Claims (8)
- 1.リボ核酸の合成方法であり、 a)式: ▲数式、化学式、表等があります▼(I)[ここで、R1は、以下の式で表され る基から選択されたピリミジン塩基またはプリン塩基から誘導された基である、 ]▲数式、化学式、表等があります▼ [ここで、R5は、HまたはCH3であり、R6、R7、およびR8は、水素原 子または環外NH2基を保護するアシル基であり、R2は、ヌクレオチド合成に 適当な酸媒体において不安定な基であり、R3は、固体支持体であり、 R4は、トリアルキルシリル基である、]で表される第1ヌクレオシドを、2′ 位の全水酸基はR4により保護されている合成リボ核酸を形成するために、式( I)で表される、1以上の同一または異なったヌクレオシド、[ここで、R1、 R2、およびR4は、上記と同様の意味を有し、R3はヌクレオチド合成に適当 なリン基である、]と連続縮合し、 b)上記合成されたリボ核酸を、式: ▲数式、化学式、表等があります▼(II)[ここで、R9は、直鎖状または分 岐したC1からC10アルキル基、R10およびR11は、同じでも異なってい てもよい直鎖状または分岐したC1からC10アルキル基であり、R9と同じで も異なっていてもよく、nは1から3の数であり、整数であっても整数でなくて もよい、]で表される脱保護剤で処理することにより、2′位の全ての水酸基を 脱保護することからなることを特徴とする、リボ核酸の合成方法。
- 2.脱保護剤が、 式: N(C2H5)3・3HF で表されるものからなることを特徴とする、請求項1に記載の方法。
- 3.脱保護剤による処理が、常温で140分間行なわれることを特徴とする、請 求項1または2に記載の方法。
- 4.R4が、t−ブチルジメチルシリル基である、請求項1ないし3のいずれか 一項に記載の方法。
- 5.R3が、式: ▲数式、化学式、表等があります▼ [ここで、R15およびR16が、同じでも異なっていてもよいC1からC4ア ルキル基である、] である、請求項1ないし4のいずれか一項に記載の方法。
- 6.R15が、メチル基であり、R16が、エチル基である、請求項5に記載の 方法。
- 7.R15およびR16が、イソプロピル基である、請求項5に記載の方法。
- 8.R2が、ジメトキシトリチル基である、請求項1ないし7のいずれか一項に 記載の方法。
