Claims (1)
- 【特許請求の範囲】 1. FKS1およびその機能的誘導体ならびにFKS2およびその機能的誘導 体よりなる群から選択される、実質的に純粋なDNA分子。 2. 微生物から単離された、請求項1記載のDNA分子。 3. 該微生物がアスペルギルス・フミガトゥス(Aspergillus f umigatus)、アスペルギルス・ニデュランス(Aspergillus nidulans)、カンジダ・アルビカンス(Candida albic ans)、クリプトコッカス・ネオフォルマンス(Cryptococcus neoformans)、ニューモシスティス・カリニ(Pneumocyst is carinii)およびサッカロミセス・セレビシエ(Saccharo myces cerevisiae)よりなる群から選択される、請求項2記載 のDNA分子。 4. 該微生物がサッカロミセス・セレビシエ(Saccharomyces cerevisiae)である、請求項3記載のDNA分子。 5. 図6の配列、図8の配列、図10のDNA配列、図11の配列、およびそ れらの機能的誘導体よりなる配列から選択される核酸配列を有する、請求項1記 載のDNA分子。 6. 図1、図2、図3および図4から選択される制限地図を有する請求項1記 載のDNA分子。 7. 該DNA分子が、そのDNAが原核生物または真核生物細胞に導入された 場合に発現され得るように調節配列に作動可能に連結している、請求項1記載の DNA分子。 8. 請求項1のDNA分子によってコードされた、実質的に精製された蛋白質 。 9. 図5のアミノ酸配列、図7の配列、図9の配列、図10のアミノ酸配列、 図12の配列、およびそれらの機能的誘導体から選択されるアミノ酸配列を有す る、請求項8記載の蛋白質。 10. 請求項1記載のDNA分子を含有する細胞。 11. YFK532−7C、R560−1C、MS14、YFK0978、Y FK1088−23B、YFK1088−16D、YFK1087−20B、お よびYFK1087−20Aよりなる群から選択される微生物。 12. 請求項8記載の精製された蛋白質に対する抗体。 13. (a)2つの微生物を培養し、第1の微生物は無傷のFKS1 DNA およびFKS2 DNAを担持し、第2の微生物は改変されたFKS1 DNA および改変されたFKS2 DNAを担持し、 (b)グルカンシンターゼに影響することが知られている化合物の定量可能な 量と共に工程(a)の微生物のアリコートをインキュベートし、 (c)テスト化合物と共に工程(a)の微生物のアリコートをインキュベート し;次いで、 (d)工程(b)および工程(c)の微生物におけるグルカンシンターゼ活性 を測定することを特徴とする、グルカンシンターゼ活性を調節する化合物を選定 する方法。 14.請求項13の方法によって選定された化合物。 15.(a)3つの微生物を培養し、第1の微生物は無傷のFKS1 DNA およびFKS2 DNAを担持し、第2の微生物は改変されたFKS1 DNA の形態を担持し、第3の微生物は改変されたFKS1 DNAの形態および少な くとも2コピーのCNA2 DNAおよび少なくとも2コピーのCNB2 DN Aを担持し、 (b)カルシニューリンに影響することが知られている化合物の定量可能な量 と共に工程(a)の3つの微生物のアリコートをインキュベートし、 (c)テスト化合物と共に該3つの微生物のアリコートをインキュベートし; 次いで、 (d)工程(b)および工程(c)のアリコートの相対的増殖を測定すること を特徴とする、カルシニューリンに影響する化合物を選定する方法。 16. 請求項15記載の方法によって選定された化合物。 17. 請求項16記載の化合物よりなる医薬組成物。 18. (a)テスト化合物を1,3−β−D−グルカンシンターゼサブユニッ トを含有する化合物と混合して混合物を得、 (b)該混合物における1,3−β−D−グルカンシンターゼサブユニット活 性を測定し、次いで、 (c)該混合物の1,3−β−D−グルカンシンターゼサブユニット活性を標 準と比較することを特徴とする、1,3−β−D−グルカンシンターゼサブユニ ット活性を調節する化合物を選定する方法。 19. 請求項18記載の方法によって選定された化合物。 20. 請求項19記載の化合物よりなる医薬組成物。 21. 請求項20記載の組成物を非ヒト患者に投与することを特徴とする治療 を必要とする患者を治療する方法。
