【発明の属する技術分野】
本発明は、一般式1に示される化合物および/またはその塩を含有することを特徴とするゲル組成物および水中油(O/W)型組成物に関するものである。より詳しくは、一般式1に示される化合物および/またはその塩を含有することにより、油を含んだゲル組成物を形成させることが可能で、更にはこのゲル組成物を水で希釈することにより均一で安定な乳化組成物を得ることができることに関する。また、配合する油の種類や量を調整することにより、ミセル、ベシクル、エマルションといった様々な水中油(O/W)型組成物を目的に応じて調製することができる。

【従来の技術】
近年、人的な安全性の面で製剤において乳化剤として使用する界面活性物質の配合量を減らしたり、より作用の温和なものを使用する研究がなされてきた。その様な中で、有益な生理活性を持つ物質、いわゆる有効成分に乳化剤の役割までも担わせる研究がなされてきた。

またヒトに対し外用する製剤に関しては、経時安定性に優れており、更には皮膚に対する刺激が低く安全性の高いことはもちろん、有効成分の経皮吸収性、皮膚細胞内富化性をはじめとする有効成分の体内動態の向上が求められてきた。

【発明が解決しようとする課題】
この様な状況の中で、化粧品等の皮膚外用剤に求められるのは有益な生理活性を持つ成分自身でゲル組成物を形成し得ること、さらにはそのゲル組成物に水を加えることにより物理的に弱い攪拌力で効率よく経時的に安定である微細な乳化粒子を形成させること、同時に皮膚に対する刺激性が低いことである。

【課題を解決するための手段】
本発明者等は、かかる実情に鑑み鋭意検討した結果、一般式1に示される化合物および/またはその塩を含有することにより、自己組織体形成能が高く、経時安定性に優れており、更には油の乳化能が高く、皮膚に対する刺激が低く安全性の高い、ゲル組成物および水中油(O/W)型組成物の発明を完成するに至った。

すなわち、本発明のゲル組成物および乳化組成物は、一般式1に示される化合物および/またはその塩を含有するものであり、自己組織体形成能が高く、経時安定性に優れており、更には油の乳化能が高く、皮膚に対する刺激が低く安全性の高い組成物である。

以下、本発明の構成について詳述する。

本発明で用いられる一般式1の物質はクロマン骨格を持ち、肌あれに対し非常に有効な防御効果を持つ化合物である。これらの化合物は抗酸化能力に優れ、生体内における活性酸素、それに由来する過酸化脂質やフリーラジカルの弊害を予防したり、血行促進を代表とする生理活性作用により育毛・発毛効果を発揮したり、例えばビタミンやその誘導体と共存することにより相乗効果的にお互いの有効性を向上させることができる。

具体的にはR1、R2はそれぞれ水素、アルキル基、アシル基、酸化エチレン、アルカリ金属、アルカリ土類金属等のひとつ以上から選ばれ、必要に応じて組み合わせてもよい。これらの選択により溶解性、ゲル形成性、自己会合性等の物性を変化させることができる。

本発明で用いられる一般式1に示される化合物は、好ましくは、R1、R2の少なくともひとつがナトリウムである。さらに含有量は、特に限定されるものではないが0.1〜25重量%とすることが好ましく、0.1重量%未満では生理活性、ゲル形成能、乳化能に劣り安定なゲル組成物および水中油(O/W)型組成物が得られない場合があり、25重量%を超えて含有することは可能であるがゲル組成物および乳化組成物として経済的でない。

また、本発明におけるグリセリンの役割は非常に大きいものがある。一般式1に示される化合物は単独でもゲルを形成したり水中油(O/W)型組成物を形成したりする能力を有するが、グリセリンを添加することによりその分散性が向上し、この化合物の持ち得る能力を存分に発揮させることが可能となる。その配合量は特に限定されないが、0.5〜60%が好ましく、0.5%未満では顕著な効果が認められない場合があり、60%を超えて配合することは可能であるが使用感触の面でべとつきを感じ、デメリットとなる場合がある。

ゲルの調製においては特に限定されないが、一般式1をグリセリンと良く混合し、この状態に油を少しずつ撹拌しながら添加していくことによりゲルを形成する方法が好ましい。この他の方法においてもゲルを形成することは可能であるが、高温での処理が必要であったり、機械的な撹拌シェアを必要としたりするため、前述の方法を用いることで手軽に安全にゲルを製造することが可能となる。

本発明の組成物は上記必須成分の他に、通常の皮膚外用剤に用いられる化粧料、医薬部外品、医薬品等の各種成分、さらに食品に用いられる成分を配合することができる。例えば油性成分、脂質、保湿剤、増粘剤、薬効成分、殺菌・防腐剤、顔料、粉体、pH調整剤、紫外線吸収剤、抗酸化剤、可塑剤、香料、アミノ酸、甘味料、着色料等を適宜配合することができる。さらにゲル形成能や乳化能を補助する役割で他の界面活性剤も配合することができる。

具体的には油性成分としては、例えば炭化水素、植物由来油、昆虫由来油、動物油脂、トリグリセライド、高級アルコール、脂肪酸、高級アルコールと脂肪酸のエステル、シリコーン油等が挙げられる。保湿剤としては、例えば多価アルコール、高分子多糖類、植物抽出エキス、微生物の代謝物などが挙げられる。増粘剤としては、例えばカルボキシビニルポリマー、キサンタンガム、メチルセルロース、ポリビニルピロリドン、ゼラチン、ベントナイト等の粘土鉱物等が挙げられる。薬効成分としては、例えば各種ビタミンおよびその誘導体、アラントイン、グリチルリチン酸およびその誘導体、各種動植物抽出物等が挙げられる。さらに乳化剤としては、例えばポリオキシエチレンアルキルエーテル、ポリオキシエチレン脂肪酸エステル、ポリオキシエチレンソルビタン脂肪酸エステル、ソルビタン脂肪酸エステル、グリセリン脂肪酸エステル、ポリグリセリン脂肪酸エステル、ポリオキシエチレン硬化ヒマシ油等の非イオン界面活性剤、ステアロイル乳酸ナトリウム等のアニオン界面活性剤、リン脂質等の両性界面活性剤、塩化アルキルトリメチルアンモニウム等のカチオン界面活性剤が挙げられる。

本発明のゲル組成物および水中油(O/W)型組成物は公知の方法により製造することができ、化粧料、医薬部外品、外用医薬品、食品等の分野を問わず利用できるものであり、その剤型は目的に応じて任意に選択でき、クリーム、乳液、ローション、ゲル、軟膏、パック、スティック等の形態とすることができる。

【発明の効果】
本発明のゲル組成物および水中油(O/W)型組成物は、一般式1に示される化合物および/またはその塩を含有するものであり、従来から一般的に用いられている界面活性物質に比べ、有益な生理活性を有しながら、ゲル化能や油の保持能に優れ、配合成分を調整することによりミセル、ベシクル、乳化粒子を形成し、経時安定性に優れており、更には皮膚に対する刺激が低く、経皮吸収性が高いため皮膚の細胞内に富化することのできる安全性に優れたものである。

これらの効果を実証するために実施例1〜12を調製し評価した。

(ゲル化方法)
以下の処方にて、一般式1に示される化合物とグリセリン(A部)を常温にて充分攪拌した後、60℃に加温、同じく60℃に加温した油相(B部)をA部に攪拌しながら徐々に加える。その後室温まで冷却してゲル組成物を得た。

以下の処方にて、一般式1に示される化合物とグリセリンの適量(A部)を常温にて充分攪拌した後、60℃に加温、同じく60℃に加温した油相(B部)をA部に攪拌しながら徐々に加える。その後70℃に加温した水相(C部)を加え、グリセリンの残りを加え撹拌しながら室温まで冷却し水中油(O/W)型乳化組成物を得た。
