酸性pHにおいて安定性を増加するよう修飾されたオリゴヌクレオチド関連出願の相互参照
本出願は、(出典明示して本明細書の一部とみなす)1986年10月28日
付出願の米国特許出願第06/924,234号の一部係属出願である1990
年7月27日付出願の米国特許出願第07/558,338号の一部係属出願で
ある。発明の背景および概要
本発明は酸性pHにおいて増加した安定性を供給するオリゴマーを供給する方
法、このようなオリゴマーをその作用部位に送達する方法、ならびに経口投与ま
たは酸に対する耐性が有利である他の投与形態のための処方における使用に関す
る。
酸性条件下でのデオキシリボ核酸(DNA)の脱プリン(塩基と糖の間のグリ
コシド結合の開裂を通じるヌクレオシジル単位からのプリン塩基の損失)が報告
されてきた(ヘベシ・エル(Hevesi,L))ら、ジャーナル・オブ・アメリカン
・ケミカル・ソサイエティ(J.Amer.Chem.Soc.)、94、4715−472
0(197))。治療用のオリゴデオキシリボヌクレオチドの経口送達は、薬剤
送達の通常条件で約4時間までの、および徐放性薬剤送達の条件で約12時間ま
での胃の酸性条件(pH1)への暴露を要求するであろう(例えば、米国特許第
4,839,177号を参照)。酸性条件でのその安定性の欠如のために、経口
投与されたオリゴデオキシヌクレオチドの充分量が無傷で有効なままであること
はありそうにない。伝えられるところによれば、リボ核酸(RNA)はそのDN
Aの相対物よりも酸性条件での脱プリンに対し、より安定であると報告されてい
るが、これは糖と塩基の間のグリコシド結合の2’−ヒドロキシルの明白な安定
化効果によるものである(ヘベシ・エル(Hevesi.L.)ら、上記)。
RNAは酸性条件下で脱プリンに耐性であるかもしれないが、生物物質中に存
在する普偏的ヌクレアーゼに対するその感受性はその治療用の有用性を限定する
。さらに、該分子の、中性から少々塩基性の状態に対する本質的な不安定性のた
めに、
薬剤としての修飾されない形態でのオリゴリボヌクレオチドの利用は実行できそ
うにない。
不運にも、リン酸結合をメチルホスホネートで置換することによる、ヌクレア
ーゼに対するRNAの保護は、不可能である。なぜなら糖の2’−ヒドロキシル
が急速にメチルホスホネート骨格を切断するからである。発明の概要
本発明は酸に耐性な形態の予め選択された塩基配列を有するヌクレオシジル単
位からなるオリゴマーを供給する方法に関する。本発明の一の態様において、オ
リゴマーが供給され、ここに、ヌクレオシジル単位は2’−O−アルキルリボシ
ル基である糖部分を有する。好ましくは、オリゴマーは本質的に中性である。好
ましいのはメチルホスホネートのヌクレオシジル間結合を有するもので、より好
ましいのは約50ないし約100%のヌクレオシジル間結合がメチルホスホネー
ト結合であるものである。好ましい2’−O−アルキルリボシル基は2’−O−
メチルリボシル基を含む。
他の態様により、酸耐性形態で供給されるオリゴマーは、メチルホスホネート
ヌクレオシド間結合からなり、好ましくは約50%ないし約100%のメチルホ
スホネート結合からなる。
付加的な態様として、本発明は、該オリゴマーが経口投与に適し、酸による分
解に対して耐性を示すような予め選択された塩基配列を有するヌクレオシジル単
位からなるオリゴマー調製方法に関する。一の態様において、該方法は2’−O
−アルキルリボシル基、より好ましくは2’−O−メチルリボシル基である糖部
分を有するヌクレオシジル単位を用いてオリゴマーを合成することからなる。好
ましくは、オリゴマーは実質的に中性である。より好ましくは、オリゴマーはメ
チルホスホネートヌクレオシジル間単位を有するよう合成される。
他の方法として、該方法はメチルホスホネートヌクレオシジル間結合を有する
オリゴマーを合成することからなる。特に好ましいのは約50パーセントないし
約100パーセントのヌクレオシジル間単位を有するオリゴマーである。
さらなる態様において、本発明はオリゴマーを哺乳動物に治療の目的のために
経口送達する方法に関し、ここに該オリゴマーはプリン塩基を有するヌクレオシ
ジル単位からなり、該方法は酸耐性オリゴマーを投与することからなる。本態様
の一の具体例において、該酸耐性オリゴマーは、2’−O−アルキルリボシル基
、より好ましくは2’−O−メチルリボシル基からなる糖部分を有するヌクレオ
シジル単位からなる。好ましくは、該オリゴマーは実質的に中性である。好まし
い酸耐性オリゴマーはメチルホスホネートヌクレオシジル間結合を有するオリゴ
マーを含み、より好ましくはヌクレオシド結合の約50ないし約100パーセン
トがメチルホスホネート結合である。とりわけ好ましい態様において、該酸耐性
オリゴマーは制御された速度で放出される形態で投与される。
数ある因子の中で、本発明は、2’−O−アルキルリボシル基である糖部分あ
るいはメチルホスホネートヌクレオシジル間結合を有するヌクレオシド単位を含
むように合成されたオリゴマーが酸触媒の脱プリンおよびこれに続く加水分解に
有利な耐性を示すという、驚くべき我々の発見に基づいている。
2’−O−アルキルリボシル基およびメチルホスホネートヌクレオシジル間結
合を有するヌクレオシジル単位からなるオリゴマーは、対応する2’−デオキシ
リボヌクレオシドメチルホスホネートよりも、RNA標的分子とより安定なデュ
ープレックスを形成するように思われるということも信じられている。
さらなる態様において、本発明は制御された速度で放出される形態の本発明の
酸耐性オリゴマーからなる医薬組成物に関する。代わりに、酸耐性オリゴマーか
らなる医薬組成物が供給され、その組成物はそれ自体酸性であるか、あるいは製
造または貯蔵の間酸性条件に暴露されてもよい。定義
本明細書中で用いるごとく、以下の語句は、特に断りのない限り、以下の意味
を有する。
「プリン」もしくは「プリン塩基」なる語は、天然のアデニンおよびグアニン
塩基のみならず、8-位で塩基置換されている塩基のごときこれら塩基の修飾物
、または、6-位で修飾されているグアニンアナログ、もしくはアデニンのアナ
ログ、2-アミノプリン、ならびに9-デアザプリン誘導体のごとき9-位に炭素
置換窒
素を有するプリンのアナログおよび他のプリンアナログをも包含する。
「ヌクレオシド」なる語は、ヌクレオシジル単位であって、それらと互換的に
用いられ、5-炭素の糖および含窒塩基よりなる核酸のサブユニットをいう。該
語には、その塩基としてA、G、C、TおよびUを有するそれらのヌクレオシジ
ル単位のみならず、偽イソシトシンおよび偽ウラシルおよび(8-置換プリンの
ごとき)他の修飾塩基のごときピリミジンアナログを含む天然の塩基のアナログ
および修飾形態も含まれる。RNAにおいては、該5-炭素の糖はリボースであ
り;DNAにおいては、2’-デオキシリボースである。ヌクレオシドなる語句
には、2’-O-アルキルリボースのごとき修飾された糖を有するものを含むかか
るサブユニットの他のアナログも含まれる。
[式中、Rは水素またはアルキルもしくはアリール基である]をいう。適当なア
ルキルもしくはアリール基には、ホスホネート結合を立体的に隠さないか、また
は互いに相互作用しないものが包含される。該ホスホネート基は、「R」もしく
は「S」の立体構造のいずれかで存在してもよい。ホスホネート基は、ヌクレオ
シジル単位を結合させるヌクレオシジル間リン基架橋(もしくは結合)として用
いられる。]
[式中、ホスホジエステル基は、ヌクレオシジル単位を結合させるヌクレオシジ
ル間リン基架橋(もしくは結合)として用いられる。]をいう。
「非-ヌクレオシド・モノマー単位」とは、その中の塩基、糖および/またはリ
ン骨格が他の化学基により置き換えられているモノマー単位をいう。
「ヌクレオシド/非-ヌクレオシド・ポリマー」とは、ヌクレオシドおよび非-
ヌクレオシド・モノマー単位よりなるポリマーをいう。
「オリゴヌクレオシド」もしくは「オリゴマー」なる語は、約100個のヌク
レオシド長を超えることもあるが、一般に約4ないし約100個のヌクレオシド
長であるヌクレオチシド間連結により結合されているヌクレオシドの鎖をいう。
それらは、酵素的な方法によっても得ることができるが、通常はヌクレオシド・
モノマーから合成する。従って、「オリゴマー」なる語は、該ヌクレオシド・モ
ノマーを結合させるヌクレオシジル間結合を有するオリゴヌクレオシドの鎖を意
味し、かくして、オリゴヌクレオチド、非イオン性オリゴヌクレオシドアルキル
-およびアリール-ホスホネート・アナログ、オリゴヌクレオチドのアルキル-お
よびアリール-ホスホノチオエート、ホスホロチオエートもしくはホスホロジチ
オエート・アナログ、オリゴヌクレオチドのホスホルアミデート・アナログ、ホ
スホトリエステルおよび他のオリゴヌクレオシド・アナログおよび修飾オリゴヌ
クレオシドのごとき、中性ホスフェートエステル・オリゴヌクレオシド・アナロ
グが含まれ、また、ヌクレオシド/非-ヌクレオシド・ポリマーも含まれる。また
、該語句には、モノマー単位間の1もしくはそれを超えるリン基結合が、ホルム
アセタール結合、チオホルムアセタール結合、スルファメート結合もしくはカル
バメート結合のごとき非-リン結合により置き換わったヌクレオシド/ヌクレオチ
ドポリマーも包含される。また、それには、該糖およびリン基の双方が、モルホ
リノ塩基アナログ、もしくはポリアミド塩基アナログのごときで置き換えられま
たは修飾されたヌクレオシド/非-ヌクレオシド・ポリマーも包含される。また、
それには、該非-ヌクレオシドの塩基、糖、およびホスフェート骨格が、非-ヌク
レオシド基で置き換わっているか、または非-ヌクレオシド基が当該ヌクレオシ
ド/非-ヌクレオシド・ポリマーに挿入されているかのいずれかのヌクレオシド/
非-ヌクレオシド・ポリマーも包含される。所望により、該非-ヌクレオシド基は
、標的配列と相互作用できるか、または標的細胞中への取り込みを変化させるこ
とができる他の小分子に結合するよう供することもできる。
「アルキル-もしくはアリール-ホスホネート・オリゴマー」なる語は、少なく
とも1個のアルキル-もしくはアリール-ホスホネート・ヌクレオシジル間結合を
有するオリゴマーをいう。適当なアルキル-もしくはアリール・ホスホネート基
には、該ホスホネート結合を立体的に隠さないか、または互いに相互作用しない
アルキル-もしくはアリール-基が包含される。好ましいアルキル基には、約1な
いし約6個の炭素原子を有する低級アルキル基が包含される。適当なアリール基
は、共役π電子系を有する少なくとも1個の環を有し、所望により置換されてい
てもよく、かつ好ましくは約10個までの炭素原子を有する炭素環式アリール基
および複素環アリール基が包含される。
「メチルホスホネート・オリゴマー」(または「MP-オリゴマー」)なる語
は、少なくとも1個のメチルホスホネート・ヌクレオシジル間結合を有するオリ
ゴマーをいう。
「中性オリゴマー」なる語は、ヌクレオシド・モノマー間の非イオン性ヌクレ
オシジル間結合(すなわち、正または負のイオン電荷を有しない結合)を有する
オリゴマーをいい、例えば、アルキル-もしくはアリール-ホスホネート結合、ア
ルキル-もしくはアリール-ホスホノチオエート、およびホスホトリエステル結合
、特に中性エチルトリエステル結合のごとき中性ホスフェートエステル結合のご
とき、ヌクレオシジル間結合;ならびに、スルファメート、モルホリノ、ホルム
アセタール、チオホルムアセタールおよびカルバメート結合のごとき非-ホスホ
ラス含有ヌクレオシジル間結合を有するオリゴマーが包含される。所望により、
中性オリゴマーは、オリゴヌクレオシドまたはヌクレオシド/非-ヌクレオシド・
ポリマーと、結合相手よりなる第二分子との間の結合体よりなっていてもよい。
かかる結合相手は、インターカレーション剤、アルキル化剤、細胞表面レセプタ
ーに対する結合基質、親油性剤、およびソラレン(psoralen)のごとき光-架橋
剤を含む核酸修飾基ならびに核酸の標的部位を切断できる基等からなってよい。
かかる結合相手は、さらに、該オリゴマーの取り込みをさらに向上させることが
でき、該オリゴマーと該標的配列との相互作用を修飾することができ、あるいは
、該オリゴマーの薬物動態的な分布を変化させることができる。本質的に必要な
のは、該オリゴマー結合体を構成する該オリゴヌクレオシドまたはヌクレオシド
/非-ヌレノオシド・ポリマーが実質的に中性であることである。
「実質的に中性」なる語は、該ヌクレオシド・モノマー間のヌクレオシジル間
結合の少なくとも約80%が非イオン性結合であるオリゴマーをいう。
「中性アルキル-またはアリール-ホスホネート・オリゴマー」なる語は、少な
くとも1つのアルキル-もしくはアリール-ホスホネート結合よりなる中性ヌクレ
オシジル間結合を有する中性オリゴマーをいう。
「中性メチルホスホネート・オリゴマー」なる語は、少なくとも1つのメチル
ホスホネート結合よりなるヌクレオシジル間結合を有する中性オリゴマーをいう
。
「酸耐性」なる語は、デオキシリボオリゴヌクレオチドと比較して、N−グリ
コシル結合の加水分解による酸触媒脱プリンに対し耐性であるオリゴマーをいう
。
「トリプレット」または「トリアド」なる語は、標的配列の1個の塩基(もし
一本鎖ならば)または複数の塩基(もし二本鎖ならば)、第二の鎖の塩基および
第三の鎖の塩基(もし標的配列が一本鎖ならば)もしくは第三の鎖の塩基(もし
標的が二重鎖ならば)間の3つのヌクレオシド塩基の水素結合した複合体をいう

本発明のオリゴマー類は独立して2’−O−アルキルリボシル基から選択され
る糖部分を有するヌクレオシジル単位からなる。適当なのは1ないし5個の炭素
原子のアルキル基である。特に好ましいヌクレオシドは2’−O−メチルリボシ
ル基を有する。
選択されたヌクレオシド間結合を有するオリゴマー鎖は当業者に公知の合成技
術によって都合よく調製することができる。例えば、ホスホジエステルおよび他
のあるリン含有ヌクレオシジル間結合を有するオリゴマーの合成には、市販の機
器、試薬およびプロトコール類を利用できる((ゲイト,エム・ジェイ(Gait,
M.J.)「オリゴヌクレオチド合成:実践的アプローチ(Oligonucleotide Synt
hesis:A Practical Approach)」、アイアールエル・プレス社(IRL Press)、
1984年;コーエン,ジャック・エス(Cohen,JackS.)「遺伝子発現のオリ
ゴデオキシリボヌクレオチド・アンチセンス・インヒビター(Antisense Inhibi
tors ofGene Expression)」、シーアールシー・プレス社(CRC Press)、フロ
リダ州、ボカ・レートン(BocaRaton,FL)1989年;および「オリゴヌクレ
オチドおよびアナログ:実践的アプローチ(Oligonucleotides and Analogues;A
Practical Approach)」、エフ・エックスタイン(F.Eckstein)編、1991
年)も参照)。ある種の非−リン含有ヌクレオシジル間結合を有するオリゴマー
の調製は、(出典明示して本明細書の一部とみなす)米国特許第5,142,0
47号に記載されている。
好ましいのは実質的に中性であるオリゴマーである。
特に好ましい態様において、これらのオリゴマーはメチルホスホネートヌクレ
オシジル間結合を有する。より好ましくは、全てのヌクレオシド間結合がメチル
ホスホネート結合である。メチルホスホネートヌクレオシジル間結合と他のヌク
レオシジル間結合との混合物を有するオリゴマーが、ある治療的適応症に好まし
く、本発明の範囲内にあるよう意図される。
好ましくは、該オリゴマーは約4ないし約40ヌクレオシドからなり、より好
ましくは、約6ないし30ヌクレオシドからなる。特に好ましいのは約8ないし
約20ヌクレオシドのオリゴマーである。有用性および投与
本明細書中で提供されるオリゴマーは、高い選択性を有する核酸のごとき標的
配列または蛋白質と高アフィニティー複合体を形成することができる。例えば、
誘導されたオリゴマーは架橋により(ソラレン(psoralen)類)、あるいは1本
または両方の鎖の開裂により(EDTA)核酸中の標的部位と結合し、次いで不
可逆的に修飾するために用いることができる。切断のための標的部位の注意深い
選択により、鎖のうち1本を、分子バサミとして、選択された核酸配列を特異的
に切断するために用いることができる。
本明細書中で提供されるオリゴマーは、不可逆的に核酸を修飾、分解、破壊し
、次いでその機能を不可逆的に阻害する核酸断片またはその標的配列と反応を起
こすことができる基と、反応するかまたはそれを修飾する核酸を包含するように
誘導化することができる。
これらのオリゴマーは、生きている細胞中のものと同一である特定の遺伝子ま
たは標的配列の発現を不活性化するか阻害するか変化させ、発現の選択的な不活
性化、阻害もしくは変化を可能とする。標的配列は、DNAまたはあるmRNA
の前駆体mRNAのごときRNAであってよい。mRNA標的配列は開始コドン
領域、ポリアデニル化領域、mRNAキャップ部位あるいはスプライス結合部を
含有する。これらのオリゴマーは不完全なまたは望ましくない生産物を生産する
か、または活性化されると望ましくない影響を引き起こす遺伝子を永久的に不活
性化するか、発現をオフにするか、または破壊するためにも使用することができ
る。
本明細書中で提供されるオリゴマーは、核酸の転写された領域と共にデュープ
レックスまたはトリプルヘリックス複合体あるいは他の形の安定な会合形態を形
成するので、これらの複合体は「アンチセンス」あるいは三本鎖療法において有
用である。本明細書中で用いる「アンチセンス」治療とは、イン・ビトロ(in v
itro)あるいはイン・ビボ(in vivo)で望ましくないDNAまたはRNA配列
を不活性化する、特異的結合オリゴマーの使用を包含する包括的用語である。
多くの病気および他の疾患は望ましくないDNAまたはRNAの存在によって
特徴付けられるが、これはある例では一本鎖であって、他の例では二本鎖である
。これらの病気および疾患は当該分野で一般的に理解されているアンチセンス治
療の原理を用いて治療することができる。アンチセンス治療は、相補性または他
の特異的結合手段を用いて(本発明の場合にはデュープレックスあるいはトリプ
ルヘリックス複合体の形成により)、特異的なDNAまたはRNA標的配列を標
的とすることを包含する。
本発明で用いるオリゴマーは、単独で投与するか、あるいは、隣接するまたは
離れた標的または前記の一般的機構を有する組み合わさった効果のアンチセンス
機構のために、オリゴマーの組み合わせを投与することができる。
治療的な適用には、経口、局所または局部投与を包含する種々の投与様式に該
オリゴマーを処方できる。製造の間に、またはこのような処方自体を、適用の部
位にて優勢な状態(例えば皮膚のアシッド・マントル(acid mantle))により
適合するようにある程度まで酸性条件にするかもしれない場合に、酸条件に接触
し得る酸耐性オリゴマーを含有する医薬処方があると有利であろう。技術および
処方は一般的に、「レミントンの医薬科学(Remington's Pharmaceutical Scien
ces)」、マック・パブリッシング社(Mack PublishingCo.)、ペンシルベニア
州、イーストン(Easton,P.A.)の最新版に見い出すことができる。該オリゴ
マーの有効成分は、投与様式および投与形態の性質に応じて、一般的に、充填剤
、増量剤、結合剤、湿潤剤、崩壊剤、界面活性剤、分解性ポリマー、または滑沢
剤を含有できる希釈剤または賦形剤のごとき担体と組み合わせる。典型的な投与
形態には、錠剤、散剤、懸濁液、乳剤および液剤を含めた液状調製物、顆粒剤、
ならびにカプ
セル剤が含まれる。
本発明のオリゴマーは、具体的には、胃の中の状態では通常の薬剤の送達下で
4時間まで、徐放の形態で送達される場合は12時間までの薬剤の酸性状態への
暴露を必要とする。ある疾患の治療には、これらのオリゴマーを徐放性形態で処
方した方が有利である。出典明示して本明細書の一部とみなすコロンボ(Colomb
o)らの米国特許出願第4,839,177号には、ある好ましい制御された速
度の放出の形態が記載されている。経口投与用には、該オリゴマーは、カプセル
剤、錠剤および液体のごとき遅延放出だけでなく通常放出の経口投与形態に製剤
化される。
これらのオリゴマーは特に局所投与のための調製物の処方に適している、なぜ
なら皮膚にはアシッド・マントル(acid mantle)があるため、これらの酸耐性
オリゴマーを含む処方は有利であると分かるであろう。出典明示して本明細書の
一部とみなす米国特許出願第07/707,879号に記載されているように、
これらのオリゴマーは皮膚と粘膜を横切るという知見を考慮すれば、これもまた
有利であろう。酸性媒体を含む処方を供給することも望ましいであろう。
局所投与のために、本発明で用いるオリゴマーは、当該分野で一般的に公知の
軟膏、膏剤、点眼剤、ゲルまたはクリームに処方される。
全身投与は、粘膜内または経皮の手段によっても可能であり、または該化合物
は経口投与できる。粘膜内または経皮投与には、浸透されるべき障壁に適当な浸
透剤が該処方に用いられる。このような浸透剤は、一般的に当該分野で知られて
おり、例えば、粘膜内投与のための肝汁酸塩およびフシジン酸の誘導体が含まれ
る。加えて、活性剤を加えて浸透を容易にすることもできる。粘膜内投与は、例
えば、吸入、または坐薬による投与に適した処方と同様、経鼻用スプレーの使用
により行うことができる。
本発明の理解の補助のために、以下の実施例が含まれ、これらは一連の実験の
結果を記載する。本発明に関連する以下の実施例は、もちろん、本発明を特に限
定ずると解釈されてはならず、本発明のこのような変形は、今公知なものもまた
は後で開発されるものも、当業者の権限の中にあって、本明細書で後に請求され
る本発明の範囲内にあると考えられる。実施例 実施例1 2’−O−メチルアデノシン合成試薬の合成
A.5’−O−ジメトキシトリチル−2’−O−メチル−3’−O−(N,N−
ジイソプロピルアミノ−O−β−シアノエチルホスフィン)−N−ベンゾイ
ルアデノシンの調製
5’−O−ジメトキシトリチル-2’−O−メチル−N−ベンゾアデノシン
(0.75g;1.09ミリモル)(バリー・アソシエーツ,インコーポレイテ
ッド(Barry Associates,Inc.))を無水の1/1 アセトニトリル/ジイソ
プロピルエチルアミンと3回共沸させた。ヌクレオシドを次いで30mlの無水

ジイソプロピルアミノ)メチル]ホスホノアミトのモノマーを用いて合成した。
固相合成を、以下の修飾をする以外は製造者の忠告に従い、バイオサーチ・モデ
ル(Biosearch Model)8750DNAシンセサイザーによってメタクリレート
ポリマーの支持台上で行った:モノマーを100mMの濃度においてアセトニト
リル中に溶解した。DEBLOCK試薬(DEBLOCK reagent)とはジクロロメタ
ン中の2.5%ジクロロ酢酸をいう。OXIDIZER試薬(OXIDIZER reagent
)は、0.25%水、25%2,6−ルチジン,72.5%テトラヒドロフラン
中の25g/Lのヨウ素をいう。CAP Aは、アセトニトリル中10%無水酢
酸をいう。CAP Bは、ピリジン中0.625%のN,N−ジメチルアミノピ
リジンをいう。カップリング時間は4分まで延長させた。
ジメトキシトリチル基を合成の最後にオリゴヌクレオチドから除去した。
オリゴヌクレオチドを次いで支持体から切断し、脱保護した。支持体が結合し
たオリゴヌクレオチドを合成カートリッジから取り出し、スクリュー・トップ(
screw top)のついたガラス製の1ドラムのバイアルの中に入れた。支持体を、
1mlのアセトニトリル/エタノール/NH4OH(9/9/1)の溶液により室
温で30分間処理した。次いで、1mlのエチレンジアミンを反応容器に添加し
、反応を6時間行わせ、完結させた。オリゴヌクレオチドを含む上清を次いで支
持体から除去し、支持体を2mlの1/1 アセトニトリル/水で2回すすぎ、
次いで上清と合した。合した溶液を水で全体の容積が30mlになるまで希釈し
、約4mlの6N塩酸で中和した。中和した溶液を、予め10mlのアセトニト
リル、10mlの50%アセトニトリル/100mM 炭酸水素トリエチルアン
モニウム、10mlの25mM 炭酸水素トリエチルアンモニウムで順次平衡化
したウォーターズ(Waters)のC−18セプーパック(Sep-Pak)カートリッジ
を用いて脱塩した。反応溶液がカラムを通過した後で、それを30mlの水で洗
浄した。生成物を次いで5mlの1/1 アセトニトリル/水で溶出した。
オリゴヌクレオチドを逆相カラム(ワットマン(Whatmann)RACII)上のH
PLCによって、50mM 酢酸トリエチルアンモニウム中のアセトニトリルの
勾配によって精製した。
カップリング時間を3分間に延ばして、立体障害2’−O−メチルモノマー試
薬の適当なカップリングを起こさせる以外は実施例1(B)の2’−O−メチル
アデノシンのモノマーを用い、化合物2について記載されたように、化合物4を
合成し、脱保護し、精製した。化合物4を、支持体に結合したデオキシアデノシ
ン上で合成した。実施例4 アデノシンオリゴリボヌクレオチドテトラマーの調製(化合物3)
オリゴリボヌクレオチドテトラマー(化合物3)を、5’−O−ジメトキシト
リチル−2’−O−tert−ブチル−ジメチルシリル−3’−O−N,N−ジ
イソプロピル−β−シアノエチルホスホルアミダイトアデノシン(ミリポア(Mi
llipore))を用いて合成した。合成は、カップリングの時間を12分間にまで
延ばし、より立体障害の2’−O−tert−ブチルジメチルシリルRNAモノ
マーが反応するようにする以外は標準的なミリゲン(Milligen)ホスホルアミダ
イド法により、ミリゲン(Milligen)8750自動DNAシンセサイザーを用い
て、1μmoleのスケールで行われた。合成は、孔を制御したガラスに結合し
た2’−O−tert−ブチルジメチルシリルアデノシン(ペニスラ・ラボラト
ーリーズ(Pennisul aLaboratories))上で行われた。全ての他のオリゴヌクレ
オチド合成試薬はミリゲン(Milligen)の標準的プロトコールによって記載され
た通りである。合成後、オリゴリボヌクレオチドは、滅菌された、RNaseの
無い状態で扱われた。水を0.5%ジエチルピロカーボネートによる一晩の処理
の後オートクレーブすることによって滅菌した。全てのガラス製品は300℃で
少なくとも4時間加熱処理した。該オリゴヌクレオチドを、最初に、支持体に結

該オリゴヌクレオチドは、カップリング時間を4分まで延長し、より立体障害
の2’−O−メチル試薬の適当なカップリングを起こさせること以外は、化合物
1について記載されたように合成し、脱保護し、精製した。該化合物を、支持体
に結合したデオキシアデノシン上で合成した。実施例A オリゴマーの酸安定性の決定
胃のpHを刺激する酸性条件下で、化合物1ないし5の相対的安定性を測定し
た。該化合物を塩化水素の水溶液、pH1で処理し、時間に対するグリコシド結
合の切断の速度は、逆相HPLCクロマトグラム上の消化生成物アデニンの出現
によって決定された。
逆相HPLC分析はベックマン・システム・ゴールド(Beckman SystemGold)
HPLCモデル126ポンプ、モデル168フォトジオドアレイ検出器、および
モデル507のオートインジェクターで行われた。ワットマン(Whatman)RA
CIIOD3(100×4.6mm)分析カラムを分析のために用いた。用いられ
た溶媒系は、0.1M酢酸トリエチルアンモニウム水溶液、pH7中のアセトニ
トリルの勾配であった。勾配は10分間にわたり0ないし2%のアセトニトリル
であって、次の10分間では2ないし60%のアセトニトリルであった。勾配が
次いで0%アセトニトリルに戻りカラムを次の注入のために平衡化した。液速は
1ml/分であった。この勾配はアデニンを出発物質および種々の副産物からき
れいに分けた。
0.1M塩酸、pH1.05の100mlのストックを滅菌水中に調製し(滅
菌方法は上記参照)、アデニンの溶出液が、ベースラインの干渉を受けていない
領
域を確かめるためにHPLC分析した。該ベースラインは所望の領域で干渉を受
けていなかった。
テトラマーの脱プリンの速度を、次いで、10.D.単位260を注入される体
積である110μlで溶解して、0時間の点を得ることにより決定した。その後
10μlの試料を各々0.5時間おきに(化合物1および2)、または1時間お
きに(化合物3ないし5)自動的に採取した。結果を以下の表Iに掲げる。
図1はテトラマーに対する無傷のアデノシンの%対時間のプロットを示す。図
2は無傷のアデノシンの%の対数対時間のプロットを示す。該対数プロットから
、テトラマーの脱プリンの速度のkA’及び半減期が計算される。表1はホスホ
ジエステル対照と比較した場合のこれらの数値ならびに脱プリンの相対速度を含
む(化合物1)。
この結果は、2’−O−メチルアデノシニルメチルホスホネートがホスホジエ
ステルデオキシアデノシンまたはメチルホスホネートデオキシアデノシンのアナ
ログのいずれよりも大いに安定(175×)であることを明白に示している。2

−O−メチルアデノシニルメチルホスホネートについて明白な小さい脱プリン値
は、2’−OMe−アデノシニルの代わりのデオキシアデノシニル残基である3
’−末端のヌクレオシドに起因しているということが最もありそうなことである
が、これは支持にはならない。脱プリンの速度の違いを示すために、pH1にお
ける処理の4時間後、約33%のホスホジエステルデオキシアデノシン、メチル
ホスホネートデオキシアデノシンのうち7%のみ、および2’−O−メチルアデ
ノシンメチルホスホネートのちょうど0.28%が脱プリンされた。
大変驚くべき観察は、メチルホスホネートデオキシアデノシニルテトラマーの
安定性であった。グリコシド結合に対するホスホネート骨格の安定化効果は予期
せぬことであった。実施例B 37℃におけるオリゴマーの酸安定性の決定
化合物1ないし4の相対する安定性が、37℃に維持された熱ブロックを用い
て実施例Aに述べられたごとく決定された。
結果を以下の表IIに示す。
図3は、pH1.0、37℃におけるアデノシニルテトラマーの、時間に対す
る%脱プリンとしてプロットした脱プリン速度を示す。図4は、時間に対する%
脱プリンの対数としてプロットしたpH1.0、37℃におけるアデノシニルテ
トラマーの脱プリン速度を示す。図5は、(2’−O−メチルA)3(dA)−
メチルホスホネートオリゴマーについてのpH1.0、37℃の酸性条件に対す
るメチルホスホネートの骨格の安定性を示す。条件は、0.1M HCl中0.
3mMのオリゴマー、37℃、pH1.0であった。検出の限界は0.2%;誤
差≦2%。アデニニルを有する種類の全体と比較した遊離アデニンの増加と、こ
れに続くHPLCにより速度が決定された。データは脱プリン、および2’−O
−メチルアデノシニルメチルホスホネートオリゴマーに対する3’−末端のデオ
キシアデノシンの切断に対し補正された。実施例C オリゴ−2’−O−メチルリボヌクレオチシドメチルホスホネートの酸安定性
オリゴ−2’−O−メチルリボヌクレオシドメチルホスホネート(A)が、適
当な保護された2’−O−メチルリボヌクレオシドメチルホスホンアミダイトの
シントンを用いて調製された。以下の注釈において、Nmは2’−O−メチルリ
ボヌクレオシドを示し、下線はメチルホスホネート結合の位置を示す。該オリゴ
マーの5’−ヌクレオチド間結合はホスホジエステル結合である。該オリゴマー
は酢酸で緩衝化されたピリジン中のヒドラジン水和物で連続して処理して脱保護
し、次いで95%エタノール中のエチレンジアミン溶液(1:1 体積/体積)
で処理した。該オリゴマーをDEAEセルロースクロマトグラフィーで精製し、

オリゴマーAはポリヌクレオチドキナーゼおよびガンマー[32P]−ATPを
用いてリン酸化された。リン酸化されたオリゴマーを次いで0.1N HClと
インキュベートするか、または37℃において一晩(〜14時間)0.1N H
