2. Description of the Related Art Cacao (Theobroma cacao) is an evergreen small tree from the tropical humid area of the United States.
Fruits with a diameter of 5 to 30 cm and a diameter of 10 cm are formed. It is said that it has been cultivated and acclimatized since 2000 years ago, and cocoa beans in fruits are used as a raw material for food and drink such as chocolate and cocoa. In addition, as the physiological actions of alkaloids contained in cocoa beans, cardiotonic action and diuretic action have been reported. On the other hand, cocoa beans are known to contain polyphenols having an antioxidant effect, but their carcinogenic preventive effect has not been reported.

Even with the remarkable progress of medicine, the number of cancer patients is increasing and is the leading cause of death in the Japanese people.
In addition, it is suggested that the number of people may increase more than ever before in the coming aging society. On the other hand, research on the occurrence of cancer has also progressed, and our dietary habits and living environment occupy most of the causes of cancer, and the mechanism thereof is being investigated.

[Means for Solving the Problems] As a result of intensive studies to solve the above problems, the present inventors have found that cacao bean, which is a main raw material of chocolate and cocoa, has an excellent physiological effect on carcinogenesis prevention. It was confirmed for the first time that there was a substance, and the present invention was completed based on this finding. That is, the present invention provides a health food or drink containing a carcinogen for preventing carcinogenesis extracted from cocoa beans using ethanol.

BEST MODE FOR CARRYING OUT THE INVENTION The method for producing a carcinogenic substance for preventing cancer according to the present invention is extraction with ethanol, which is harmless to the human body. As the cocoa beans used as a raw material, various forms such as cocoa shell, cocoa nibs, cocoa mass, and defatted cocoa mass are used, but defatted cocoa mass is preferable because the extract can be made into powder. Defatted cocoa mass is obtained by removing oil from cocoa beans obtained by removing a portion of the cocoa shell called cocoa shell from cocoa beans. The oil content may be removed by a known method.

As the extraction method, 300 to 800 parts by weight, preferably 400 to 700 parts by weight of an ethanol solution is added to 100 parts by weight of cocoa beans, and the mixture is heated at room temperature for several hours to 4 hours.
Stir for 8 hours, preferably for about 24 hours. As an ethanol solution, water to ethanol is 60:40 to 0:10.
It is possible to use a solution having a ratio of 0, preferably 30:70 to 0: 100.

The ethanol contained in the extract obtained by filtration may be removed under reduced pressure, and then freeze-dried or vacuum-dried to give a powder. This extract may be further purified by chromatography or the like, if necessary. As an example, in the examples, open column chromatography (one of the most popular methods currently used in Diai
on HP2MG) was performed to obtain crude polyphenol, which is the main component of carcinogen. The carcinogenic substance thus obtained may be either a concentrated liquid or a powder, depending on the intended use of food and drink.

By the way, the carcinogenic substance in the food is naturally not taken alone, but is taken as a mixture of several components. In addition, synergistic actions between carcinogens often produce excellent effects. Therefore, rather than studying the effects of the individual components of the carcinogen, it is necessary to understand the development of the overall effects of the multiple components contained in the food. Based on such a viewpoint, the present inventors have confirmed that the extract obtained from cocoa beans has a carcinogenic effect.
The effect of suppressing mutations caused by carcinogens and mutagens, which trigger cell carcinogenesis, was evaluated as an index.

It is well known that the Ames method using Salmonella typhimurium is used as a method for screening the mutagenicity of chemical substances. As the number of substances detected as mutagens and confirmed to have carcinogenicity has increased, this method has come to be widely used as a simple method for detecting carcinogenic substances. In addition, by using a substance whose carcinogenicity has been confirmed in advance in this test system, it has become possible to create a state in which mutation occurs, and to evaluate the mutation suppressive action of the substance, that is, the carcinogenic preventive action (T .Osawa,
et al, Enviromental and Molecular Mutagenesis, 18 ,
35-40, 1991). As a result of conducting an antimutagenicity test by modifying such Ames method, it was found that the cocoa extract has a strong carcinogenic preventive effect.

Example 1 Method for Producing Carcinogenic Substances To 100 parts by weight of defatted cocoa mass was added 500 parts by weight of 80% ethanol, and the mixture was stirred and extracted at room temperature for 24 hours. The extract was filtered and concentrated under reduced pressure. The obtained concentrated liquid was freeze-dried to obtain a cocoa extract (80% ethanol extract). Next, 30 ml of the supernatant obtained by further centrifugation of the obtained concentrated solution was used for Diaion HP2.
MG column (manufactured by Mitsubishi Chemical Corporation, size 35 mm id x 310 m
m) was applied, and alkaloids were eluted and removed with an eluent of purified water: ethanol = 80: 20. Furthermore, an active fraction having a high polyphenol content was obtained with an eluent of purified water: ethanol = 20: 80. Further, add 30 ml of this fraction to Si
It was applied to a lica gel column (size 35 mm id × 310 mm) and chromatographed. Elution with a mixed solution of chloroform: methanol gave an active concentrated fraction in an eluent of chloroform: methanol = 60: 40. The outline of the purification method is shown in FIG. The active fraction and the active concentrated fraction were used for the test after lyophilization.

Example 2 Carcinogenesis prediction by antimutagenicity test
Evaluation of anti-protective effect 50 μl of test substance, 100 μl of mutagen, metabolic activating enzyme S-9mix (mixture of S-9 for mutagenicity test and cofactor I for Ames test (Oriental Yeast)) 300 μl of Salmonella typhimurium TA98
100 μl was added, mixed well, and incubated at 37 ° C. for 20 minutes. Immediately after incubation, soft agar solution (agar 2.4
2.5 ml of 1/10 amount of 0.5 mM histidine-biotin solution was added to 400 ml of NaCl, 12.0 g of NaCl), mixed well, and spread on a minimal glucose agar plate medium. After standing at 37 ° C. for 2 days, the number of returned colonies that had grown was counted. As the test substance, the active fraction obtained in Example 1 and the active concentrated fraction were used. As the mutagen, Trp-P-2, which is one of the heterocyclic amines derived from foods whose carcinogenicity has been confirmed, was used. This thermal metabolite of tryptophan, Trp-P-2 itself does not have carcinogenicity, but is absorbed into the body, metabolized by enzymes in the liver, converted into an active body with strong carcinogenicity, and bound to DNA. It is known to induce mutations by doing.
