Claims (1)
- 【特許請求の範囲】 1. 核酸分子が、一つまたはそれ以上のサルモネラ菌血清型のDNAまたはR NAと選択的にハイブリッド化する、下記の配列: を含む1本鎖DNA、およびそれと相補的なDNA配列、およびその類似体およ びフラグメントであることを特徴とする、サルモネラ菌(Salmonella)種の検出 および同定のための核酸分子。 2. DNAに基づく検出システムにおけるプローブまたはプライマーとして使 用するための請求項1に記載の核酸分子。 3. 核酸分子が、下記の配列: およびそれと相補的なDNA配列、およびその類似体およびフラグメントの少 なくとも一つを含むことを特徴とする、請求項1に記載の核酸分子。 4. 核酸分子が、一つまたはそれ以上のサルモネラ菌血清型のDNAまたはR NAと選択的にハイブリッド化する、下記の配列: およびそれと相補的なDNA配列、およびその類似体およびフラグメントの少 なくとも一つを含むことを特徴とする核酸分子。 5. 核酸分子が、更に、固体支持体の標識および/または結合のためのハイブ リッド化領域または非ハイブリッド化領域を含むことを特徴とする、請求項1〜 4の何れか一項に記載の核酸分子。 6. 請求項1〜5の何れか一項に記載の核酸分子の一つまたはそれ以上を、D NAに基づく検出システムにおけるプローブまたはプライマーとして使用するこ とを特徴とする、一つまたはそれ以上のサルモネラ菌血清型の検出方法。 7. DNA検出システムが、ポリメラーゼ鎖延長反応(PCR)、自己-維持配列 複製(3SR)、Q-ベータレプリカーゼ増幅システム、リガーゼ増幅反応(LAR)お よび機能的に類似の変異体からなる群から選択された増幅システムを採用するこ とを特徴とする、請求項6に記載の方法。 8. 増幅システムがPCRであることを特徴とする、請求項7に記載の方法。 9. 少なくとも二つのオリゴヌクレオチドプライマーを使用し、これらのプラ イマーが標的DNAの相対する鎖と相補的であることを特徴とする、請求項8に 記載の方法。 10. プライマーが、ST11/ST14対およびST11/ST15対からなる群から選択され、 各対のプライマーが標的DNAの相対する鎖と相補的であることを特徴とする、 請求項9に記載の方法。 11. 巣籠もりプライマーを使用することを特徴とする、請求項8〜10の何れか 一項に記載の方法。 12. 請求項3で定義したST15、あるいはその相補的配列またはそのDNA類 似体もしくはフラグメントをプローブとして使用することを特徴とする、請求項 6に記載の方法。 13. Salmonella Typhimurium を検出することを特徴とする、請求項6に記載 の方法。 14. 請求項3で定義した核酸分子ST22、あるいはそれと相補的なDNA配列ま たはその類似体もしくはフラグメントを使用することを特徴とする、請求項13に 記載の方法。 15. 少なくとも下記の成分: a)ポリメラーゼ b)請求項2に記載の少なくとも二つのオリゴヌクレオチドプライマー を含む、PCR技術を用いたサルモネラ菌種検出に使用されるキット。 16. 少なくとも下記の成分: a)ポリメラーゼ b)固定化用手段または標識用手段を備えた請求項2に記載の少なくとも二つ のオリゴヌクレオチドプライマー を含む、DIANA技術を用いたサルモネラ菌種検出に使用されるキット。 17. 少なくとも下記の成分: a)逆転写酵素 b)請求項2に記載の少なくとも二つのオリゴヌクレオチドプライマー(両方 のプライマーはポリメラーゼ結合部位を有する) を含む、3SR技術を用いたサルモネラ菌種検出に使用されるキット。 18. 少なくとも下記の成分: a)リガーゼ b)請求項2に記載の少なくとも二つのオリゴヌクレオチドプライマー(これ らオリゴヌクレオチドは請求項1の配列に隣接する) を含む、LAR技術を用いたサルモネラ菌種検出に使用されるキット。 19. 少なくとも下記の成分: a)RNA指導性RNAポリメラーゼ b)5'-MDV-1構造またはそのフラグメントを有するRNAプローブ c)請求項2に記載の少なくとも一つのオリゴヌクレオチドプライマー(この プライマーは固定化されているか、または固定化可能である) を含む、LAR技術を用いたサルモネラ菌種検出に使用されるキット。
