルガル」アルギネートは天然に得られるアルギン酸及びその塩を指す
。主としてケルプから得られる天然由来のアルギン酸は商業的に入手可能であっ
て、例えばKELACIDTM(かってMerck & Co.,Inc.に帰属していたが、現在はMons
anto CompanyのKelco Div.

ネシウム塩、アンモニウムの塩、及びプロピレングリコールアルガルアルギネー
トである。本発明でもっとも好ましいのは天然に得られるアルガルアルギン酸ナ
トリウムであり、これは例えばかってはMerck & Co.,Inc.に帰属していたが、
現在はMonsanto CompanyのKelco DivisionがKELTEX,KELGIN及びKELTONEの商標で
商業的に販売しているようなものである。
ペクチンは多数の負に荷電したガラクツロン酸残基を含む枝分かれした分子か
らなる植物細胞壁多糖類である。その負の電荷のためペクチンは高度に水和され
ており、そして容易にカチオンと結合するため、本発明の乳濁液のゲル化外相を
形成するために適している。
ローカストビーンガムはイナゴマメ、Ceratonia siliguaのさやの抽出物であ
る。これは商業的に入手可能で、しばしば食品例えばアイスクリーム、ソーセー
ジ、及びチーズの安定剤として使用される。
アニオン性ヒドロコロイドは外相の重量に基づいて0.1〜5重量%、例えば
0.25〜2.5重量%の量でゲル化外相に存在するのが好ましい。磨耗しうるゲル化
された外相が必要な、例えば乳濁液が制汗薬として使用される場合、ヒドロコロ
イドは典型的には外相の重量に基づいて0.75〜1.25重量%の量で存在する。別に
薬剤放出系への適用のような場合は、ゲル化相が体液と接触したときくずれうる
ものであることが求められ、アニオン性ヒドロコロイドは一般にゲ
ル化外相の0.5〜0.6重量%の量で存在する。
本発明の乳濁液のいくつかの適用、例えば制汗薬、ボディーローションなどの
場合には、場合により外相はさらに香水を含むことがあり、典型的には外相重量
に基づいて0.75〜1.25重量%の量で存在する。外相は所望により着色剤を含有し
てよい。
ゲル化された外相は場合により防腐剤も含有することがあり、好ましい防腐剤
はn−プロピルp−ヒドロキシベンゾエートである。防腐剤は少量使用するのが
適当であり、例えばゲル化された外相の約0.2重量%を超えないことである。
さらにゲル化された外相は、殺生物剤を場合により含有することがあり、典型
的には外相重量に基づいて0.05〜2.5重量%の量存在する。
1つ又はそれより多い界面活性剤を外相に含有させることも適当であり、適当
な界面活性剤の例にはジエタノールアミドセチルホスフェート、ポリオキシエチ
レンひまし油、ポリオキシエチレン水素添加ひまし油、ポリオキシエチレントリ
グリセライド、ポリオキシエチレンラノリン、ポリオキシエチレンラウレート、
ポリオキシエチレンステアレートなどがある。一般に界面活性剤は外相重量に対
して1〜3重量%の水準で存在し、一層好ましくは外相重量に対して1.5〜2.5重
量%の水準で存在する。
アニオン性ヒドロコロイドは典型的にはカルシウム、マグネシウムなどのよう
な適当なカチオンによりゲル化される。
そのような多価ゲル化用カチオンの場合、これらは塩化カルシウム、塩化マグ
ネシウム、硫酸カルシウム、硫酸マグネシウムなどのような塩で供給するのが適
当である。ゲル化多価カチオンは一般に
外相重量に対して0.05重量%を超えない水準で乳濁液の外相に存在する。
アニオン性ヒドロコロイドがゲランガムからなる場合、ナトリウム、カリウム
などのような一価カチオンをゲランガムのゲル化に使用するのが好ましく、カリ
ウムが特に好ましい。適切には一価カチオンは塩化ナトリウム、塩化カリウム、
クエン酸三ナトリウム、クエン酸三カリウムなどのような塩で供給され、カリウ
ムの場合、好ましい塩はクエン酸三カリウムである。
ゲル化用一価カチオンは外相重量に対して0.5重量%を超えない水準で外相に
存在するのが有利である。
ゲル化用一価カチオンを使用する場合、外相はカルシウム、マグネシウムなど
のような多価イオンを本質的に含有しないのが好ましい。ゲル強度を増すために
多価イオンを使用することはこの技術分野における正常な実行手段であるから、
熟練した作業者はゲル化量の多価イオンをこのように回避することをもっとも異
常なことと考えるであろう。
望ましくは非水性中間相は揮発性シリコーン油、石油、パラフィン、又は植物
油例えばオリーブ油、落花生油、ひまし油、綿実油又は菜種油などを含めること
ができる油相からなる。このようにして、水相に分散された油中水型乳濁液の三
相乳濁液が本発明により提供される。本発明の有利な油相はシリコーン油、好ま
しくは揮発性シリコーン油であるが、もちろんその他の上述の油も中間相に同様
に使用することができると認識される。
上で述べたように、本発明のねらいはゲル化された外相とはなじみ合わない活
性成分を含む水性内相を非水性相の中に封じ込めるこ
とである。適切にはこの活性成分はアニオン性ヒドロコロイドとの間でその崩壊
又は沈殿を引き起こすという点でなじみ合わないカチオン性物質を含有している
。
そのようなカチオン性物質の例は、典型的にはアルミニウム及び/又はジルコ
ニウム塩のような塩として存在する多価金属イオン、典型的にはアルミニウムク
ロロハイドレート、アルミニウム−ジルコニウムクロロハイドレートなど、カチ
オン性薬剤例えば塩酸ベラパミル、マレイン酸クロロフェニラミンなど、及びカ
チオン性界面活性剤例えば塩化ベンザルコニウム、セチルトリメチルアンモニウ
ムクロライド、ラウリルジメチルアンモニウムクロライドなどを含む。
上記の活性カチオン性成分の範囲から本発明の乳濁液がいくつかの適用をもつ
ことを認識することができる。
活性カチオン性成分がアルミニウムイオンを含む第一の実施態様においては、
本発明の乳濁液は制汗薬としての適用がある。そのようなアルミニウムイオンを
含む制汗薬は本発明における使用に適するアニオン性ヒドロコロイドのようなゲ
ルとのアルミニウム含有物質の不適合性の理由から従来は作られなかった。
カチオン性薬剤が中間油相で封じ込められた水性内相に存在する本発明の第二
の実施態様においては、本発明の乳濁液は徐放性薬剤系として有用であり、この
場合ゲル外相は体液と接触するとゆっくり浸食されて封じ込められた薬剤を放出
する。
活性成分がカチオン性界面活性剤である本発明のさらに別の実施態様において
は、個人衛生用製品、例えばボディーローション、加湿剤、クリームなどが本発
明により提供される。本発明の乳濁液を
使用する個人衛生用製品の提供は、よく知られた油中水型乳濁液の加湿特性及び
水中油型乳濁液の望ましい皮膚感触特性が達成されることが組み合わされる点で
望ましいことである。
適切には内相は活性成分の水性溶液からなり、この場合溶媒は典型的には水で
ある。活性成分は溶解度の限界まで存在するのが望ましく、そしてアルミニウム
クロロハイドレート(これが好ましい成分である)を活性成分として使用する場
合、これを内相の重量に基づいて80重量%の量まで含有させることができる。活
性成分をその溶解度の限界まで水相溶媒中に含有させることは活性成分の量に余
裕を与える点で有利であり、さもないとゲル化外相と適合しないため、本発明の
乳濁液に組み込めないであろうと考えられる。
活性成分の水性溶液が非水性相に分散される一次乳濁液は最初に作るのが好ま
しい。45〜85重量%の水性溶液を15〜55重量%の非水性相に分散させるのが望ま
しい。適当な55〜85重量%の一次乳濁液をその後に、ゲル化性物質例えばゲラン
ガムなどの水性分散液の重量百分率均衡の範囲内で分散させる。
疎水性界面活性剤は一般に非水性相に含有される。適当な界面活性剤の例には
セチルジメチコンコポリオール、ポリソルベート60、ソルビタンモノラウレート
、ソルビタンモノステアレート、ソルビタンモノオレエート、ソルビタンモノパ
ルメート、ソルビタントリオレエート、ポリエチレングリコール−6−ソルビタ
ン蜜蝋、ポリエチレングリコール−20−ソルビタン蜜蝋、セテス−20−ステアラ
テース−2−ステアレース−20−オレース−2、アルミニウムマグネシウムヒド
ロキシドステアレートなどがある。典型的には界面活性剤は非水性相の重量に基
づいて0.5〜2.5重量%の水準で存在する。
ポリソルベート60を使用する場合、この界面活性剤は一般に1.5〜2.5重量%の水
準で含有され、一方セチルジメチコンコポリオールは典型的には0.5〜1.5重量%
の水準で使用する。
本発明の特に好ましい乳濁液は、典型的には上で説明したように実質的に一価
カチオンによりゲル化されたゲランガムからなる外相、シリコーン油からなる中
間相、及び活性カチオン成分特にアルミニウムクロロハイドレート又はアルミニ
ウム−ジルコニウムクロロハイドレートを含む水性内相からなる。この特に好ま
しい乳濁液は制汗薬としての使用に適しており、そしてさらに本発明により、ゲ
ランガムからなる外相、シリコーン油からなる中間相、及びアルミニウムクロロ
ハイドレート又はアルミニウム−ジルコニウムクロロハイドレートからなる群か
ら選択される活性カチオンを含む水性内相からなり、そして活性カチオン性成分
を含む内相は中間相により封じ込められている乳濁液からなる制汗薬が提供され
る。
本発明の制汗薬は一般に「スティック」型制汗薬であり、そして乳濁液は実質
的に固体である。本明細書で記述する「スティック」型制汗薬は典型的には容器
内に納めるように形作られた実質的に固体の本発明乳濁液の本体であり、それに
よりこの本体は容器に関連して前進した及び後退した位置の間を動くことができ
る。
本発明によってはなおその上、乳濁液がアニオン性ヒドロコロイドからなるゲ
ル化外相、中間相、及びゲル化外相と適合しない活性成分を含有する水性内相か
らなっており、それにより活性成分を含有する内相は中間相により封じ込められ
ている乳濁液のゲル化外相を与える実質的に上で説明したようなアニオン性ヒド
ロコロイドの使用が提供される。
本発明によりさらに、活性成分を含む水相を非水性相に一次二相乳濁液が生成
するように分散させ、一次乳濁液をゲル化性物質の水性分散液と混合し、そして
そのゲル化をもたらすことからなる実質的に上で説明した乳濁液の製造方法が提
供される。
適切にはゲル化性物質は実質的に上で説明したような1つ又はそれより多いア
ニオン性ヒドロコロイドからなり、この場合ゲランガムの使用が特に好ましい。
同様に非水性相及び活性成分も実質的に上で説明した通りであり、この場合非水
性相は揮発性シリコーン油からなり、そして活性成分はアニオン性ヒドロコロイ
ドとなじみ合わないカチオン性物質からなるのが好ましい。
望ましくは、疎水性界面活性剤例えば上述のセチルジメチコンコポリオールは
非水性相と、最初に述べた水性相をその中に分散させる前に、緊密に混合する。
そのような界面活性剤の使用は一次二相乳濁液の安定化を達成するために有効で
ある。
非水性相は最初に比較的低いエネルギー及び剪断の攪拌を与えるのが有利であ
り、典型的には水性相をそれに添加する間、パドル攪拌機などのような攪拌機を
使用して350〜450rpm(好ましくは390〜410rpm)の速度で攪拌する。その後比較
的高いエネルギー及び剪断の攪拌を使用し、典型的には1400〜1600rpm、好まし
くは1480〜1520rpmの速度で攪拌し、これにより安定な一次乳濁液が形成される
。
典型的には一次乳濁液をゲル化性物質の水性懸濁液と混合する前に50〜60℃の
範囲の温度に加熱する。
適切にはこの方法はゲル化性物質、典型的には上で説明したようなアニオン性
ヒドロコロイドを、一次乳濁液と混合する前、水のよ
うな水性媒体中に分散させることを含む。生成する水性分散液は一般に水和を実
行するために80〜90℃の範囲の温度に加熱し、その後DEAセチルホスフェート又
は上で説明したようなその他の同様な界面活性剤を添加する。分散液は一次乳濁
液と混合する前50〜60℃の範囲の温度に冷却するのが適当である。場合により一
次乳濁液を水性分散液と混合して得られる混合物は更に65〜70℃の範囲の温度に
加熱してよい。
ゲランガムをアニオン性ヒドロコロイドとして使用する場合、ゲル化はゲル化
用カチオン、典型的にはカリウム又はナトリウムのような一価イオンの添加によ
り達成するのが望ましい。適切にはこの混合物を冷却し固化させてゲルを形成す
る。本発明の好ましい態様によればクエン酸三カリウムを使用する。
水性相及び非水性相、ゲル化カチオンなどの重量百分率は実質的に上で説明し
た通りである。同様にこの方法には、場合によりさらに上で説明した香水、防腐
剤などのような成分とゲル化性物質の水性分散液との配合を加えてもよい。
本発明を次の実施例により例証するが、この実施例は例示のみを目的とする。
